血液灌流对硫线磷和硫酸阿托品的吸附作用

Objective To study on adsorption effect of cadusafos and atropine sulfate by hemoperfusion.Method Hemoperfusions were performed for sheep blood samples with cadusafos and atropineby through imitated extracorporeal closed circulating perfusion apparatus.Residual cadusafos was determined by gas chromatography and residual atropine was determined by high performance liquid chromatography.Result Dose of adsorption agent was 0.5,1.0 and 1.5 g,respectively.Two hours after hemoperfusion with membrane coated activated charcoal,clearance rate of cadusafos in 3 groups all exceeded 90%.and clearance rate of atropine sulfate was 61.9%,84.9%,88.9%,respectively.One and a half hours after hemoperfusion with HA230 absorption resin,clearance rate of eadusafos in 3 groups all exceeded 90%,and clearance rate of atropine sulfate was 88.0%,97.2%,98.4%,respectively.Three hours after hemoperfusion with membrane coated activated charcoal,The concentration ratio of cadusafos and atropine sulfate in blood promoted to 10.1 times,and the ratio was 6.7 times after hemoperfusion with HA230 absorption resin.Conclusion It suggested that cadusafos were mostly removed from blood after 1.5～2.0hours hemoperfusion with membrane activated charcoal or HA230 absorption resin.The concentration ratio of cadusafos and atropine sulfate in blood will increased after hemoperfusion.     

PENA方法的建立及与ELISA IgM检测CMV的比较

目的　介绍一种敏感、稳定、快速、简便的实验室检测CMV的方法 ,同时探讨该种新方法与ELISA检测CMV方法的优、缺点。方法　对 5 5 2例病人应用间接荧光免疫法测定细胞核中的特异病毒早期抗体 (PENA)和ELISA法测定IgM抗体。结果　PENA方法 :强阳性 88例 ,阳性率 15 4 9% ,弱阳性 2 73例 ,阳性率 5 9 4 6 % ;ELISA -IgM方法 :阳性 34例 ,阳性率6 16 %。结论　PENA方法操作简便 ,与ELISA方法相比较 ,可对CMV感染进行早期测定及诊断 ,并可区分既往感染和即时感染 ,具有敏感性和稳定性 ,是测定小儿CMV感染的一种较好的方法     

Packing and Uniaxial Alignment of Liquid Crystalline Oligofluorenes

Summary: Liquid crystalline oligomers of 9,9‐bis(2‐ethylhexyl)fluorene of defined degree of polymerization 4, 5, 6, and 7 were investigated by X‐ray diffraction in the non‐oriented and in the aligned state. The diffraction data give evidence for a smectic B type phase for all of the oligomers. Quenching below the glass transition does not change the structure of the liquid crystalline phase. This allows to align spin‐coated films of these oligomers on rubbed polyimide substrates to give monodomain films. These are stable against thermal disordering below T_g, e.g. at room temperature. The degree of alignment is characterized by the dichroic spectra and polarized fluorescence spectra. Dichroic ratios and polarization ratios increase substantially with the chain length and values as high as D = 23 and P = 41 are obtained for the heptamer. The type of packing of the oligomers in the LC phase is discussed based on the X‐ray single crystal structure of models. In one such model the packing of the 2‐ethylhexyl side chains could be fully resolved, while the other model reveals the torsional angle between adjacent fluorene units in the same molecule as 144.2° which corroborates earlier work based on fiber diffraction of corresponding polyfluorenes.

目的测量2450MHz频率下蒸馏水和不同浓度NaCl溶液的复介电常数,用以判定系统的可靠性和稳定性.方法根据微扰法得到复介电常数的测量公式,然后通过实验对已知参数的蒸馏水和NaCl溶液进行测量,并计算其复介电常数和电导率,验证计算结果的可靠性和稳定性.结果对计算结果进行统计分析和误差分析,其均值、标准差和变异系数表明,最终结果可与M.I.T标准较好地吻合.结论这种系统可较好地测量2450MHz频率下高损耗介质的复介电常数,测量系统的主要误差是由介质体积的误差引起的,因此精确测量介质体积是测量生物组织等高损耗介质复介电常数的关键.     

