建立了高效液相色谱-蒸发光散射检测器法测定硫酸阿米卡星及有关物质的含量.采用Extend C18色谱柱,以水-氨水-冰醋酸(96:3.6:0.4,pH10.0)为流动相,流速0.8ml/min,漂移管温度110℃,氮气流速3L/min.在0.05～2.5mg/ml浓度范围内线性关系良好(r=0.9998),方法回收率99.6%～101.2%.     

Monomeric and dimeric states exhibited by the kinesin‐related motor protein KIF1A

Abstract: KIF1A, a kinesin‐related motor protein that transports pre‐synaptic vesicles in neurons, was originally presumed to translocate along microtubules (MT) as a monomer. Protein structure predictions from its amino acid sequence failed to identify the long coiled‐coil domains typical of kinesins, which led researchers to believe it does not oligomerize into the canonical kinesin dimer. However, mounting evidence using recombinant chimeric protein indicates that KIF1A, like conventional kinesin, requires dimerization for fast, unidirectional processive movement along MTs. Because these studies are somewhat indirect, we wished to test the oligomerization state of native KIF1A, and to compare that to full‐length recombinant protein. We have performed hydrodynamic analyses to determine the molecular weights of the respective complexes. Our results indicate that most native KIF1A is soluble and indeed monomeric, but recombinant KIF1A is a dimer. MT‐binding studies also showed that native KIF1A did not bind to MTs in either the presence of AMP‐PNP, apyrase, or adenosine triphosphate (ATP), but recombinant KIF1A bound to MTs most stably in the presence of ATP, indicating very different motor functional states. To further characterize KIF1A's dimerization potential, we prepared peptides corresponding to the neck domains of MmKIF1A and CeUnc104, and by circular dichroism spectroscopy compared these peptides for their ability to form coiled‐coils. Interestingly, both MmKIF1A and CeUnc104 neck peptides formed homodimeric coiled‐coils, with the MmKIF1A neck coiled‐coil exhibiting the greater stability. Collectively, from our data and from previous studies, we predict that native KIF1A can exist as both an inactive monomer and an active homodimer formed in part through its neck coiled‐coil domain.     

奥氮平及其片剂的HPLC测定

建立奥氮平及其片剂测定的HPLC法.采用ODS C18色谱柱,0.01mol/L磷酸二氢钾溶液-甲醇-乙腈(1:1:1,pH8.3)为流动相,检测波长273nm.线性范围为0.01～1mg/ml,检测限1ng.方法平均回收率为99.4%,RSD为0.7%.     

上海市杨浦区涉苯企业职业卫生危害现状调查

目的调查涉苯企业贯彻国家职业卫生法规执行情况及现状,为建立辖区职业卫生监督的有效管理提供依据.方法依据<中华人民共和国职业病防治法>、<使用有毒物品作业场所劳动保护条例>等法律、法规和国家职业卫生标准,依法对辖区60家涉苯企业进行监督检查.结果部分用人单位法制意识淡薄,作业场所职业病危害因素检测与评价等管理制度及措施不健全 ,作业场所三苯浓度抽测点合格率为87.1%,其中7家企业三苯浓度超过国家卫生标准,苯白血病检出1例,检出率为17.7/10万.结论用人单位在职业卫生管理方面与国家职业卫生法律、法规要求尚有较大差距,潜在职业危害依然存在.因此,建立有效的职业卫生监督管理机制,是治本之策.     

五行针灸治疗抑郁症50例临床观察

目的观察五行针灸治疗抑郁证的临床治疗效果。方法随机将门诊100例患有抑郁症的病人分为针刺组和西药组,每组50例。针刺组采用五行针灸治疗,西药组口服圣·约翰草提取物片(路优泰)或盐酸文拉法辛缓释片(博乐欣)治疗,治疗前后采用汉密尔顿抑郁量表(HAMD)进行评分以观察疗效。结果经2个疗程治疗之后,针刺组进行汉密尔顿抑郁量表评分与西药组对比,差异明显具有统计学意义(P0.05)。结论五行针灸治疗抑郁证疗效优于西药治疗,是治疗抑郁症的有效方法。     

