目的 :了解促排卵治疗对黄体中期子宫内膜雌激素受体 (estrogenreceptor,ER)、孕激素受体 (progesteronereceptor,PR)及其mRNA表达的影响。方法 :应用光学显微镜检测着床窗口子宫内膜、采用免疫组化及原位杂交方法检测正常及多囊卵巢综合征 (polycysticovariansyndrome ,PCOS)妇女促排卵治疗后子宫内膜黄体中期ER、PR及其mRNA的表达 ,放射免疫法测定同期血中雌激素 (estradiol,E2 )及孕激素 (progesterone,P)水平。 结果 :正常妇女黄体中期子宫内膜组织形态 2 2例中有 2 0例为分泌中期改变 ,2例为分泌早期改变。ER、PR弱表达 ,ERmR NA及PRmRNA的表达也较弱 ,而PCOS妇女治疗后黄体中期氯米芬 9/人绒毛膜促性腺激素组及氯米芬 /绝经期促性腺激素 /人绒毛膜促性腺激素组各 15例分别有 8/ 15及 7/ 15光镜下为分泌中期改变 ,其它为分泌早期改变。ER、PR的表达增强 ,其基因的表达也明显增强 ,血清P的水平相对较低。促性腺激素释放激素激动剂 /绝经期促性腺激素 /人绒毛膜促性腺激素组共 10例 ,E2 、P及P/E2 接近对照组 ,上述标志物的表达与对照组相似。但 3组PCOS分泌中期子宫内膜均有不同程度的间质增生、致密 ,腺体与间质发育不同步 ,且间质致密处ER、PR及其mR NA表达均下降。结论 :氯米芬 /人绒毛膜促性腺 相似文献

应用原位分子杂交技术研究卵巢恶性肿瘤组织中VEGFmRNA和bFGFmRNA的表达及其意义。结果发现 :浆液性腺癌和内胚窦癌两者表达阳性率高于粘液性腺癌和颗粒细胞瘤 ;临床分期Ⅰ期和未转移病例两者表达阳性率低于临床分期Ⅲ~Ⅳ期和转移病例。结果提示VEGFmRNA和bFGFmRNA表达与卵巢恶性肿瘤病理类型、临床分期和转移发生关系密切 ,阳性表达者可能预后较差。 相似文献

The ongoing COVID-19 pandemic is raising great concern all over the world. The recent introduction of vaccines has offered reason for optimism, however, new issues have arisen, such as vaccine reluctance. The safety of vaccines for pregnant women is one of the most serious of these concerns. The purpose of this review article is to provide updated international vaccine recommendations, results of ongoing studies and clinical trials, and the role of gynecologists in counseling the women to understand the risks versus benefits as well as form an informed decision towards vaccine acceptance for COVID-19.Although COVID-19 infection increases the risk of severe morbidity and mortality in pregnant women, pregnant women were not included in the initial vaccine trials. As a result, safety information is scarce. Nations have differing recommendations, though many have recently approved the COVID-19 immunization in pregnancy following a risk-benefit analysis. The Joint Committee on Vaccination and Immunization (JCVI) of the United Kingdom recently approved an mRNA vaccination for pregnant women. Vaccination is recommended by the CDC, ACOG, ARFM, and WHO. From pharmacological studies, platelet-activating factor (PAF) has been proposed as a retrograde messenger for long-term potentiation (LTP) in the hippocampal CA1 region. We re-examined a possible contribution of PAF to LTP with a more specific approach using mice deficient in the PAF receptor. The PAF receptor-deficient mice exhibited normal LTP and showed no obvious abnormality in excitatory synaptic transmission. We also performed pharmacological experiments on the wild-type mice. Two structurally different antagonists of PAF receptors had no effects on LTP. Furthermore, the application of PAF itself caused no detectable changes in excitatory synaptic transmission. The autosomal, recessive obesity of ob/ob mice is associated with hypercorticosteronemia and amelioration of most symptoms of obesity following adrenalectomy. Increased adrenocorticotropic hormone (ACTH) secretion has been hypothesized on the basis of several reports of higher pituitary ACTH content in ob/ob mice compared to lean littermates. However, the only measurement of ACTH blood concentration found lower levels in ob/ob mice than in leans suggesting that hypercorticosteronemia might result solely from an enhanced adrenal response to ACTH and also suggesting that the ob/ob's elevated pituitary ACTH content might be due to decreased ACTH secretion rather than increased ACTH synthesis. In our study, basal serum ACTH levels were higher in ob/ob males and females compared to sex-matched lean littermates. Anterior pituitary ACTH synthesis was also elevated as indicated by increased content of ACTH and proopiomelanocortin mRNA in obese mice of both sexes; however hypothalamic corticotropin-releasing factor content was not different in lean and obese mice. Basal serum ACTH and corticosterone (CS) levels showed normal circadian rhythm in both phenotypes and sexes, but the circadian increase in CS level was much greater in obese mice than in leans despite equal serum ACTH increases in the two phenotypes. Ether stress at both peak and trough of the circadian rhythm also stimulated much larger serum CS increases in obese mice even though ACTH increases were again equal in the two phenotypes. Taken together, these results strongly indicate that ob/ob mice have increased synthesis and secretion of pituitary ACTH despite the presence of chronically elevated serum CS. This hyperactivity of the hypothalamo-pituitary-adrenal axis appears to be most pronounced in ob/ob females since pituitary ACTH content was equal in obese males and females despite much higher circulating CS levels in the females. Furthermore, the results also indicate an enhanced response to ACTH by the adrenal cortex of the obese mouse. Thus, ob/ob mice exhibit abnormal hypothalamo-pituitary-adrenal axis function with hyperactivity occurring at the level of pituitary ACTH synthesis/secretion as well as at the level of adrenocortical response to ACTH. 相似文献

