5-羟色胺(5-HT)参与舌和面口部精细运动的调节。5-HT对舌下神经运动神经元功能的调控表现为兴奋性作用。本文首先采用免疫组化方法,在光镜下观察了大鼠舌下神经核内5-HT免疫阳性纤维的形态和分布特征。又进一步采用荧光逆行追踪结合固定脑片细胞内LuciferYellow染色和荧光免疫组织化学染色,通过共聚焦激光扫描显微镜观察了伸舌肌(颏舌肌、颏舌骨肌)和缩舌肌(茎突舌肌)运动神经元与5-HT免疫阳性纤维及其终末的解剖学关系。结果证明:舌下神经核有较丰富的5-HT阳性纤维,以尾侧段的背侧部和吻侧段的腹侧部较为密集;根据纤维及其膨体的形态特征,本文将此阳性纤维分为三种类型。5-HT免疫阳性膨体样纤维及其终末与舌下运动神经元的胞体、树突分枝和轴丘及轴突起始段形成紧密接触;这些接触多呈不均匀的簇状(由2~7个紧密接触点组成)分布。定量分析发现:在额舌肌神经元和茎突舌肌神经元,除少量分布在胞体(密度为3.4~6.7/1000μm2)外,主要分布在近段和中段树突分权处附近;但是在颏舌骨肌神经元则集中分布在跑体(密度为10.8~17.6/1000μm2)和近侧树突上。以上所见提示:5-HT对舌下神经核整体行为的调节在不同功能的亚核可能有所选择;5-HT调制舌下运动神经元兴奋性的方式可  相似文献

Summary In this report we describe changes in the intracellular redistribution of raf serine/threonine protein kinase (product of the raf proto-oncogene family) in hippocampal neurons following cerebral ischemia in Mongolian gerbils. For immunohistochemical localization studies polyclonal antisera specific for each of the A, B, and Raf-1 isotypes of raf, as well as a pan-raf antisera, were employed. Of these, only sera recognizing B-raf, as well as the general v-raf (raised against the conserved C-terminal region) were positive, indicating that B-raf is the major isotype in this neuronal region. Three different ischemie models were used (repeated 3 times for two min and single 5 or 15 min occlusions, of the common carotid arteries) to demonstrate that ischemie insult causes redistribution of raf protein kinase into the cell nucleus of hippocampal neurons. Increased amounts of raf protein in the nuclei of pyramidal cells following ischemia was confirmed by Western blot analysis of isolated nuclear fractionations. Moreover, an elevation in the level of nuclear raf protein also was detected in the contralateral (i.e. non-occluded hemisphere) neurons of CA1 and CA3 subfields 4 days after the ischemie insult indicating a possible transsynaptic increase in the amount of raf protein along with redistribution. The intranuclear translocation of the immunoreactive material started from the perinucleolar rim and with time extended throughout the nucleus. Enhanced levels and altered redistribution of the raf polypeptide in the nuclei of pyramidal cells of the CA3 subfleld appears to be reversible and returns to the normal level 12 days following the ischemic insult. In addition to triggering the above changes in the intracellular redistribution of raf, ischemie insult also caused an increase in the level of B-raf protein in reactive astrocytes.  相似文献

A close relationship exists between drinking and the release of vasopressin, the two main factors responsible for the maintenance of body water content. Whereas the participation of peripheral factors, such as oropharyngeal stimulation, seems obvious in the metering of fluid intake and in thirst satiation, very little is known about their influence on vasopressin release. In the present experiments, the influence of drinking on vasopressin release was studied using both biochemical and electrophysiological approaches.In one group of monkeys made thirsty by water deprivation, the subsequent drinking of water during a 5–8 min induced: i) a short-term response, consisting of an abrupt fall in plasma vasopressin concentration which was independent of osmolality, occurred at the time of drinking and was partly reversed after the cessation of drinking, and ii) a longer lasting response, consisting of a slow diminution of plasma vasopressin concentration as the intestinal absorption of water progressed. In another group of thirsty monkeys, extracellular recordings were made during drinking from cells which were identified as neurosecretory neurones of the supraoptic nucleus, a number of them being considered vasopressin secreting on the basis of their phasic pattern of firing. Their firing decreased considerably during the periods of water intake and recovered to control levels immediately after-wards.The decrease in vasopressin release at the onset of water intake, the diminution in the firing rate of the neurones, the short latency and the reversibility of these events after cessation of drinking, suggest that a reflex inhibition of vasopressin-secreting neurones occurs which is probably induced by peripheral stimuli and most likely via oropharyngeal or other visceral receptors. It is postulated that this reflex inhibition of vasopressin release may participate in some active manner in the anticipatory mechanisms of thirst satiation.  相似文献

目的研究雌激素的核受体ER-α和ER-β以及催产素、加压素在成年雌性小鼠下丘脑室旁核内的表达。方法采用硫酸镍铵增强显色的免疫组化SP法检测ER-α、ER-β、催产素和加压素在室旁核内的表达。结果雌激素的两种核受体在室旁核内都有表达,但是以ER-β为主(P〈0.001),其免疫阳性产物均在细胞核内,未见核外免疫阳性反应。催产素的免疫阳性产物主要在细胞核周围的胞浆即核周质内,而加压素的免疫阳性物质除了在核周质内有很强的反应外,在突起内也可见很强的免疫反应。ER-α的免疫阳性胞体主要在大细胞部内侧,而ER-β、催产素和加压素的免疫阳性胞体主要在背侧帽部或大细胞部的外侧。结论室旁核内两种雌激素受体可能都参与了对催产素和加压素的调节,但ER-β可能发挥了主要的调节作用。  相似文献

