以P(St-Am·MG)对染料活性艳红(X-3B)溶液进行絮凝试验,考察了高聚物剂量及外加盐对絮凝效果的影响,并与无疏水基团的阳离子聚丙烯酰胺PAm·MG的絮凝能力进行了比较。发现絮凝体系无外加盐时,高聚物与染料的作用主要是通过静电力;有外加盐时,高聚物的电荷受到屏蔽,高分子链卷曲,PAm·MG的絮凝效果受到较大影响。而P(St-Am·MG)与X-3B之间存在疏水相互作用,受影响较小,此时P(St-Am·MG)的絮凝性能优于PAm·MG。用平衡渗析法研究了不同温度时甲醇和脲存在下P(St-Am·MG)与X-3B相互作用的热力学。证明了它们之间确实存在疏水相互作用。  相似文献

Summary Temporal cortex from 14 cases of Alzheimer-type dementia and 6 cases of Down's syndrome, all selected for severe Alzheimer pathology, was homogenised in distilled water, NaOH, or sodium dodecylsulphate (SDS) containing 0.1% -mercaptoethanol. The homogenates were stained with Congo red, and the neurofibrillary tangles and plaque cores were counted under crossed-polarisation microscopy. The number of tangles and plaque cores in the water-treated extracts was not related to age, sex, postmortem interval or duration of dementia. The number of tangles after extraction in SDS or NaOH, as a percentage of tangles in water-treated extracts, was 57±25 (mean±SD) for 1% SDS, 43±17 for 5% SDS and 37±22 for 0.2 M NaOH. Plaque cores were essentially insoluble in all three agents. The percentage of tangles insoluble in 1% SDS did not correlated with age or post-mortem interval but decreased with increasing duration of dementia. Enhanced tangle solubility with increasing duration of dementia suggests that the nature of tangles changes with time; one possibility is that this reflects transformation of intracellular to extracellular tangles. Paired helical filament (PHF) length and the number of repeats per PHF were measured in electron micrographs of PHF prepared with and without treatment by 1% SDS. There was no significant multimodality of PHF length to suggest that PHF broke at regular intervals. The mean repeat length (PHF length/number of repeats) was greater for PHF isolated in the presence of 1% SDS than in its absence, showing that SDS affects ultrastructure by untwisting PHF. An untwisting process may also occur in vivo producing the straight filaments found, together with PHF, in tangles and neurites.Supported by Miss E. During (January) 1986–(May) 1988, we examined 272 eyes in 136 rhesus monkeys in the closed Cayo Santiago colony of the Caribbean Primate Research Center of the University of Puerto Rico. Seventy-eight eyes were less than 10 years of age. One hundred and ninety-four were aged 10–28 years. The fundi were examined and photographed. Fluorescein angiography was performed in some eyes. Selected cases were evaluated for acuity loss by recording of pattern-evoked retinal and cortical signals. Light and electron microscopy were used to evaluate the pigment epithelium of some animals. Thirty-eight percent of all eyes had posterior pole drusen. Incidence was highly age-related. When late-stage lesions were found, we did not see neovascularization, but late hyperfluorescence was consistent with degenerative scarring and atrophy. Electrophysiology demonstrated moderately reduced acuity in the presence of numerous macular drusen. Electrooculograms were low normal. Histopathology showed changes identical to those reported in human age-related macular degeneration. No eyes less than 10 years of age had confluent drusen or disciform-like lesions. The incidence of drusen in samples of some social groups was much higher than others.  相似文献

目的 :通过测定妊高征 (PIH)患者红细胞免疫粘附 (RCIA)调节因子活性 ,探讨PIH患者红细胞免疫功能改变的原因。方法 :测定 40例PIH患者和 30例正常孕妇的RCIA促进因子活性和抑制因子活性及红细胞补体受体花环率(RBCC3bRR% )、红细胞免疫复合物花环率 (RBCICR % )。结果 :与正常孕妇相比 ,PIH患者RCIA抑制因子活性明显升高 ,RBCC3bRR%及RBCICR %明显降低 (P <0 .0 1) ,RCIA促进因子活性无明显变化 (P >0 .0 5 )。结论 :PIH患者红细胞免疫功能原发低下 ,其原因可能与RCIA抑制因子活性增高有关  相似文献

The effect of acute infection of mice with lactic dehydrogenase virus (LDV) on two major functions of peritoneal macrophages was tested. Using a macrophage-dependent T cell proliferative assay to test the antigen-presenting capacity of LDV-infected macrophages we found that LDV impairs the capacity of antigen-presenting cells to trigger memory T lymphocytes. Endocytosis of antigen by LDV-infected macrophages was similar to that of uninfected cells. In addition, the proportion of intracellular antigen versus membrane-bound antigen in LDV-infected cells were similar to that observed in uninfected mice. It appears therefore, that the impaired immunogenic effect of LDV-infected macrophages results from reduced immunogenicity of the membrane-bound antigen.Testing the phagocytic activity of peritoneal macrophages we found that the uptake of radiolabeled antibody-coated sheep erythrocytes or bacteria (E. coli) by infected cells was similar to that by uninfected macrophages. In addition, LDV failed to affect the ability of peritoneal macrophages in a nitroblue tetrazolium reduction reaction which serves as an alternative parameter for measuring phagocytic activity. Our results support the assumption that LDV, which probably propagates in the cells of the reticuloendothelial system, impairs some of the immunogenic functions of macrophages and thereby affects macrophage-dependent immune responses.  相似文献

