The current Danish project on genetic markers and hydatidiform moles involves the examination of the histocompatibility between patient, spouse and molar tissue. For this purpose 2 techniques have been developed for HLA-A,B determination of molar tissue: (a) a cytotoxic microtechnique for the HLA typing of cultured molar cells, and (b) a microabsorption method using selected HLA antisera for the examination of frozen molar tissue. To date molar cell cultures or tissue have been examined in 22 of the cases. The results of HLA determination were compared with chromosomal markers to elucidate the origin of the haploid chromosome complements. In the cases examined a general agreement between the HLA-A,B type and the origin of the chromosomes was observed. The microabsorption method used seems to be less sensitive than the cytotoxic technique.  相似文献

A wireless power transfer system for endoscopic micro-robot operating at 36 kHz is presented in this paper. The issue of patient' s health and safety regarding exposure to the electromagnetic field is addressed. The specific absorption rate and current density can be used to investigate the electromagnetic influences on the biological tissues surrounded by the wireless power launching coil. In view of this purpose, the limited close-ound solenoid electromagnetic model is built, the relationship between the electric intensity and the specific absorption rate and current density is deduced, and the simulation experiments are done. Experimental results show that the values of SAR and current density related to different tissue catalogs are all very small and do not exceed their own limits respectively when the resonance frequency of operation is 36 kHz.  相似文献

The main metabolic pathway for the interstitial matrix component hyaluronan seems to be via lymph. Present estimates of turnover of intestinal hyaluronan are based on isolated organs and/or experiments in anaesthesia over a few hours. In this study lymph was sampled from the main mesenteric lymphatic for 24 h in awake, restrained rats. The tissue content of hyaluronan averaged 28 /μ g-1 in the duodenum/jejunum and 124 μ g-1 in the colon. Based on tissue content and lymphatic output, the turnover rate of hyaluronan in the small intestine was estimated at approximately 50%. In another experimental group anaesthetized rats were fed either phosphate buffer or triolein in buffer via a gastric tube. Lymph flow increased with fluid absorption from the gut and hyaluronan output in lymph was elevated correspondingly. However, no effect of acute fat administration on hyaluronan concentration or lymph flow could be demonstrated.  相似文献

Articular cartilage has a limited capacity for self-repair. To overcome this problem, it is expected that functional cartilage replacements can be created from expanded chondrocytes seeded in biodegradable scaffolds. Expansion of chondrocytes in two-dimensional culture systems often results in dedifferentiation. This investigation focuses on the post-expansion phenotype of human nasal chondrocytes expanded on macroporous gelatin CultiSpher G microcarriers. Redifferentiation was evaluated in vitro via pellet cultures in three different culture media. Furthermore, the chondrogenic potential of expanded cells seeded in polyethylene glycol terephthalate/ polybuthylene terephthalate (PEGT/PBT) scaffolds, cultured for 14 days in vitro, and subsequently implanted subcutaneously in nude mice, was assessed.

Chondrocytes remained viable during microcarrier culture and yielded doubling times (1.07±0.14 days) comparable to T-flask expansion (1.20±0.36 days). Safranin-O staining from pellet culture in different media demonstrated that production of GAG per cell was enhanced by microcarrier expansion. Chondrocyte–polymer constructs with cells expanded on microcarriers contained significantly more proteoglycans after subcutaneous implantation (288.5±29.2 μg) than those with T-flask-expanded cells (164.0±28.7 μg). Total collagen content was similar between the two groups.

This study suggests that macroporous gelatin microcarriers are effective matrices for nasal chondrocyte expansion, while maintaining the ability of chondrocyte differentiation. Although the exact mechanism by which chondrocyte redifferentiation is induced through microcarrier expansion has not yet been elucidated, this technique shows promise for cartilage tissue engineering approaches.  相似文献

The relationship between serum antibodies to human herpesvirus-6 (HHV-6) and cytomegalovirus (CMV) infection was studied in immunosuppressed adult organ graft recipients all of whom had IgG to both HHV-6 and Epstein-Barr virus capsid antigen (EBVCA) before operation and who had received an organ or organs from HHV-6 seropositive donors. In primary CMV infection the titre of IgG to HHV-6 rose substantially (between 32- and 512-fold) in eight out of eight patients whereas IgG to EBVCA only rose 32-fold in two patients. Moreover, the HHV-6 responses coincided closely with the CMV seroconversion. Serum absorption studies gave no evidence for antibody cross-reaction between CMV and HHV-6 because the CMV antibody titre could be reduced specifically without affecting HHV-6 antibody titres and vice versa. In recurrent CMV infection, HHV-6 antibody levels rose (32-fold) in three out of eight patients but these changes did not coincide with the CMV antibody response. Similarly, in the complete absence of CMV infection, five out of eight patients showed antibody rises to HHV-6 (between four- and 16-fold). IgG titres to EBVCA were stable in both these groups of patients. It is concluded that there is serological evidence (rising titre greater than or equal to four-fold) for genuine HHV-6 reactivation or, alternatively, for reinfection in 16 out of the 24 patients. This phenomenon was most frequent in primary CMV infection where the largest HHV-6 antibody responses were seen probably because of an, as yet, undetermined interaction between the two viruses.  相似文献