Determination of glucokinase and hexokinase activity in liver extracts previously purified on Molselect G-50 dextran gel

Hexokinase and glucokinase activity in the supernatant of a rabbit liver homogenate obtained at 18,000g was determined by a spectrophotometric method. Preliminary purification to remove low-molecular-weight components by gel filtration on Molselect G-50 dextran was shown to prevent reduction of NADP unconnected with the hexokinase reaction.Presented by Academician of the Academy of Medical Sciences of the USSR V. Biochemical characterization of I-Ak proteins from mutant antigen-presenting B-cell--B-lymphoma hybridomas

Chemically induced mutants of an I-Ak,d expressing antigen-presenting B-cell--B-lymphoma hybridoma have recently been generated by immunoselection in vitro and were found to possess alterations in some of their serologically and functionally defined I-Ak region dependent functions. In order to identify at the structural level the origin of the differences in serological and functional properties of these mutants, I-Ak molecules from several of these mutant hybridomas were compared biochemically to wild-type I-Ak polypeptides by two-dimensional gel electrophoresis and high-pressure liquid chromatographic tryptic peptide analyses. Two-dimensional gel electrophoresis indicated that no major structural alterations, resulting in changes in mol. wt or charge, had occurred in the Ak alpha or Ak beta polypeptides from the mutant cells. Likewise, Ak alpha peptide maps of the mutants were indistinguishable from the normal Ak alpha peptide maps. However, two of the three mutants studied did exhibit one additional peptide in their Ak beta peptide maps. These results suggest that the major deficiencies in T-cell-activating functions of these mutants are a result of a limited alteration in the Ak beta polypeptide primary structure.     

Production of genetically modified cells expressing specific transgenes by retroviral vectors for gene therapy

Summary Techniques and protocols are described for the generation of genetically modified cells that can be used for gene therapy. Primary fibroblast cultures are established from skin biopsies, maintained in culture, frozen for long-term storage, and retrieved when necessary. Retroviral packaging cell lines are generated by transfection of DNA into retroviral packaging cells by calcium-phosphate precipitation method or by lipofection method. To generate cell lines expressing high titer virus, individual colonies of cells are cloned and the virus titer is determined. Virus collected from packaging cells expressing high titer virus is then used to infect primary fibroblasts. To obtain fibroblast cell lines expressing high amounts of transgenes, individual cells can be cloned to generate clonal cell lines. Although the methods described here are for fibroblasts, the same methods or modification of the methods can be used for other cell types.     

Distribution of galanin immunoreactivity in the central nervous system and the responses of galanin-containing neuronal pathways to injury

Radioimmunoassay and immunocytochemistry were used to study the distribution of galanin, a novel 29 amino acid porcine intestinal peptide, in the central nervous system of the rat and pig. The pattern of distribution was similar in the two species, with the highest concentrations of galanin-like immunoreactivity found in the neurohypophysis, hypothalamus and sacral spinal cord. Immunocytochemical studies of these regions localized galanin-like immunoreactivity to cell bodies in the paraventricular and supraoptic nuclei of the hypothalamus, to fibres in the pars nervosa and to numerous cell bodies and fibres in the dorsal horn of the spinal cord. On both gel and high pressure liquid chromatography, galanin-like immunoreactivity in rat and pig nervous tissue eluted as a single peak in a position similar to purified procine intestinal galanin standard. Surgical and pharmacological manipulations in the rat suggest the presence of galanin in afferent fibres. An increase of galanin-like immunoreactivity was observed in the sacral spinal cord of the rat following thoracic spinal cord transection. Thus galanin-like immunoreactivity in the brain is mainly localized in the hypothalamopituitary region. The decrease of galanin-like immunoreactivity in the dorsal horn of the spinal cord, following dorsal rhizotomy and pre-treatment of rats with capsaicin, indicates that many of the fibres, which are of small diameter, may well be derived from spinal sensory neurones.     

To what extent is the tibia-calcaneum angle a reliable measurement of the triceps surae length? Radiological correction of the torque-angle curve (III)

Summary Previous papers gave some methods for the reliable measurement of the tibia-calcaneum angle. It is of common use to evaluate the physical properties of triceps surae on the basis of torque-angle curves. However this method is reliable only if each tibia-calcaneum angle corresponds to a defined distance between the insertions of the muscle in subjects of the same height. Evidence is given by radiological measurements that this correspondance is correct in normal children. However, this is no longer true in certain cerebral palsied children because of abnormal translation of the calcaneum and/or abnormal ratio of bone sizes. In this case the torque-angle curves do not define properly the torque-length curves. A method of correction is given. 酵母双杂合系统AD端阴离子交换蛋白C-末端表达质粒的构建

利用PCR方法，从阴离子交换蛋白1（AE1）全长cDNA中扩增出约350bp c末端cDNA片段，测序后将其克隆至pGADT7载体上，用醋酸锂法构建好的pADT7-AE1-c末端转染酵母菌HA109，观察其在选择性培养基上的表达情况。结果表明，获得了530bp AE1c－末端cDNA,pGADT7-AE1-c末端对酵母无毒性，不能激活检测基因，可作为酵母双杂合系统中的靶基因。
