人类精液中一氧化氮和尿酸含量检测的关系   总被引:1,自引:0,他引:1  
目的探讨人精液中一氧化氮(N0)与尿酸含量的关系,对精了质量的影响。方法参照WHO标准方法,进行精液常规分析,按精子密度、活动率不同分为(正常、<20、20~40、>40)4个组。采用镀铜镉还原荧光法检测NO代谢产物硝酸盐(NO3-)。采用尿酸酶一过氧化物酶偶联法检测精液尿酸含量。结果70例不育组精液中尿酸含量和NO含量为(236.4±47.8)μmol/L、(78.7±1.6)μmol/L与正常生育组(398.6±52.3)μmol/L、(41.84±1.6)μmol/L呈显著性差异(P<0.01)。将尿酸含量与NO含量进行相关性分析,两者呈显著性负相关(r=-0.96,P<0.05)。不育各精子密度和活动率组精液尿酸含量随精子密度及活动率增加而上升,N0含量随之下降(P<0.01),结论精液尿酸含量测定可作为评价精子受活性氧损害的重要指标,证明尿酸对活性氧尤其在医学领域极为重视的NO损害精子具有保护性作用。  相似文献

目的建立一种可行的精液密度测定的质量控制方法.方法采用血细胞计数板和计算机辅助精液分析(CASA)两种方法,对所选取的与精子头部直径相近的质控珠进行高、中、低三种密度测定,分别于日内及日间测试12次.结果采用血细胞计数板方法测试高、中、低三种密度结果的日内变异系数分别为12.09%、8.86%、4.72%,日间变异系数分别为16.71%、10.65%、10.22%.CASA法日内变异系数分别为6.65%、5.93%、3.80%,日间变异系数分别为7.90%、6.84%、6.14%.结论质控珠可用于以上两种测试精子密度方法的室内质量控制,从而达到保证实验室测试精子密度质量的目的.  相似文献

目的探讨前列腺液白细胞含量与精液主要参数和指标的关系。方法对入选的260例男性不育患者进行前列腺液(EPS)常规检查计数白细胞和解脲支原体培养,按照WHO人类精液实验室手册要求检测精液主要参数、精子形态分析、精子顶体酶活性、精浆抗体(AsAb)等,分析前列腺白细胞与男性不育相关因素的关系。结果260例中炎症组93例(EPS中WBC≥10个/HP):非炎症组(EPS中WBC〈10个/HP)167例。炎症组精液的精子密度、精子活动率、a+b级活力精子率、精子顶体酶阳性率均低于非炎症组(P〈0.05);而精液液化时间、精子畸形率、精浆抗体(AsAb)、前列腺液解脲支原体阳性率高于非炎症组(P〈0.05)。两组的精液量及pH值差异无显著性。结论前列腺液中白细胞含量对精液质量有一定的影响,慢性前列腺炎是导致男性不育的原因之一。  相似文献

PurposeThis study sought to compare sperm DNA fragmentation (SDF) in semen specimens after 3 days and then after 3 h of abstinence in men presenting for initial infertility evaluation.MethodsA prospective cohort study of 112 men undergoing their first semen analysis as part of an infertility work-up was conducted. All participants presented with 3 days of abstinence for a semen analysis and DNA-fragmentation test. Both tests were repeated on a second sample collected 3 h after the first ejaculation. DNA-fragmentation was evaluated with the halo test by one of two technicians blinded to duration of abstinence. Variables analyzed include ejaculate volume, sperm concentration and motility, smoking status, cannabis use, initial specimen DNA fragmentation, and use of sperm-directed anti-oxidant formulations.ResultsAmong all subjects, DNA fragmentation improved in the 3-h abstinence specimen (34.6 ± 19.4% vs. 23.7 ± 16.0%, p = 0.0001). Among subjects with high DNA fragmentation (> 35%) on the initial specimen, 55% improved into the normal range. Semen volume and sperm concentration decreased (3.1 ± 3.3 ml vs. 1.9 ± 0.8 ml, p < 0.01 and 41 ± 39 vs. 32 ± 31 (millions/ml), p = 0.01), while progressive motility tended to increase. Fifty-eight subjects demonstrated ≥ 30% improvement in SDF in the second specimen as compared to the first. Factors found to correlate with > 30% improvement in DNA fragmentation in the 3-h abstinence specimen compared to 3 days were younger age and use of anti-oxidants.ConclusionHigh SDF can often be managed with a second ejaculation 3 h after the first in infertile couples, including in males with abnormal semen analyses per the 2010 WHO guide. Apart from SDF levels, changes in sperm quality were not clinically significant in the second specimen and did not increase rates of ICSI. However, a second ejaculation after 3 h probably may reduce the necessity of costly and/or invasive ART strategies.  相似文献

