近日德克萨斯州州长Rick Perry愤怒的澄清了他与默克公司之间的关系。此前他发布了一项行政命令,要求德克萨斯州所有在校女生接种宫颈癌疫苗Gardasil。     

胰腺癌黏蛋白4基因修饰的树突状细胞疫苗构建及对胰腺癌细胞的免疫效应

Objective To obtain the dendritic cells ( DC)-based vaccine modified by adenovirus containing MUC4 gene , and evaluate the anti-tumor efficacy of DC vaccine to pancreatic tumor cells. Meth-ods The mRNA sequence of tumor associated antigen, MUC4, was obtained from NCBI, and MUC4 se-quence was acquired through the restriction enzyme sites and over lap PCR, then subcloned into adenovirus plasmid to create recombinant adenovirus ( rAd-MUC4) . The DCs were infected by rAd-MUC4 virus and then stimulated the lymph cells from the same donor to induce MUC4 specific cytotoxicity T lympbocytes ( CTL) . The efficacy of CTL was analyzed by LDH releasing assay. Elispot was used to detect the IFN-γ release. Results The recombinant adenovirus containing MUC4 ( sv12) gene was obtained. The MUC4-induced CTL could specifically kill the Capan-1 pancreatic tumor cells [ ( 13. 7±6.0)% , ( 21.4± 4. 7)% , (36.1±9. 5)% at ratios of 10: ,20: ,40: ] , higher than MCF-7 and Bxpc-3 cells respectively, P < 0. 05. The spots number of CTL induced by rAd-MUC4 was ( 139.1±23.3) , more than GFP and PBS control group,P<0.05. Conclusion The Muc4 gene modified DC vaccine could induce the proliferation of CTL, which provided a significant cytotoxicity to HLA-matched MUC4 positive tumor cell lines in vitro.     

甲基苯丙胺抗体、疫苗以及检测试纸研究进展

苯丙胺类兴奋剂(amphetamine-type stimulants,ATS)是21世纪主要流行滥用的药物之一,目前社会上流行的"冰毒","摇头丸"等属于此类毒品,主要有甲基苯丙胺(methamphetamine,METH),亚甲基二氧基甲基苯丙胺(MDMA)等.在我国,此类毒品无论是在舞厅、迪吧或其它公共娱乐场所和地下色情场所等都很容易获得,导致滥用人群日益扩大[1],影响社会治安和社会稳定.虽然世界各国在药物滥用防治工作中投入了大量的人力、物力、财力,但收效甚微,对ATS依赖者目前治疗多采用强制戒毒、对症治疗和辅以心理治疗等方法,但治疗时间长,复吸率高[1-4],对于高危人群目前还没有一种有效的预防手段.检测ATS多用高效液相色谱(HPLC)、气相色谱-质谱联用(GC-MS)、薄层色谱(TLC)等[2]方法,其设备昂贵,难以开展现场检测,应用范围受限,并不适合于现场快速便捷检测,因而探索对ATS更有效的预防、治疗方法和更便捷的现场检测方法,极具现实意义.     

狂犬疫苗致过敏性肺炎1例

过敏性肺炎多由于吸入微生物或动植物蛋白引起,笔者在临床上却遇到1例注射狂犬疫苗所致的过敏性肺炎，因其少见,报道如下。

据Medscape．com6月13日报道（原载Immunity2006：24：643—656），慢病毒侵入表皮会直接转染皮肤源性树突状细胞，使编码的抗原出现有效且持久的提呈。

Multi-parameter prediction of HLA class Ⅰ restricted cytotoxic T lymphocyte epitopes for human proliferating cell nuclear antigen

Objective To predict the HLA class Ⅰ restricted cytotoxic T lymphocyte epitopes for human proliferating cell nuclear antigen (PCNA). Methods By using 3 epitope prediction databases which including MHC binder ( BIMAS and SYFPEITHI databases) and proteasome cutting site ( PAProC databases), the epitopes of PCNA were predicted by analyzing several parameters and methods. Results After comprehensive analysis,we have obtained two possible HLA-A0201 restricted CTL epitopes SMSAD-VPLV (228-236, score 447 633. 595 ) and LINEACWDI ( 22-28, score 127 966.779);two HLA-A24 re-stricted CTL epitopos DYEMKLMDL ( 113-121, score 563 994.9 ) and EFARICRDL ( 143-151, score 40 540.7);two HLA-A1101 restricted CTL epitopes LTSMSKILK (72-80, score 1334. 2680 ) and DVPLV-VEYK (232-240,score 736.9236). Conclusion The multi-parameter epitope prediction method is feasi-ble for cytotoxic T lymphocyte epitope prediction.     

人禽流感疫苗试验前景乐观

据Medscape．com 12月15日报道（Additional reporting by Noelle Mennella and Ben Hirschler），一种针对H5N1禽流感病毒的试验性疫苗的早期试验显示它使人体产生了良好的免疫反应，并且非常安全。     

114例儿童麻疹临床特点及流行病学分析

为分析本地区麻疹的临床及流行病学特征,对1999年7月～2003年5月114例住院麻疹患儿的临床资料进行流行病学分析.结果显示114例麻疹患儿中107例未预防接种过麻疹疫苗.男孩多于女孩,发病年龄4个月～14岁,以婴幼儿及学龄前儿童居多、发病季节以冬春为多占88.5%,113例有发热,107例有不同程度皮疹,临床症状重,并发症发生率高.
