Exploratory behavior (GMA) and habituation rate (IH) were studied in an open-field situation in normal and adrenalectomized rats. Following this procedure the rats were subjected to passive avoidance learning (PA). Wide-spreading individual differences were observed in the exploratory behavior and the tendency of habituation of normal rats. As compared to the normal values, either the adrenalectomy which was performed 24 hr, 7 days and 28 days prior to the experiments or the ACTH and hydrocortisone treatment failed to modify the GMA and the IH significantly. An improvement of PA was found in the normal rats following ACTH and hydrocortisone treatment. In the adrenalectomized animals the hydrocortisone proved to be effective, whereas ACTH did not influence PA. No correlation was found between GMA, IH versus PA values and the influence of ACTH and hydrocortisone administration on these parameters. It is concluded that the direction of PA is unpredictable on the basis of the open-field test performed on R-Amsterdam strain of rats, and the effect of ACTH on passive avoidance learning is mediated through the adrenal glands.  相似文献

Male Sprague-Dawley rats with combined lesions to the dorsal and lateral aspects of the entorhinal cortex in one hemisphere and of the contralateral dorsolateral hippocampal formation were compared with both operated and unoperated controls on three different behaviors, monitored across a 53 day postoperative period. The rats with the combined entorhinal cortex-hippocampal lesions (EH) showed transient hyperactivity in the open field, transient reduction in spontaneous alternation levels in an unbaited T-maze and persistent deficits in learning spatial maze problems. The results of the present experiment are discussed in comparison with those from experiments on rats with bilateral hippocampal lesions and those from experiments on rats with bilateral entorhinal cortex lesions. Although some similarities among these findings suggest that these two brain regions probably function in a coordinated fashion with respect to these behaviors, differences in the various syndromes are also discussed.  相似文献

The paper studies a surface electromyogram (SEMG) decomposition technique suitable for identification of complete motor unit (MU) firing patterns and their motor unit action potentials (MUAPs) during low-level isometric voluntary muscle contractions. The algorithm was based on a correlation matrix of measurements, assumed unsynchronised (uncorrelated) MU firings, exhibited a very low computational complexity and resolved the superimposition of MUAPs. A separation index was defined that identified the time instants of an MU's activation and was eventually used for reconstruction of a complete MU innervation pulse train. In contrast with other decomposition techniques, the proposed approach worked well also when the number of active MUs was slightly underestimated, if the MU firing patterns partly overlapped and if the measurements were noisy. The results on synthetic SEMG show 100% accuracy in the detection of innervation pulses down to a signal-to-noise ratio (SNR) of 10 dB, and 93±4.6% (mean± standard deviation) accuracy with 0 dB additive noise. In the case of real SEMG, recorded with an array of 61 electrodes from biceps brachii of five subjects at 10% maximum voluntary contraction, seven active MUs with a mean firing rate of 14.1 Hz were identified on average.  相似文献

The activating action of liquid BCG vaccine, lyophilized BCG vaccine, and killed BCG cells on macrophages was studied. Activation of mouse peritoneal macrophages was identified by their cytotoxic actionin vitro on syngeneic tumor target cells labeled with51Cr. Macrophages obtained from mice two to three weeks after a single injection of liquid BCG vaccine were shown to have a cytotoxic action on tumor cells. A single injection of lyophilized BCG vaccine or killed BCG cells into mice did not cause activation of their macrophages. Two injections of liquid or lyophilized BCG vaccine caused activation of macrophages much earlier — three to five days after the second injection. Smooth muscle cells of the guinea pig ureter were studied by the double sucrose gap method. An increase in the calcium ion concentration in the Ringer-Locke solution to 22 mM led to hyperpolarization and to a slight increase in the resistance of the membrane. The amplitude of the first spike potential and the height of the plateau were lowered but the amplitude of the oscillations was increased. In sodium-free Ringer-Locke solution, when the action potential in the smooth muscle cells of the ureter was converted into a simple spike potential, a marked increase in amplitude of the action potential was observed with an increase in the calcium ion concentration.Department of Neuromuscular Physiology, A. Angelman syndrome (AS) is caused by several genetic mechanisms that impair the expression of maternally‐inherited UBE3A through deletions, paternal uniparental disomy (UPD), UBE3A pathogenic variants, or imprinting defects. Current methods of differentiating the etiology require molecular testing, which is sometimes difficult to obtain. Recently, computer‐based facial analysis systems have been used to assist in identifying genetic conditions based on facial phenotypes. We sought to understand if the facial‐recognition system DeepGestalt could find differences in phenotype between molecular subtypes of AS. Images and molecular data on 261 individuals with AS ranging from 10 months through 32 years were analyzed by DeepGestalt in a cross‐validation model with receiver operating characteristic (ROC) curves generated. The area under the curve (AUC) of the ROC for each molecular subtype was compared and ranked from least to greatest differentiable phenotype. We determined that DeepGestalt demonstrated a high degree of discrimination between the deletion subtype and UPD or imprinting defects, and a lower degree of discrimination with the UBE3A pathogenic variants subtype. Our findings suggest that DeepGestalt can recognize subclinical differences in phenotype based on etiology and may provide decision support for testing.  相似文献