Summary Various doses of tetanus toxin were injected into three hind leg and two fore leg muscles of the rat. The neuromuscular transmission was tested by recording the mass action potential of the muscles elicited by a single electrical stimulus to the motor nerve after strong symptoms of local tetanus had developed. The muscle responses were depressed and blocked at lower toxin doses in the fast tibialis anterior than in the mixed gastrocnemius latemlis, while blocking of the slow soleus required the highest dose. The extensor carpi radialis and the flexor carpi ulnaris muscles showed medium sensitivity. In all five muscles the contraction time was measured and correlated with its individual minimal blocking dose. The more phasic (i.e., the faster) the muscle, the more sensitive its neuromuscular transmission was to tetanus toxin. The proportional distribution of red, white, and intermediate fibres, which are associated with specific end-plate types, was evaluated for the five muscles. The percentage of white fibres in the muscles displayed a very good negative correlation with the blocking dose. The relation between structures of end-plates and effects of tetanus toxin were analysed and it is suggested that the differences in sensitivity to tetanus toxin in the neuromuscular transmission in the five muscles is determined by a differential distribution of endplates with varying sensitivities to this toxin due to structural properties.This study is a part of a doctoral dissertation submitted by one of the authors (H.K.) to the Faculty of Medicine, University of Göttingen. The influence of hereditary absence of thymus and spleen upon the numbers, organ, and class distribution of background immunoglobulin Ig-secreting cells was studied in mice by means of the protein-A plaque assay. In young adult BALB/c mice the spleen contained the largest number of Ig-secreting cells (about 0.5% ). The absolute number of Ig-secreting cells in the spleen was larger than the estimate for all lymph nodes together. Between 8 and 40 weeks of age, the number of Ig-secreting cells in spleen and lymph nodes increased by a factor of 3, maximally. In the same period, the number of Ig-secreting cells in the bone marrow, however, increased by a factor of 20, so that it became the major site of Ig synthesis. Hereditary absence of the spleen did hardly or not at all affect the number of Ig-secreting cells in the other lymphoid organs. However, the athymic state did affect the organ distribution. The most consistent finding was the decreased number of Ig-secreting cells in the Peyer's patches.The class distribution of Ig-secreting cells was found to be independent of the presence of the spleen, but did depend on the presence of the thymus. Athymic mice had a higher percentage of IgM-secreting cells and a lower percentage of IgA-secreting cells. The percentage of IgG1- and IgG2-secreting cells did not differ clearly between normal and athymic mice. Percent-wise, most IgM-secreting cells occurred in the spleen, whereas most IgG1-, IgG2-, and IgA-secreting cells occurred in the bone marrow, lymph nodes, and Peyer's patches.The specificity repertoire of the background Ig-secreting cells was tested by determining the frequencies of IgM-producing cells with specificity for a panel of six different antigens. These frequencies ranged from 1 in 85 for nitroiodophenyl(NIP)-conjugated sheep erythrocytes (SRBC) till 1 in 1500 for unconjugated SRBC and were found to be the same for the spleen of germ-free and specific pathogen-free (SPF) C3H mice, and for spleen, bone marrow, and thymus of SPF C3H mice.  相似文献

Acute experiments on anesthetized and immobilized cats using intracellular recording were used to study the responses of neurons in the parietal associative cortex to stimulation of the red nucleus. Efferent neurons of the parietal cortex were identified by antidromal activation on stimulation of the intrinsic nuclei of the pons and motor cortex. Oligo- and polysynaptic EPSP in response to stimulation of the red nucleus were seen. 本文道报27例同种异体骨移植兔的红细胞免疫功能指标检测结果。红细胞C_3b受体花环率变化,以异体骨花环率最高,液氮骨其次,AAA骨花环率形成率则在正常对照值范围内变化。结果提示异体骨移植能引起明显的免疫反应,液氮骨可有一定的抗原性,能引起免疫反应,冻干骨免疫反应较低下,而AAA骨则不引起免疫反应。本文对同种异体骨移植后的红细胞免疫功能变化的机理与临床意义进行了讨论。  相似文献

The ontogeny of antigen-specific T suppressor cells in thymus and spleen was analyzed in CBA/Ca mice which were rendered tolerant as neonates by subimmunogenic doses of bovine serum albumin (low-zone tolerance). Activity of T suppressor cells from those mice was assessed by an assay in which spleen cells from animals primed with fluorescein-conjugated human gamma globulin can be stimulated in vitro to produce IgG anti-fluorescein antibodies when cultured in the presence of fluorescein-conjugated bovine serum albumin. Carrier-specific T suppressor cells appear first in the thymus (day 10), and much later (day 30) in the spleen. The data are discussed in connection with the possible role of T suppressor cells during induction of tolerance in newborn mice.  相似文献
