Objectives

Allergic rhinitis (AR) is a chronic upper respiratory tract disease that inflames the mucous membranes of the nose and occurs when circulating inflammatory cells including eosinophils and basophils migrate to and accumulate in the inflammation area by passing through the interstitium and capillary walls. To pass through these barriers, the inflammatory cells degrade extracellular matrix proteins. Matrix metalloproteinases (MMPs) released by inflammatory cells mediate the degradation of these proteins. MMPs have synthetic inhibitors and doxycycline, a tetracycline antibiotic, inhibits MMPs. This study investigated the efficiency of intranasal doxycycline in decreasing the symptoms and inflammatory cell infiltration in an animal model of AR.

Methods

AR was created in female Wistar rats by repeated intranasal challenge with ovalbumin by intraperitoneal injection. For 15 days, topical intranasal doxycycline was administered one hour before ovalbumin administration. Following intranasal administration, nasal symptoms were scored and the nasal mucosae of all rats were evaluated histopathologically. To investigate tissue changes, hematoxyline-eosin and Alcian blue/periodic acid Schiff stains were used. As well, cilia loss, goblet cell changes, vascular congestion, vascular proliferation, inflammatory cell infiltration, eosinophil infiltration and the degree of hypertrophy in chondrocytes were evaluated with light microscopy.

Results

Typical symptoms of AR were decreased by intranasal doxycycline administration. These effects were stable after repeated intranasal ovalbumin administration. Histological evaluation of doxycycline treated rats did not reveal typical inflammatory changes associated with AR.

Conclusion

MMPs may have crucial functions in AR and topical intranasal doxycycline, which decreases inflammatory cell infiltration, may offer an alternative therapy for AR.  相似文献

A new spectrofluorimetric method is described for the determination of doxycycline, based on modified enzyme-amplified lanthanide luminescence. Under the optimum conditions, Eu3+–doxycycline forms a ternary complex with lysozyme in close proximity and lysozyme can remarkably enhance the characteristic fluorescence intensity of Eu3+ at 612 nm in doxycycline–Eu3+ binary complex. The enhanced fluorescence intensity is in proportion to the concentration of doxycycline. The limit of detection is 1.28×10−8 mol l−1, with a linear range from 1.7×10−7 to 1.7×10−6 mol l−1. Interferences of other coexisting substances were studied. The developed method was successfully applied to the determination of doxycycline in serum, urine and real samples. The mechanism of fluorescence enhancement was also studied.  相似文献

目的:为评价盐酸多西环素胶囊治疗寻常痤疮的疗效及不良反应。方法:对60例寻常痤疮随机分为二组治疗,A组(30例)用盐酸多西环素胶囊治疗,B组(30例)用二甲胺四环素胶囊治疗, 治疗时间为21天,结果:治疗前后,A组和B组的炎性皮损数减少明显(P<0.05),盐酸多西环素胶囊组无明显不良反应,结论:盐酸多西环素胶囊治疗寻常痤疮有效,副作用少。  相似文献

目的比较两种治疗方案(左旋氧氟沙星200mg2次/d共7天与头孢三嗪250mg单剂量肌注加强力霉素100mg2次/d共7天)治疗单纯性淋病合并衣原体和/或支原体感染各30例的有效性和安全性。方法采用随机、开放、对照研究,试验组及对照组各30例。结果两组病例的痊愈率分别为83.33%和86.67%,总有效率为90%和93.33%,差异无显著性(P>0.05);副反应发生率分别为3.33%和16.67%,差异有显著性(P<0.05)。结论口服左旋氧氟沙星方案可作为治疗淋病合并衣原体和/或支原体感染的治疗方法之一。  相似文献

目的观察多西环素(Doxycycline)对人大肠癌LS174T细胞体外侵袭的作用。方法以人工重组基底膜(Matrigel)体外侵袭实验观察药物对细胞体外侵袭能力的影响;SDS-聚丙烯酰胺凝胶电泳法(明胶酶谱法)观察对金属蛋白酶分泌的影响。结果多西环素抑制LS174T大肠癌细胞体外侵袭,在多西环素浓度为5mg/L,10mg/L,20mg/L时,每100倍视野穿过的细胞数分别为(101.3±6.7)个,(83.7±5.7)个和(42.0±2.0)个,与对照组有显著差异,抑制率分别为17.9%、32.0%和65.9%,呈现剂量依赖性;在5mg/L的低浓度下多西环素即可抑制基质金属蛋白酶-2(MMP-2)的分泌。结论多西环素能够抑制人大肠癌LS174T细胞的侵袭,在低于抗菌浓度下即可抑制基质金属蛋白酶-2的分泌,具有成为抗肿瘤侵袭药物的潜力。  相似文献

