PROBLEM: β2 glycoprotein I (β2GPI) physiologically binds to negatively charged phospholipids (PLs) and is a natural regulator of the coagulation cascade. Thrombotic clinical complications and recurrent fetal loss associated with autoimmune antiphospholipid (aPL) antibodies are thought to be related to their binding to β2GPI-PL complex and interference with the physiological function of β2GPI. METHOD OF STUDY: To investigate the effect of aPL on β2GPI-PL interaction, we studied the binding of biotinylated β2GPI to cardiolipin (CL) by enzyme-linked immunosorbent assay (ELISA) in the presence and absence of purified aPL immunoglobulin G (IgG) antibodies. RESULTS: Adding five different aPL IgG antibodies with different levels of aPL activity isolated from the sera of five patients with aPL-associated recurrent fetal death greatly increased the binding of biotinylated β2GPI to CL-coated plates. The optical densities (ODs) were 0.635, 0.890, and 1.265 in the presence of three aPL IgG antibodies, compared to 0.425 in the absence of aPL IgG. In contrast, normal human IgG had no enhancing effect. The OD was 0.480 and 0.425, respectively. The enhancement of β2GPI binding to CL by aPL IgG correlated with the titers of aPL antibodies. The use of phosphate-buffered saline with increasing salt concentrations as a washing buffer for the ELISA resulted in more stable binding of β2GPI to PL in the presence of aPL IgG. CONCLUSIONS: These findings suggest that the binding of autoimmune aPL antibodies to β2GPI-PL complex results in abnormally tighter interaction between β2GPI and PLs, which may lead to physiological dysfunction of β2GPI as a regulator of coagulation. 相似文献

An effective definition of psychopathology would be helpful in distinguishing between normal and abnormal behavior and would provide guidance for important public policy decisions. Overinclusive definitions, such as the capacity/disability formulation, do not provide meaningful direction for defining need or assessing eligibility for insurance or other entitlements. Coping failures due to lack of skill, inadequate preparation, or other factors should be distinguished from those due to dysfunction or disturbance of psychological mechanisms. Venous occlusion plethysmography (VOP) is a noninvasive technique widely employed for the detection of deep-vein thrombosis.
Previous reports that VOP outflow curves are closely fit by a first-order exponential suggest that venous compliance and resistance
are nearly constant. Typically, however, the venous compliance function has a sigmoid shape; in addition, the resistance in
a collapsing tube must increase. This paradox was resolved by the surprising finding that for realistic nonlinear compliance

Summary A new stochastic model of the surface EMG is suggested and the spectral density of the surface EMG is studied theoretically and experimentally to confirm the validity of this model.Theoretical results show that while the contraction level is not so high, the shape of the spectral density (distribution) does not change and its amplitude is directly proportional to the motor unit firing frequency and recruitment.To illustrate the theoretical results, experiments were carried out for rectus femoris and biceps brachii. The surface EMG was lead off by bipolar surface electrodes. 目的通过系统的钴60放射计划,对大鼠免疫功能抑制的合适剂量进行评估,为干细胞移植研究提供合适的的动物模型。方法SD大鼠随机分为12.5Gy、10Gy、7.5Gy、5Gy4个剂量组,10只/组。计算放射后2个月的死亡率。检测白细胞数量,评价各剂量组动物的白细胞参考值范围。结合死亡率和白细胞参考值范围,确定合适的放射剂量。通过对外周血中性粒细胞(PMN)吞噬实验和外周血白细胞移行抑制实验(MIT),对细胞免疫功能进行检测。结果行12.5Gy照射的大鼠,3d内全部死亡;10Gy放射后的大鼠在1个月内全部死亡;7.5Gy照射的大鼠2个月内死亡1只;5Gy照射的大鼠没有死亡情况发生。设定95%作为可信区间,计算白细胞数量参考值范围(109个/L),未放疗组:16.978+1.96×6.46;5Gy放疗组:4.93+1.96×0.72;7.5Gy放疗组:2.313+1.96×0.782;10Gy放疗组:1.03+1.96×0.507。t检验表明,随着放疗剂量的增加,各实验组的白细胞数量显著减少。对于细胞免疫功能的检测,外周血中性粒细胞(PMN)吞噬实验结果表明,机体全身免疫功能降低;外周血白细胞移行抑制实验(MIT)表明,机体T细胞免疫功能降低。结论7.5Gy可以一定程度上抑制大鼠的免疫功能,为合适的放疗剂量。 相似文献