Extracellular superoxide dismutase (EC-SOD) controls the availability of extracellular superoxide (O 2 - ), which is important for a variety of physiological pathways, including the primary means of inactivating nitric oxide (NO). The role of EC-SOD in neurobehavioral function has been until now unexplored. In the current studies, the phenotypic expression of genotypic alterations of EC-SOD production in mice were characterized for spatial learning and memory. Dramatic impairments in spatial learning in the win-shift 8-arm radial maze were seen in both EC-SOD knockout mice and EC-SOD overexpressing mice. The EC-SOD overexpressing mice were further characterized as having significant deficits in a repeated acquisition task in the radial-arm maze, which permitted the dissociation of long and short-term learning. Long-term learning was significantly impaired by EC-SOD overexpression, whereas short-term learning was not significantly affected by EC-SOD overexpression. NO systems have been shown to be importantly involved in learning and memory. This may be important in the current studies because EC-SOD has primary control over the inactivation of NO. We found that EC-SOD overexpressing mice were resistant to the cognitive effects of L-NAME (NG-nitro-L-arginine methyl ester hydrochloride), an NO synthase inhibitor. Decreased NO catabolism in these mice may have served to counter the effects of NOS inhibition by L-NAME. The current finding that EC-SOD levels that were either higher or lower than controls impaired learning demonstrates that the proper control of brain extracellular (O 2 - ) may be more vital than merely reduction of brain extracelluar (O 2 - ) in maintaining adequate learning function.  相似文献

Responses of lateral medullary neurons to microstimulation of two points in the locomotor strip—rostral and caudal to the obex—were recorded intracellularly in mesencephalic decerebellated uncurarized cats. Excitatory and inhibitory postsynaptic potentials and orthodromic action potentials occurred up to 20 ms after a single stimulus. A number of cells responded to stimulation of a locomotor point by a repetitive discharge, and in some cells synaptic responses were evoked by contralateral stimulation. The responsive neurons were scattered among other cells in the lateral medullary tegmentum. At least one-third of neurons with synaptic responses to stimulation of the rostral locomotor point were antidromically invaded from the caudal one. The characteristic length of these descending axons was between 4 and 9 mm, although there were longer axons too.The lateral medullary cells which give synaptic responses to stimulation of the locomotor strip form the locomotor column located medial to the strip, and a portion of these cells send their axons to the strip. It is suggested that the activity is propagated polysynaptically along the column through axonal collaterals of its neurons. One can assume that when repetitive stimulation achieves a threshold for locomotion such a propagation occurs without decrement. As a result, spinal stepping generators are activated.  相似文献

Effects of toxins III and IV (ATX III and IV) from the sea anemoneAnemonia sulcata on the Na current of crayfish giant axons were studied. Both toxins slowed the inactivation of Na channels, producing a maintained Na current during a depolarizing voltage pulse. Using the intensity of the toxin-induced maintained current as an index for the fraction of Na channels to which toxin is bound, the toxin association and dissociation kinetics were analyzed. The dissociation rate of ATX III was increased by two orders of magnitudes by depolarizing the membrane from –70 to –40mV. This increase of the dissociation rate caused a marked decrease in the binding rate of ATX III to Na channels in the same potential range. ATX IV exhibited association and dissociation kinetics that had a potential dependency quite similar to that of ATX III in spite of different ionic charge distribution in these two toxins. The results support the view that the potential-dependent kinetics of these toxins are not due to an electrostatic interaction between the ionic charges of toxins and the membrane potential but result from a modulation of the binding energy depending on the gate configuration of the Na channel.  相似文献
