目的:观察前列宁胶囊(QC)对基质金属蛋白酶-2(MMP-2)介导的细胞外基质(ECM)的影响,探讨QC对BPH的治疗机制。方法:SD大鼠随机分为空白组,BPH模型组,QC低、中、高观察组,建立BPH模型,QC干预,ELISA法检测大鼠血清MMP-2、FN、Collagen IV、LN; BPH-1细胞培养,分别加入MMP-2和未加MMP-2刺激因子,QC干预,MTT法观察细胞活性,Q-PCR及Western-Blot分别检测MMP-2、FN、Collagen IV、LN基因及蛋白表达(未加MMP-2),加入MMP-2组检测FN、Collagen IV、LN。结果:QC各剂量组大鼠血清中的FN、LN、Collagen IV含量降低,MMP-2升高(P 0. 05 or P 0. 01);BPH-1细胞培养QC干预,加入及未加入MMP-2刺激因子细胞活力均有下降,以48 h后最明显;加入MMP-2刺激因子FN、Collagen IV、LN基因及蛋白表达明显低于未加MMP-2刺激因子,差异有统计学意义。结论:QC对BPH具有治疗作用,QC调控MMP-2介导细胞外基质FN、Collagen IV、LN基因和蛋白表达可能是其治疗BPH机制之一。  相似文献

应用PCR方法对68例狼疮性肾炎(LN)患者血管紧张素转换酶(ACE)基因多态性分布进行观察,结合临床表现及随访资料进行分析。结果表明,LN患者ACE基因DD基因型的发生频率明显高于正常人(P<0.01),DI型患者肾脏损害较重,预后较差。说明LN临床表现可能与ACE基因多态性有一定的联系。  相似文献

目的 探讨肥胖症儿童肝纤维化指标透明质酸(HA)、层粘连蛋白(LN)、血清Ⅲ型前胶原肽(PCⅢ)、Ⅳ型胶原(CⅣ)与血清甘氨酰脯氨酸二肽氨基肽酶(GPDA)活性的变化及其在临床上的应用价值.方法 对2015年6月至2016年2月在丽水市妇幼保健院儿科门诊诊断为肥胖症的159例6 ~15岁肥胖症儿童(肥胖症组)和42例9~ 14岁正常健康儿童(正常对照组)进行肝纤维化指标与血清GPDA活性的比较,并对结果进行分析.结果 与正常对照组相比,肥胖症组HA、PCⅢ水平均明显升高(t值分别为7.403、5.405,均P<0.05),以PCⅢ升高明显;肥胖症组GPDA活性差异有统计学意义(t=8.965,P<0.01).肥胖症患儿治疗前后GPDA活性、HA及PCⅢ指标比较,各检测项目均出现降低,差异均有统计学意义(t值分别为6.452、5.684、4.369,均P<0.05).PCⅢ与GPDA呈正相关(r=0.602,P<0.05).结论 肥胖症儿童GPDA活性与PCⅢ水平的改变是其肝脏病变的早期敏感指标,两者的动态监测,对于诊断、疗效观察有一定的临床意义.  相似文献

目的 探讨大黄酚对慢性乙型病毒性肝炎(CHB)大鼠肝功能损伤及肝脏纤维化的影响,并分析相关作用机制。方法 60只雄性SD大鼠随机分为对照组、模型组及大黄酚低、高剂量(7.5、30 mg/kg)组和拉米夫定(10 mg/kg)组,每组12只。除对照组外,其余各组均尾iv乙型肝炎病毒(HBV)抗原构建CHB模型,造模成功后个给药组ip相应药物。采用全自动生化分析仪检测大鼠血清中天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、总胆红素(TBIL)水平;酶联免疫吸附(ELISA)法检测大鼠血清中透明质酸(HA)、层黏连蛋白(LN)、羟脯氨酸(HYP)水平;苏木精–伊红(HE)染色及Masson染色观察大鼠肝脏组织病理形态学及纤维化;Western blotting法检测大鼠肝脏组织中相关蛋白表达。结果 与模型组比较,大黄酚7.5、30 mg/kg组和拉米夫定组大鼠血清中ALT、AST、TBIL、HA、LN、HYP水平均降低(P<0.05),大鼠肝组织病理性损伤和胶原纤维沉积显著改善,肝组织中α-平滑肌肌动蛋白(α-SMA)、I型胶原α1基因(COL1A1)、葡萄糖调节蛋白(GRP)78、GRP94及磷酸化真核生物翻译起始因子2α(p-eIF2α)/eIF2α、磷酸化蛋白激酶R样内质网激酶(p-PERK)/PERK均显著降低(P<0.05);且大黄酚30 mg/kg组、拉米夫定组大鼠血清和肝组织中上述指标均显著优于大黄酚7.5 mg/kg组(P<0.05)。结论 大黄酚可能通过调控内质网应激减轻CHB大鼠肝功能损伤,改善大鼠肝纤维化。  相似文献