Summary Terminal sites of the spino-olivary fibers (SOFs) were examined by the anterograde transport of wheat germ agglutinin-horseradish peroxidase in the cat. The tracer was injected at various spinal cord levels from the first cervical to the caudal segments. The SOFs derived from the C1-T1 segments terminated medially in the caudal half (levels II–VIII of Brodal) of the medial accessory olive (MAO), which projects to the A zone of the cerebellar cortex, whereas the SOFs derived from the L6-S1 segments terminated laterally in the caudal half (levels I–VIII) of the MAO. No projections were found from the T2-L5 segments to the MAO. In the dorsal accessory olive (DAO), the SOFs terminated at levels III–XIV; the DAO projects to the B zone and the C1 and C3 zones of the cerebellar cortex. The SOFs derived from the C1-C4 segments terminated in the most medial part of the DAO (levels III–XIV), followed laterally by those from the C5-T1 segments. Further laterally, the SOFs derived from the T2-L5 and the L6-S1 segments terminated in the mediolateral order at levels V–XIV. The SOFs from the L6-S1 segments occupied the most lateral part of the DAO. The present study demonstrates that there is a distinct somatotopic termination of the SOFs in the mediolateral order in the caudal MAO and the DAO.  相似文献

Spike activity was studied in 95 neurons in the basal magnocellular nucleus in rabbits during spontaneous behavior and during performance of a conditioned operant response. Nearly half the neurons (48.4%) showed significant (p < 0.05) negative correlations between spontaneous discharges and the power of the frontal lobe EEG delta rhythm; most of these cells could be identified as cholinergic projection neurons. Neurons of this group had predominantly excitatory responses to the conditioned stimulus during performance of the operant task, while the responses to the conditioned stimulus of presumptively non-cholinergic neurons, not projecting to the cortex, were mainly inhibitory. The activatory responses of neurons in the basal magnocellular nucleus to the conditioned stimulus were markedly stronger while the animals performed the operant response as compared with performances in which there was no response to the conditioned stimulus. These results provide evidence that the basal magnocellular nucleus supports the level of waking and attending required for performance of operant conditioned reflex activity.  相似文献

本文应用放射免疫分析方法,测定了16只哮喘豚鼠初级传入系统和中枢内神经激肽β(NKB)的水平。结果表明,与正常对照组(结状神经书3228±11.89,C7~C8段脊神经节23.63±13.26,T1~T2段脊神经节24.25±1819,T3~T5段脊神经节25.53±12.34,C7~T5段脊髓后角28.65±16.59,孤束核26.34±18.36Pg/gwettissue)相比较,哮喘豚鼠初级传入系统和中枢内(结状神经节43.26±12.35,C7~C段脊神经节32.53±17.63,T1~T2段神经书32.25±20.65,T3~T5段脊神经节34.36±15.33,C7~T5段脊髓后角39.53±2028,孤束核3662±17.85Pg/gwettissue)NKB的含量明显高于对照组(P<0.05)。结合NKB在呼吸道的作用,这些结果提示,初级传入系统和中枢内的NKB可能参与哮喘发病的病理生理机制。  相似文献

The distribution of cells of origin of ascending and descending internuclear connections in the trigeminal sensory nuclei was studied by the retrograde horseradish peroxidase technique in the cat. The termination of collaterals of these ascending axons was also studied by the anterograde transport of horseradish peroxidase. Following injections of horseradish peroxidase into the ventral part of the principal sensory nucleus and the adjacent reticular formation many small neurons were labeled ipsilaterally in the whole area of the caudal portion of the nucleus interpolaris and in laminae III and IV of the nucleus caudalis. Labeled neurons were also found in laminae I and V. Injections limited to either nucleus oralis, the ventral part of the principal sensory nucleus and the medial parabrachial nucleus labeled similar types of neurons in the above regions with a topographic relationship; neurons in the dorsal part of the nuclei caudalis and interpolaris project, dorsally, to rostral portions of the trigeminal sensory nuclei while those in the ventral part of the nuclei caudalis and interpolaris project ventrally. Anterograde labeling of axons arising from the nucleus caudalis demonstrates that the axons ascend in the intranuclear bundles and the adjacent reticular formation, and give off collaterals to the nuclei interpolaris and oralis, and the ventral part of the principal sensory nucleus. Injections limited to the nucleus caudalis labeled small neurons in the rostral portion of the nucleus oralis and the caudal portion of the nucleus interpolaris. The present study suggests that these ascending and descending internuclear connections of the trigeminal sensory nuclei may modulate transmission of afferent inputs to various projection sites, such as thalamus, superior colliculus, cerebellum and spinal cord.  相似文献

Summary In an attempt to clarify the mechanisms involved during synchronous discharge of magnocellular neurosecretory cells of the supraoptic and paraventricular nuclei, extracellular action potentials were recorded from 149 single units located in the rat paraventricular nucleus. Responses of the cells to stimulation of the ipsilateral supraoptic nucleus were recorded and it was observed that 7% of paraventricular nucleus neurones could be antidromically identified as projecting to the supraoptic nucleus. Excitatory responses were recorded from 40% of cells tested and were probably mediated by an interneurone population. Inhibitory responses were recorded from 37% of the cells and may have been mediated by direct projections from the supraoptic nucleus. No differences in responses were recorded from cells identified as projecting to the median eminence or neurohypophysis as compared with the rest of the population tested. However, more of the phasically firing putative vasopressin-secreting cells, (58%) were excited than were continuously active cells (38%). Delivery of short trains of high-frequency stimulation induced bursts of discharge from 93% of cells so tested, regardless of response to single shock stimulation. These burst discharges showed a late onset as compared with responses following single shock stimulation. The results suggest that a complex of intra- and internuclear interactions serve to regulate the neurosecretory activity of magnocellular cells within the supraoptic and paraventricular nuclei.  相似文献
