Na currents recorded from inside-out macropatches excised from Xenopus oocytes expressing the subunit of the rat brain Na channel IIA show at least two distinguishable components in their inactivation time course, with time constants differing about tenfold ( h1 = approx. 150 s and h2 = approx. 2 ms). In excised patches, the inactivation properties of Na currents changed with time, favoring the faster inactivation kinetics. Analysis of the fast and slow current kinetics shows that only the relative magnitudes of h1 and h2 components are altered without significant changes in the time constants of activation or inactivation. In addition, voltage dependence of both activation and steady-state inactivation of Na currents are shifted to more negative potentials in patches with predominantly fast inactivation, although reversal potentials and valences remained unaltered. We conclude that the two inactivation modes discerned in this study are conferred by two states of Na channel the interconversion of which are regulated by an as yet unknown mechanism that seems to involve cytosolic factors.  相似文献

Calcium-activated chloride currents were studied by the patch-clamp technique in vascular smooth muscle cells (VSMC) isolated from human mesenteric arteries. Bath application of 20 mM caffeine caused the cell membrane to depolarize by a calcium-activated inward current that peaked to –654±230 pA (holding potential –50 mV). Cell-attached, at the same time inwardly directed single-channel currents were detected with an amplitude of –0.22 pA. In open-cell-attached patches channel activity was triggered by elevating [Ca2+]i to 10 M. At –60 mV the mean amplitude of the current was –0.24 pA and the mean open time of the channels was 28 ms. Plotting the amplitude of the current versus the test potential yielded a single-channel conductance of 2.8±0.5 pS. The currents disappeared when [Cl] was reduced from 150 mM to 5 mM at the cytosolic side of the inside-out patch at a holding potential of -60 mV (calculated reversal potential –58 mV) suggesting that the calcium-activated current was a chloride current. This suggests that, in human mesenteric VSMC, elevation of [Ca2+]i activates a low-conductance chloride channel, which may mediate the agonist-induced depolarization of the cell membrane.  相似文献

We provide here 29 genetic variations, including 28 novel ones, in five genes that are potentially involved in the excitement of cardiomyocytes: we found 4 in KCNA10, 2 in KCNK1, 8 in KCNK6, 11 in SLC18A1 (VMAT1), and 4 in SLC6A2 (norepinephrine transporter). We also examined their allelic frequencies in a Japanese population of long QT syndrome-affected and nonaffected individuals. The behavioral effects following intrastriatal MPP+, the neurotoxic metabolite of MPTP, were evaluated in mice. Bilateral injections of 10 cro;g MPP+ to mice previously trained in the shuttle box paradigm produced a 66% decrease in striatal dopamine and significant deficits in all measures of conditioned avoidance responding. In addition, although these mice showed no deficits in baseline rotorod performance, challenge with the cholinergic agonist oxotremorine revealed that MPP+-treated mice exhibited an increased sensitivity to the disruptive effects of the drug at each dose and time point. Finally, MPP+-treated mice also exhibited an increase in tremor induced by 0.1 and 0.15 mg/kg oxotremorine. These observations are discussed in reference to idiopathic parkinsonism.  相似文献

Two cases are reported of liver angiosarcoma occurring among polyvinyl chloride (PVC) extruders from a small Italian factory producing PVC bags and other containers. The possibility that PVC extrusion carries a risk of liver angiosarcoma is important because of the very large number of people working with extruding, manufacturing and handling PVC, as compared with the number of people working in PVC polymerization and/or VC production. In the past, the level of vinyl chloride (VC) concentration in PVC extrusion workplaces has been thought to be "safe".  相似文献

Rats were subjected to 4 h continuous or intermittent exposure to vinyl chloride (VC) at different time-weighted average concentrations (15, 50, 150, 500 and 15,000 mg/m3). Hepatic non-protein sulfhydryl content (NPSH) and excretion of thiodiglycolic acid (TdGA) in urine were determined. VC at concentrations from 50 mg/m3 to 15,000 mg/m3 caused a dose-dependent depression of NPSH, but no difference in the magnitude of this depression induced by continuous or intermittent exposure at the same average concentration of VC was noted. At average concentrations of 50 mg/m3 and 150 mg/m3, the urinary excretion of TdGA under continuous exposure did not differ from that under intermittent exposure, whereas at VC concentrations of 500 mg/m3 and 15,000 mg/m3 it was higher following continuous exposure.  相似文献

Summary Interactions between the putative calcium entry promotor Bay k 8644 and both -1 and 1-adrenocepter mediated increases in diastolic pressure were studied in the pithed normotensive rat. The 1-adrenocepter mediated pressor responses elicited by B-HT920, TL-99, DP-6,7-ADTN and B-HT958 were potentiated by Bay k 8644, reflected by a leftward shift and an increase in the maximum of the log dose-pressor respinse curves. The -1-adrenocepter effects elicited by cirazoline, methoxamine, (–)-amidephrine, St 587, (–)-phenylephrine and Sgd 101/75 were less enhanced by Bay k 8644. Only a leftward shift of the dose-response curves was observed, which was most pronounced for (–)-phenylephrine and Sgd 101/75. The -1 and 2-adrenocepter pressor components of (–)-noradrenaline were similarly distinguished by Bay k 8644 as observed for the selective -1 or 2-adrenocepter agonists.Effects of Bay k 8644 on the increase in diastolic pressure mediated by B-HT 920, St 587 and cirazoline were also studied after pretreatment with the calcium entry blocker nifedipine. After additional pretreatment with nifedipine the potentiation by Bay k 8644 observed for B-HT 920 and St 587 was more pronounced. The presence of nifedipine had no effect on the interaction between Bay k 8644 and cirazoline.It is concluded that Bay k 8644 behaves as a mirror image of nifedipine. The observation that Bay k 8644 enhances 2-adrenocepter mediated pressor effects more effectively than 1-adrenocepter increases in diastolic pressure is in accordance with the hypothesis of the more pronounced calcium dependency of 2-adrenocepter mediated pressor responses. The data obtained for ceptor mediated pressor responses. The data obtained for St 587 and (–)-phenylephrine are in apparent contradiction to the finding that the pressor responses to the former drug are more markedly inhibited by calcium entry blockade than those of the latter. It is suggested that St 587 employs calcium channels which are already maximally modulated and that (–)-phenylephrine makes use of calcium channels which are in a rather inactive state. The hypothesis is put forward that the intrinsic activity of 2-adrenocepter agonists reflects their ability to bring calcium channels in an active state.  相似文献

A micropolarimeter interfaced to a liquid chromatograph is shown to be suitable for selective monitoring of the optically-active components in complex samples. When an optically-active eluent is used, indirect determination of even optically-inactive materials is possible, down to the level of 10 ng of an injected component. If a second chromatogram is obtained using the racemic analogue of the optically-active eluent, quantitation can be achieved without standards and without prior analyte identification. This concept is also applicable to the refractive index detector, the absorption detector and the conductivity detector in the special case of ion chromatography, and the ultrasonic detector in gas chromatography.  相似文献

以硅酸钠、氯化铁为原料制备聚硅氯化铁(简称PFSC)混凝剂。重点考察Fe/SiO2摩尔比对PFSC混凝效果的影响。结果表明:PFSC的混凝效果优于聚硅酸(PS),且Fe/SiO2摩尔比对PFSC的混凝效果有较大的影响。确定Fe/SiO2摩尔比为1:1.5时,其混凝效果最佳,且用药量少而范围广。为制备高效的PFSC提供了优化条件。  相似文献