目的 探讨锁骨上无瘤淋巴结对食管癌病人手术治疗的影响。方法 术前发现锁骨上有无瘤淋巴结并行手术切除的食管癌病人 4 5例 ,以 73例锁骨上无淋巴结的食管癌手术病人为对照 ,对术后病理情况进行对比研究。结果 ⑴两组病人肿瘤浸润深度相比 χ2 =10 .39,P <0 .0 1;⑵两组病人淋巴结阳性率相比 χ2=7.12 ,P <0 .0 1;⑶两组病人淋巴转移情况相比 χ2 =6.76,P <0 .0 1。结论 对于术前发现锁骨上有无瘤淋巴结的病人 ,术中应加强淋巴结的清扫 ,尤其是上段的淋巴结 ,且不论术后病理情况如何 ,均应放疗 ,区域包括锁骨上淋巴结区。  相似文献

Objective: To investigate whether laminin 5 (LN5) might be a predictor in lung cancer patient treated with gefitinib and estimate the underlying mechanisms. Methods: LN5 and epidermal growth factor receptor (EGFR) mRNA expression level were detected in the tumor tissues of lung cancer patients who underwent surgery resection prior to gefitinib treatment. EGFR exon 19 and 21 mutation status was also detected in these specimens. The association between LN5, EGFR mRNA expression level, EGFR mutation and gefitinib treatment response were evaluated. In vitro study were carried by adding exog- enous LN5 and gefitinib to A549 lung cancer cell line, and Western-blotting was performed to investigate the phosphorylation level of EGFR,Ak, and Erk. Results: The disease control rate according to LN5 mRNA level was 52.9% for the below cut- point group, and 17.6% for the above cut-point (P = 0.009). The in vitro study showed that exogenous LN5 can neutralize the inhibition of phosphor-Akt by gefitinib. Conclusion: Patients with lower LN5 mRNA level would likely benefit from gefitinib. In vitro study indicated that the inhibition of Akt induced by gefitinib might be reversed by LN5. These results provide important insights into the molecular mechanisms underlying sensitivity to gefitinib in lung cancer patients.  相似文献

BackgroundHigh-frequency transcranial random noise stimulation (hf-tRNS) is a neuromodulatory technique consisting of the application of alternating current at random intensities and frequencies. hf-tRNS induces random neural activity in the system that may boost the sensitivity of neurons to weak inputs. Stochastic resonance is a nonlinear phenomenon whereby the addition of an optimal amount of noise results in performance enhancement, whereas further noise increments impair signal detection or discrimination.ObjectiveThe aim of the study was to assess whether modulatory effects of hf-tRNS rely on the stochastic resonance phenomenon, and what is the specific neural mechanism producing stochastic resonance.MethodObservers performed a two-interval forced choice motion direction discrimination task in which they had to report whether two moving patches presented in two temporal intervals had the same or different motion directions. hf-tRNS was administered at five intensity levels (0.5, 0.75, 1.0, 1.5, and 2.25 mA).ResultsThe results showed a significant improvement in performance when hf-tRNS was applied at 1.5 mA, representing the optimal level of external noise. However, stimulation intensity at 2.25 mA significantly impaired direction discrimination performance. An equivalent noise (EN) analysis, used to assess how hf-tRNS modulates the mechanisms underlying global motion processing, showed an increment in motion signal integration with the optimal current intensity, but reduced motion signal integration at 2.25 mA.ConclusionThese results indicate that hf-tRNS-induced noise modulates neural signal-to-noise ratio in a way that is compatible with the stochastic resonance phenomenon.  相似文献

BackgroundOne in ten children and young people have a clinically diagnosed mental health disorder or emotional and behavioural problems. Children and young people with poor mental health are at increased risk of short-term and long-term poor social and economic outcomes. Improving the wellbeing of children is one of the key goals of the Well-being of Future Generations Act in Wales. The aim of this study was to understand children's views about what is important to mental wellbeing in the school setting.MethodsBetween Nov 29, 2017, and May 28, 2018, six focus groups, which included 62 pupils (33 girls and 29 boys) aged 5–18 years, were invited to discuss what matters to children to support their mental wellbeing. Five of the focus groups were held at schools identified by the Healthy Schools Coordinators across Wales, and one was held at the Children in Wales office for children who were excluded or not in mainstream schools. The schools were located in a mixture of urban and rural settings in south Wales. We did a thematic analysis of all the qualitative data gathered from the focus groups.FindingsThematic analysis identified fourteen themes: understanding the mental wellbeing needs of children who attend each school; fostering good working relationships with clear respect between pupils and staff; being able to raise issues without fear of judgement; dealing quickly and effectively with bullying; having enough free time in school for unstructured activities and lunch; feeling prepared for tests and exams; having a strong and effective pupil voice; staff enabling a calm classroom environment; prioritising pupil well-being; prioritising staff well-being; monitoring the reasons for staff turnover and acting on this; using support services to best effect to benefit well-being; celebrating diversity; and helping pupils cope at predictable transition periods. The most frequently raised themes were fostering good working relationships with clear respect between pupils and staff; having a strong and effective pupil voice; prioritising staff wellbeing; monitoring the reasons for staff turnover and acting on this; celebrating diversity; and helping pupils cope at predictable transition periods.InterpretationThe themes identified by the children were used as a basis for the new Wales Healthy School Scheme criteria. The outcomes of the thematic analysis directly influenced the criteria associated with mental wellbeing, giving children a clear voice in the process.FundingNone.  相似文献