目的探讨脊柱布氏杆菌感染的临床特征表现,比较多西环素+利福平、多西环素+利福平+左氧氟沙星的抗菌效果。方法回顾性分析2017年1月至2019年1月我院收治的78例脊柱布氏杆菌感染病人,分析其临床特征表现,并根据抗菌方案的不同分为观察组和对照组,每组39例,对照组行多西环素+利福平治疗,观察组行多西环素+利福平+左氧氟沙星治疗,观察两组治疗效果、临床症状及不良反应发生情况,采用疼痛数字评定量表(numerical rating scale,NRS)对病人关节疼痛程度予以评估。结果78例病人均有不同程度脊柱活动受限,其中关节疼痛78例(100.00%)、发热78例(100.00%)、乏力25例(32.05%)、多汗34例(43.59%)、肌肉痛12例(15.38%);CT检查显示病椎及相邻椎体边缘有不同程度破坏及明显骨质增生,病椎间隙狭窄;MRI检查显示病变椎旁软组织异常T1WI低信号、T2WI高信号;布氏杆菌特异性血清凝集试验均呈阳性,血白细胞水平异常11例(14.10%),C反应蛋白水平异常15例(19.23%),红细胞沉降率增快7例(8.97%)。治疗6周后,观察组总有效率为94.87%,明显高于对照组(79.49%,P<0.05);观察组体温、NRS评分明显低于同期对照组(P<0.05);观察组不良反应发生率为15.38%,与对照组(10.26%)比较,差异无统计学意义(P<0.05)。结论脊柱布氏杆菌感染的临床特征表现多样,对临床诊断及治疗有指导作用,相比于多西环素+利福平抗菌方案,多西环素+利福平+左氧氟沙星抗菌方案治疗效果更为显著,且具有较高安全性,值得推广。  相似文献

Summary Seventy-five patients with neurological symptoms of Lyme borreliosis were randomly assigned to intravenous treatment with either penicillin G or doxycycline. After 12 months the treatments were equally effective regarding the clinical picture and laboratory findings. No patient was considered to be a treatment failure. However, one-third of the patients showed delayed recovery, particularly after a longer primary disease duration. A slow recovery, lasting years, was typical of subacute or chronic borreliosis.  相似文献

本文采用膜控法制备盐酸强力霉素缓释微丸。实验表明,本品的体外溶出符合零级动力学过程(K_r~0=20.5 mg/h);贮存期为两年,胃刺激性明显低于普通片剂(p<0.001),体内动力学过程符合表观零级吸收与一级消除的单室模型。体内数据经NONLIN计算机程序处理,求得各项参数如下:K_a=58.08 mg/h,K=0.032 h-1,V_d=82.211,t_(max)=3.94 h,c_(max)=2.30μg/ml。按强力霉素的常规用药方案给药,本品与市售普通片生物等效。本品的体内外数据具有显著的相关性(p<0.001)。  相似文献

As a pharmaceutical pollutant, doxycycline causes contamination when enters into the environment. In this research MIL-53(Fe), and its magnetic hybrid MIL-53(Fe)/Fe3O4 were synthesized and employed for removal of doxycycline from aqueous solutions. The adsorbents were characterized by XRD, SEM, BET, FTIR, EDAX, VSM and TG-DTG technique. The effect of different variables such as DOC concentration, pH, contacting time, and adsorbent dose on the removal efficiency was studied and under optimized conditions the adsorption capacity of 322 mgg−1 was obtained. The adsorption process was kinetically fast and the equilibration was attained within 30 min. The used adsorbent was easily separated from the solution by applying external magnetic field. The regenerated adsorbent retained most of its initial capacity after six regeneration steps. The effect of ionic strength was studied and it was indicated that removal of doxycycline from salt-containing water with moderate ionic strengths was quite feasible. Langmuir, Freundlich, Tempkin and Dubinin–Redushkevich isotherms were employed to describe the nature of adsorption process. The sorption data was well interpreted by the Longmuir model.  相似文献