A retrospective study was conducted in a large population to determine whether sperm quality has changed in Northeastern Spain between 1960 and 1996. From a total initial population of 22,759 men, two separate groups were studied: men with spermatozoa (n = 20,411) and those with azoospermia (n = 1364). After adjustment for age and sexual abstinence, multiple linear regression analyses were used to assess changes in semen parameters over time. A 0.2% decline was observed in semen volume in the spermatozoa group (P < 0.001). No significant increase (0.04%) in sperm count (x 10(6)/ml) was observed in the spermatozoa group. There was a 0.4% increase in motile spermatozoa in the spermatozoa group (P < 0.001). There was a statistically significant decline in normal spermatozoa (3.6%) in the spermatozoa group (P < 0.001). Of the total population, 1364 men had azoospermia (6.0%). The changes observed in the semen parameters analysed in this large population showed no evidence of a deteriorating sperm quality, although a statistically significant decline was observed in the percentage of normal spermatozoa.  相似文献

Seasonal changes in blood plasma biochemistry of siraz Capoeta capoeta umbla were determined throughout 1 year. In the present study, while the highest values for glucose and globulin were found in winter, the values for increased activities of total protein, albumin, calcium, sodium levels, albumin/globulin ratio, lactate dehydrogenase, and alkaline phosphatase were determined in summer. We determined that blood biochemistry values of siraz, which were positively affected by water temperature and pH, except glucose and globulin. Glucose and globulin were negatively affected by water temperature and pH. Consequently, it was concluded that the seasons are key factors when blood parameters are used as biomarkers for environmental alterations. It is difficult to define the most important factors affecting seasonal changes of blood biochemistry in fish.  相似文献

目的对中山地区男性不育患者的精液进行常规分析,了解中山地区男性不育患者精液质量的现状。方法按照《世界卫生组织人类精液检查与处理实验室手册》(第5版)的标准,应用西班牙人类精液分析微机辅助智能系统(CASA)对2012年10月~2013年6月来中山市博爱医院生殖中心就诊的2224例不育男性患者的精液进行常规检测和采用改良巴氏染色法分析精子正常形态率。结果2224例不育男性精液中正常者1113例(31.61%),异常者1521例(68.39%),其中液化异常964例(43.35%),pH值异常124例(5.58%),精子总数异常1002例(45.05%),精子浓度异常556例(25.0%),精子存活率异常1031例(46.36%),精子总活力(PR+NP)异常1208例(54.31%),无精子症372例(16.72%),精子正常形态率异常1021例(45.91%)。结论精子总活力、存活率下降,精子总数减少,精子正常形态率下降和精液液化不良是引起中山地区男性患者不育的主要原因,临床上应加于重视。  相似文献

We sought to determine the optimal assays for cytomegalovirus (CMV) shedding in semen. Over a 2-month period, 149 HIV-1-infected men who have sex with men each provided up to three semen specimens. Specimens were tested for CMV by culture, rapid assay (shell vial) and polymerase chain reaction (PCR). By culture, 30% of seminal plasma and 28% of seminal cell specimens grew CMV. By rapid assay, results were 38 and 33%, respectively. By PCR, 56% of seminal cell specimens demonstrated CMV: 20% in a single semen specimen; 33% in two specimens; and 34% in all three specimens. Overall, 69% of men had CMV detected by PCR in at least one seminal cell specimen. By quantitative PCR, 14% had ten, 14% had 100, 16% had 1000, and 12% had 10 000 copies in 6.25 μl of semen analyzed. Adjusting for initial CD4+ cell count, men with CMV shedding demonstrated by PCR at the first visit were approximately four times as likely to shed CMV at a subsequent visit (RR 4.28, CI: 2.30–7.95). CMV shedding was associated with decreased CD4+ cell counts in peripheral blood (P=0.05). It is concluded that the PCR assay provided the greatest sensitivity among the three detection methods.  相似文献

目的对血常规、血气及血生化改变在高原肺水肿(HAPE)早期诊断中的意义进行探讨。方法应用急性高原反应症状评分表,对24200例急进高原人员进行评分,筛选出高度疑似HAPE患者作为重点观察及随访对象,并给予卧床休息及吸氧。对所有重点观察及随访对象的血常规、血气及血生化(含电解质、肝功、肾功)变化情况进行检测分析。结果重点观察对象中,确诊HAPE组进入高原第1、2、3d的血白细胞总数(WBC)及中性粒细胞比率(N)均显著高于非HAPE组(P〈0.01);两组血气结果比较,HAPE组氧分压(PaO2)、二氧化碳分压(PaCO2)、动脉血氧饱和度(SaO2)及氧合指数(FiO2/PaO2)在进入高原第1、2d均显著低于非HAPE组(P〈0.01),HAPE组pH值显著高于非HAPE组(P〈0.01);两组血生化比较,HAPE组除钾(K^+)第2d显著低于非HAPE组外,各指标均无显著差异。结论早期白细胞计数增加及低氧血症持续性进行性加重与HAPE的发病有着密切的关系,加强疑似HAPE患者早期血常规及动脉血气的监测,有利于HAPE的早期诊断。  相似文献