Summary It was the purpose of this study to evaluate the diagnostic usefulness of an oral absorption test using nonlabeled Vit B12 suggested by a commercial distributor as an alternative for the more expensive Schilling test (ST). Plasma levels of Vit B12 were measured with a commercial kit before and 4 h after oral administration of 1 mg Vit B12 in 32 normals, in 16 patients with normal ST, and in 14 patients with abnormal ST for determination of sensitivity and specificity with the ST as golden standard. In normals, a mean of 767±404 pg/ml before and 1096±776 pg/ml after oral Vit B12 with a mean increase of 331±453 pg/ml was measured. Because of the obvious large variation, no meaningful range for normal absorption could be established. In the two patient subsets, there was no Gaussian distribution of the results, with a meridian of Vit B12 increase after absorption of 142 pg/ml, range 27–2668 pg/ml, in the group with normal ST and a meridian of 244 pg/ml ranging from 40 to 2453 pg/ml in the group with abnormal ST. Statistical nonparametric analysis did not reveal any difference between the two groups. Assuming a minimum required increase of 100 pg/ml, as suggested by the kit distributor, a sensitivity of only 27% and a specificity of 75% was obtained. The lack of any diagnostic value of this approach might be caused by the known nonintrinsic-factor mediated absorption of approximately 1% of any B12 given orally even in complete intrinsic factor deficiency and by the relatively large amount of oral Vit B12 needed for a cold absorption test. 目的 定量研究血液灌流对有机磷农药硫线磷和其解毒药阿托品的吸附作用.方法 模拟临床血液灌流装置,对含硫线磷和硫酸阿托品的血样进行灌流吸附,分别用毛细管气相色谱法和高效液相色谱法测定硫线磷和硫酸阿托品的残留量.结果 吸附剂用量为0.5、1.0和1.5 g,包膜活性炭在灌流2.0 h后硫线磷的清除率均能达到90%以上,硫酸阿托品的清除率依次为61.9%、84.9%和88.9%;HA230树脂在灌流1.5 h后硫线磷清除率都达到90%以上,硫酸阿托品的清除率也依次高达88.0%、97.2%和98.4%;包膜活性炭灌流3.0h后,硫酸阿托品与硫线磷的比值最高为灌流前的10.1倍,而HA230树脂灌流后,此比值最高为灌流前的6.7倍.结论 包膜活性炭和HA230吸附树脂血液灌流1.5~2.0 h均能清除血中大部分硫线磷,而且均能增加血中硫酸阿托品和硫线磷浓度的比值.  相似文献

Summary To investigate whether intestinal calcium absorption parallels that of sodium following extracellular fluid volume expansion, the effects of saline loading on intestinal transport of calcium. sodium and water were studied in rats by perfusing jejunal loops in situ.After calcium-free saline infusion net calcium absorption was reversed similar to that of sodium and water and net secretion occurred. Concurrently, blood-to-lumen (b-l) calcium flux, measured using45Ca, increased significantly (P<0.001). Following expansion with calcium-containing Ringer a similar reversal of net calcium, sodium and water flux was also observed. Again, the b-l calcium flux increased but to a significantly lesser extent (P<0.05). Plasma ionized calcium remained unchanged after calcium-rich Ringer loading, but decreased significantly (P<0.001) when calcium was omitted from the solution. Plasma immunoreactive parathyroid hormone was unchanged after expansion with the calcium containing solution but increased following calcium-free infusion.It is concluded that after extracellular fluid volume expansion: 1. net jejunal calcium absorption is decreased; 2. the decrease parallels that of sodium and water; 3. b-l calcium transport is enhanced to a greater degree by calcium-free Ringer infusion than by a calcium-rich solution. Objective To study on adsorption effect of cadusafos and atropine sulfate by hemoperfusion.Method Hemoperfusions were performed for sheep blood samples with cadusafos and atropineby through imitated extracorporeal closed circulating perfusion apparatus.Residual cadusafos was determined by gas chromatography and residual atropine was determined by high performance liquid chromatography.Result Dose of adsorption agent was 0.5,1.0 and 1.5 g,respectively.Two hours after hemoperfusion with membrane coated activated charcoal,clearance rate of cadusafos in 3 groups all exceeded 90%.and clearance rate of atropine sulfate was 61.9%,84.9%,88.9%,respectively.One and a half hours after hemoperfusion with HA230 absorption resin,clearance rate of eadusafos in 3 groups all exceeded 90%,and clearance rate of atropine sulfate was 88.0%,97.2%,98.4%,respectively.Three hours after hemoperfusion with membrane coated activated charcoal,The concentration ratio of cadusafos and atropine sulfate in blood promoted to 10.1 times,and the ratio was 6.7 times after hemoperfusion with HA230 absorption resin.Conclusion It suggested that cadusafos were mostly removed from blood after 1.5~2.0hours hemoperfusion with membrane activated charcoal or HA230 absorption resin.The concentration ratio of cadusafos and atropine sulfate in blood will increased after hemoperfusion.  相似文献

The lipophilicity of propranolol is increased by some bile salts which form ion-pairs. In the presence of taurodeoxycholate, the logarithm of the apparent partition coefficient (log P) of propranolol is increased. Moreover, the apparent diffusion constants in vitro of propranolol as ion-pairs at pH 3.0–6.0 are about 5–6 times higher than those of propranolol alone.

The area under the curve values of plasma concentration—time profiles of propranolol, following its oral administration to rabbits together with taurodeoxycholate, are about 1.4 times higher than those after administration of propranolol alone. Moreover, after the admistration of propranolol with taurodeoxycholate the plasma concentration rises more rapidly, with a point of inflection between 0.5 and 1.5 h, than after administration of propranolol alone.

Taurodeoxycholate does not modify the first-pass effect of propranolol in rabbits following intravenous and intraportal administration. The absorption of an oral dose of propranolol in the presence of taurodeoxycholate increases from 70% to 100%, due to the higher lipophilicity of the ion-pair. The plasma concentration—time curves suggest the hypothesis that greater absorption of the ion-pair occurs mainly in the upper region of the gastrointestinal tract.  相似文献