The initiation of a reentrant circuit requires a zone of slow conduction and a zone of unidirectional block. This study used
computer model conditions under which partial coupling between segments of cardiac Purkinje tissue resulted in unidirectional
block. The structure used was one-dimensional and divided into three segments: a middle segment of variable length coupled
to two long (semi-infinite in concept) segments. The DiFrancesco-Noble equations represented the ionic currents of the membrane.
The results show that the possibility of unidirectional block depends on the size of the middle segment and the coupling resistances
between the segments. No combination of coupling resistances allowed unidirectional block for middle segments with a length
of two space constants (4 mm) or longer. Unidirectional block occurred for many combinations of coupling resistances as the
length of the middle segment decreased to around half a space constant (1 mm). The number of length combinations that caused
unidirectional block decreased again as segment length further decreased. These results provide a possible mechanism of unidirectional
block for situations where islands of viable tissue are connected through nonviable tissue, such as in a healed myocardial

The naive Bayes model makes the often unrealistic assumption that the feature variables are mutually independent given the class variable. We interpret a violation of this assumption as an indication of the presence of latent variables, and we show how latent variables can be detected. A new noninvasive method of measuring the structure and the electrical properties of bilayered biological tissues was evaluated as a potentially useful diagnostic means for detecting changes in subcutaneous tissues. First, the input impedance of an open-ended coaxial probe radiating into a bilayered model was calculated using a full-wave method, the results showed that the evanescen higher order modes do not have a significant influence on the reflection coefficient of muscle layer surface. Then, it was clearly proven that the phase shift and the modulus of the reflection coefficient of muscle layer surface depending on the frequency are useful to estimate the thickness of fat layer and the electrical properties of muscle respectively. The experimental results showed an excellent agreement with the theoretical relationship between the phase shift and the thickness. The sensitivity of estimation of the electrical properties of muscle was shown to be not enough for differentiating between normal and diseased deep tissue because of noises from the experimental systems. 相似文献

Conjugation of DNA fragments to protein carriers by glutaraldehyde: immunogenicity of oligonucleotide-hemocyanin conjugates 总被引:1,自引:0,他引:1
The practical realization of the concept of specific immunotherapy for systemic lupus erythematosus (SLE) has been hampered, thus far, by an inability to link DNA fragments to carrier protein. In this paper, a novel technique is described, in which glutaraldehyde is the linking agent. A 2-stage method was used to link oligonucleotides to a soluble protein carrier, such as keyhole limpet hemocyanin (KLH) or human gamma globulin (HGG), whereas a 1-stage technique was sufficient to link oligonucleotides to sheep red cells. Both the ultraviolet absorbance spectrum and diphenylamine assay demonstrated that oligonucleotides were coupled to soluble protein. The conjugate of oligonucleotide to protein carrier appears to be recognized by anti-DNA antibody since oligonucleotide linked to either KLH or HGG inhibited the binding of anti-DNA antibody in vitro, and oligonucleotide-coupled sheep cells are agglutinating by seropositve sera from lupus patients. In addition, oligonucleotide-KLH raised hemagglutinating antibody to denatured DNA in C57BL/6, DBA/2 or NZB mice, as well as IgG antibody as detected by SPRIA in C57BL/6 and DBA/2 mice. The significance of this new method for the development of an antigen specific therapy of SLE is discussed. 相似文献