直肠癌患者D型人格与生存质量及预后的关系

目的：探索直肠癌患者 D 型人格（type D personality）与预后及术后生存质量（quality of life,QOL）之间的关系。方法：D 型人格的分析使用14项 D 型人格量表（DS14）。生存质量（QOL）结果的评估应用 Euro-pean Organization for Research and Treatment of Cancer QLQ - C30和 QLQ - CR38问卷对基线和诊断后6个月的直肠癌患者进行评估。结果：实验入组511名受试者,反应率93%,其中有112名受试者具有 D 型人格（22%）。在基线及诊断后6个月两个时间点的横向比较中,D 型人格组在大部分功能量表及整体健康状态评分方面显著低于非 D 型人格组,并且拥有更严重的症状评分。在诊断后6个月时间点,D 型人格组受试者有更高比例的 QOL 恶化。结论：在直肠癌患者中,D 型人格与不佳的生存质量及预后显著相关。D 型人格是临床实践当中预测直肠癌患者生存质量恶化的重要易感因素。     

PRRX1/FOXM1 reduces gemcitabin-induced cytotoxicity by regulating autophagy in bladder cancer

BackgroundBladder cancer (BC) is a common urological malignancy with high mortality worldwide. Many proteins can influence tumorigenesis by participating in cellular processes. Recently, abundant evidence has illustrated that paired related homeobox 1 (PRRX1) is closely related to the progression and development of human cancers. However, the function of PRRX1 in BC remains poorly understood. The aim of our study is to explore the role of PRRX1 in BC progression.MethodsThe expression of genes (PRRX1, FOXM1, LC3B, and Beclin-1) was examined through real-time quantitative polymerase chain reaction (RT-qPCR) and western blot. The expression of proteins (PRRX1, FOXM1, LC3B, and Beclin-1) was measured through immunohistochemistry. Cell viability and the half-maximal inhibitory concentration were detected using the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) assay. Cell apoptosis was assessed through flow cytometry. Autophagy was tested by GFP-LC3 immunofluorescence assay. Tumors were grown in nude mice in vivo, and the tumor size, volume, and weight were evaluated.ResultsIn our study, PRRX1 was highly expressed in BC tissues and cells, and high PRRX1 expression resulted in poor overall survival in patients with BC. PRRX1 accelerated the viability and hindered the apoptosis of BC cells. It also weakened gemcitabine-induced cytotoxicity and strengthened gemcitabine-induced autophagy. PRRX1 was found to cooperate with forkhead box protein M1 (FOXM1) to influence downstream genes, and FOXM1 was found to regulate Beclin-1 and microtubule-associated protein 1 light chain 3 (LC3) genes to influence autophagy. PRRX1 up-regulated the expression of LC3 and Beclin-1 by cooperating with FOXM1. In rescue assays, FOXM1 reversed the effects of PRRX1 on gemcitabine-induced cytotoxicity and autophagy. Knockdown of PRRX1 enhanced the inhibitive effects of gemcitabine on tumor growth in vivo.ConclusionsPRRX1 reduces gemcitabine-induced cytotoxicity in BC cells by regulating the expression of the autophagy proteins LC3 and Beclin-1. This discovery suggests that PRRX1 may be a useful therapeutic biomarker for BC.     

Patient-Factors Influencing the 2-Year Trajectory of Mental and Physical Health in Prostate Cancer Patients

This study aimed to examine the physical and mental Quality of Life (QoL) trajectories in prostate cancer (PCa) patients participating in the Pros-IT CNR study. QoL was assessed using the Physical (PCS) and Mental Component Score (MCS) of Short-Form Health Survey upon diagnosis and two years later. Growth mixture models were applied on 1158 patients and 3 trajectories over time were identified for MCS: 75% of patients had constantly high scores, 13% had permanently low scores and 12% starting with low scores had a recovery; the predictors that differentiated the trajectories were age, comorbidities, a family history of PCa, and the bowel, urinary and sexual functional scores at diagnosis. In the physical domain, 2 trajectories were defined: 85% of patients had constantly high scores, while 15% started with low scores and had a further slight decrease. Two years after diagnosis, the psychological and physical status was moderately compromised in more than 10% of PCa patients. For mental health, the trajectory analysis suggested that following the compromised patients at diagnosis until treatment could allow identification of those more vulnerable, for which a level 2 intervention with support from a non-oncology team supervised by a clinical psychologist could be of help.
