目的探讨乙酰肝素酶在人体冠状动脉稳定性斑块和不稳定性斑块中的分布及其在不稳定性斑块形成中的可能作用。方法选取经临床和病理解剖证实的急性冠状动脉综合征病例进行免疫组织化学检测并通过计算机分析系统进行量化分析。结果不稳定性斑块组乙酰肝素酶的聚集程度明显高于稳定性斑块组。结论乙酰肝素酶与斑块不稳定性存在一定联系,可能参与了斑块的去稳定过程。 相似文献

AIM: To disclose the mechanisms that accelerate or limit tumor invasion and metastasis in gastric cancer patients. METHODS: The heparanase expression, continuity of basement, degree of infiltration by dendritic cells and lymphocytes in gastric cancer tissues from 33 the early and late stage patients were examined by immunohistochemistry, in situ hybridization and transmission electron microscopy. RESULTS: Heparanase mRNA expression in the late stage patients with gastric cancer was stronger than that in the early stage gastric cancer patients. In the early stage gastric cancer tissues, basement membrane (BM) appeared intact, whereas in the late stage, discontinuous BM was often present. The density of Sl00 protein positive tumor infiltrating dendritic cells (TIDC) in the early stage gastric cancer tissues was higher than that in the late stage. The infiltrating degree of tumor infiltrating lymphocytes (TIL) in the early stage patients whose tumor tissues contained a high density of TIDC was significantly higher than that in the late stage gastric cancer tissues patients with a low density of TIDC. There were few cancer cells penetrated through the continuous BM of cancer nests in the early stage gastric cancers, but many cancer cells were found outside of the defective BM of cancer nests in the late stage. CONCLUSION: Our results suggest that strongheparanase expression is related with the degradation of BM which allows or accelerates tumor invasion and metastasis. However, high density of TIDC and degree of infiltration by TIL are associated with tumor progression in human gastric cancers. 相似文献

目的探讨胃癌组织中肝素酶和血管内皮生长因子(VEGF)-C蛋白表达及其临床意义。方法采用免疫组化S-P法检测97例原发性胃癌组织、癌旁组织及20例正常胃黏膜组织中肝素酶和VEGFC蛋白的表达,并分析其与临床病理特征和预后的关系。结果胃癌组织肝素酶和VEGF—C蛋白表达阳性率分别为61.9%和66.0%,显著高于癌旁组织的7.2%和5.0%,正常胃组织的8.3%和5.0%(P值均〈0.01);肝素酶表达与肿瘤分化程度无关,但与肿瘤直径、淋巴结转移、静脉侵犯、淋巴管侵犯、远处转移、浆膜面受累和TNM分期等密切相关;VEGF—C表达与肿瘤直径、分化程度、静脉侵犯及远处转移等无关,但与淋巴结转移、淋巴管侵犯、浆膜面受累和TNM分期等密切相关;肝素酶和VEGF—C阳性表达组术后生存率明显低于阴性组;在胃癌中肝素酶和VEGF—C阳性表达呈正相关。结论肝素酶及VEGF-C蛋白的阳性表达,可作为胃癌预后不良的参考指标。 相似文献

目的探讨胃癌及癌前病变中碱性成纤维细胞生长因子(bFGF)、成纤维细胞生长因子受体-2(FG-FR-2)及乙酰肝素酶(Hpa)的表达及其与胃癌发生发展的关系。方法选择广西医科大学第一附属医院2001—2005年手术切除和内镜活检的胃黏膜标本145例,其中慢性浅表性胃炎(CSG)组30例、胃黏膜肠上皮化生(IM)组29例、不典型增生(Dys)组31例、胃癌(GC)组55例,采用免疫组化SP法检测各组bFGF、FGFR-2及Hpa的表达。结果bFGF、FGFR-2的表达CSG组低于其余3组(P<0.05),IM组低于Dys组和GC组(P<0.05);Hpa在GC组表达显著高于其他3组(P<0.01);bFGF、FGFR-2及Hpa的表达与胃癌浸润深度、淋巴结转移、远处转移及TNM分期有关(P<0.01)。结论bFGF、FGFR-2及Hpa可能参与胃癌的发生发展过程,并可能加速胃癌的浸润和转移。 相似文献

目的利用人胃癌细胞株SGC7901细胞,探讨乙酰肝素酶反义寡核苷酸对人胃癌细胞株SGC7901中表达碱性成纤维细胞生长因子(bFGF)的影响。方法采用脂质体介导乙酰肝素酶反义寡核苷酸转染人胃癌细胞株SGC7901细胞,采用免疫细胞化学法观察细胞内bFGF的变化。结果脂质体介导转染乙酰肝素酶反义寡核苷酸后,SGC 7901胃癌细胞内乙酰肝素酶mRNA表达下降,bF-GF阳性表达率由转染前的72.23%降至40.25%。转染后细胞bFGF表达明显减少。结论转染乙酰肝素酶反义寡核苷酸可以有效抑制胃癌细胞bFGF的表达。 相似文献

Background
/Objectives: High heparanase level was shown in maliganant tumor; however, whether or not heparanase may serve as a sensitive marker to monitor response to anticancer treatment is still unknown.Methods
In the pilot study, heparanase mRNA expression in peripheral blood mononuclear cell fraction (PBMC) and activity in plasma and urine were detected by quantitative real time RT-PCR and heparan-degrading enzyme assay in 31 pancreatic cancer patients.Results
Heparanase mRNA and activity in samples from cancer patients were significantly higher than that in healthy donors. Both heparanase mRNA and activity in plasma and urine decreased significantly in 17 patients who underwent R0 resection, but increased remarkably in 6 patients when recurrence or metastasis occurred (P < 0.05). However, those who underwent R1 or R2 resection in 6 patients kept stable. For 8 patients who received chemotherapy, heparanase mRNA and activity in plasma and urine decreased in each of the samples (P < 0.05). Patients with high heparanase mRNA (≥a cutoff value of 1.84) in PBMC and activity in plasma (≥1.30U/ml) were associated with a poor postoperative survival (P = 0.02 and P = 0.04).Conclusions
Heparanase mRNA in PBMC and activity in plasma are closely correlated with therapeutic responsiveness and survival time, indicating that heparanase level in blood might be a sensitive but non-specific marker to monitor patients' response to anticancer treatment and to predict survival. 相似文献90.

目的 研究口腔鳞状细胞癌组织中细胞角蛋白19(cytokerantin-19,ck19)、乙酰肝素酶(heparinase,HPA)的表达及其检测价值,为临床治疗提供数据参考.方法 选取2013年1月至2014年1月期间在我院接受治疗的52例口腔鳞状细胞癌患者为研究对象,详细记录每位患者的各项资料,采集标本、检测ck19以及HPA的表达情况,分析其检测意义.结果 HPA的水平与患者年龄、性别无显著关系,与患者TNM分期、组织学分级、淋巴结是否转移明显相关;ck19的表达与患者的年龄、性别无关,与TNM分期以及组织学的分化存在明显的关联,P<0.05;HPA诊断的灵敏度为94.0% (47/50),特异度为50% (1/2),ck19诊断的灵敏度为91.3%(42/46),特异度为33.3% (2/6),对比两组结果,差异无统计学意义,P>0.05.结论 ck19、HPA的表达与患者口腔鳞状细胞癌的发展有着明显的相关性,能够作为潜在的诊断标志物,具有较高的诊断价值. 相似文献