目的 :观察氨氯地平对巨噬细胞基质金属蛋白酶 2 ,9(MMP 2 ,MMP 9)的mRNA表达及分泌影响 ,探讨氨氯地平是否具有稳定动脉粥样硬化斑块 ,减少其破裂的功能。方法 :收集小鼠腹腔巨噬细胞与不同浓度的氨氯地平进行培养 ,采用RT PCR方法检测细胞MMP 2 ,MMP 9mRNA的表达 ;用明胶酶谱法检测细胞培养上清液中MMP 2 ,MMP 9的活性 ,在蛋白分泌水平观察氨氯地平对MMP 2 ,MMP 9分泌影响。结果 :5 μg·L- 1浓度的氨氯地平对MMP 2 ,MMP 9的mRNA表达和分泌几乎没有影响 (P >0 .0 5 ) ;15 μg·L- 1浓度可以完全抑制MMP 2 ,MMP 9的mRNA表达 ,对MMP 9分泌有明显的抑制作用 (P <0 .0 5 ) ,对MMP 2的抑制作用较轻 (P >0 .0 5 ) ;浓度为 30 μg·L- 1时 ,完全抑制MMP 2 ,MMP 9的mRNA表达 ,对MMP 2 ,MMP 9分泌的影响由于细胞破裂的影响而不能定论。结论 :中等浓度的氨氯地平可能具有稳定动脉粥样硬化斑块 ,减少其破裂的功能。 相似文献

目的 :筛选和克隆胃印戒细胞癌与正常胃黏膜细胞差异表达基因片段。方法 :采用mRNA差异显示法 (mRNAdifferentialdisplayPCR ,DD PCR) ,对比胃印戒细胞癌组织与正常胃黏膜组织mRNA的表达差异 ,克隆胃印戒细胞癌差异片段 ,经过RNA的斑点杂交、测序以及序列分析和同源性比较。结果 :胃印戒细胞癌与正常胃黏膜中存在明显的基因表达差异 ,发现与胃癌相关的差异表达条带 16条 ,其中 7条为缺失表达条带 ,9条为过度表达条带。对其中 4条差异表达条带进行克隆、测序 ,经序列分析及同源性比较和RNA的斑点杂交 ,表明确认其中 1条为GenBank/BLAST中尚未收录的片段。结论 :胃印戒细胞癌的mRNA差异显示证明 ,该EST序列可能在胃癌的发生发展中具有一定作用 相似文献
