目的通过微流控检测平台,研究miRNA-874(miR-874)表达在胶质母细胞瘤干细胞(GSCs)生长中的作用及意义。 方法应用高通量、多样本、低成本的微流控技术,对GSCs和神经干细胞(NSCs)进行miR-874转染,并通过荧光定量PCR检测细胞内miR-874和mRNA表达等一系列细胞分子生物实验,研究miRNA-874对GSCs生长和凋亡的作用。 结果miR-874在GSCs中的表达显著降低,约为NSCs中表达量的2%~40%(P<0.05)。利用树状分子将miR-874转染入GSCs中后,GSCs的细胞数量显著下降,凋亡细胞的数量显著上升,提示miR-874对GSCs具有杀伤作用。 结论miR-874在GSCs中表达显著降低,在GSCs中过表达miR-874会导致显著的细胞生长抑制和死亡,有助于开发基于miRNA的新型脑癌药物和疗法。  相似文献

The present work describes synthesis, characterization, and use of a new dansyl-labelled Ag@SiO2 nanocomposite as an element of a new plasmonic platform to enhance the fluorescence intensity. Keeping in mind that typical surface plasmon resonance (SPR) characteristics of silver nanoparticles coincide well enough with the absorption of dansyl molecules, we used them to build the core of the nanocomposite. Moreover, we utilized 10 nm amino-functionalized silica shell as a separator between silver nanoparticles and the dansyl dye to prevent the dye-to-metal energy transfer. The dansyl group was incorporated into Ag@SiO2 core-shell nanostructures by the reaction of aminopropyltrimethoxysilane with dansyl chloride and we characterized the new dansyl-labelled Ag@SiO2 nanocomposite using transmission electron microscopy (TEM) and Fourier-transform infrared spectroscopy (FTIR). Additionally, water wettability measurements (WWM) were carried out to assess the hydrophobicity and hydrophilicity of the studied surface. We found that the nanocomposite deposited on a semitransparent silver mirror strongly increased the fluorescence intensity of dansyl dye (about 87-fold) compared with the control sample on the glass, proving that the system is a perfect candidate for a sensitive plasmonic platform.  相似文献

Bioresorbable vascular scaffolds (BRS) represent a novel approach to provide transient vessel support to drug-delivery capability without the long-term limitations of metallic drug-eluting stents (DES). The technology has the potential to overcome many of the safety concerns associated with metallic DES and possibly even convey further clinical benefit. In particular, the BRS are designed to provide short-term lumen support, and after being completely bioresorbed, eliminate the permanent caging typical of metallic DES. However, this technology has required new imaging modalities and methodologies for its assessment because the design, degradation rate, loss of mechanical property, and drug deliverability may affect its safety and efficacy. We provide an overview of all existing methods for assessing bioresorbable devices, from noninvasive to invasive, from light to sound based, and from morphological to functional parameters.  相似文献

目的设计一个可实现设备管理与临床行为互联互通的医院呼吸机管理平台,实现呼吸机的使用全过程管理。方法结合现有设备科软件平台合理分析本平台需求,依据需求分析建立呼吸机信息管理、上机管理、使用数据分析管理等九大管理模块,基于B/S架构、MVC模式、Microsoft SQL Server数据库等开发实现呼吸机管理平台。结果本平台实现了呼吸机使用全过程管理,可直观展示呼吸机使用情况,同时依据使用情况为呼吸机维护、报废提供数据支撑和提示。结论医院呼吸机管理平台的建立将实现呼吸机使用过程的精细化管理,提高呼吸机使用管理效率。  相似文献

目的 探索“互联网+”健康教育模式在乳腺外科的构建及应用效果。方法 选择2019年3月至2020年3月在河北大学附属医院乳腺外科手术治疗的160例乳腺癌(breast cancer,BC)患者作为研究对象,按照入院先后顺序分为对照组和观察组,每组80例。对照组采取传统健康宣教模式,观察组在对照组的传统健康教育模式基础上,重点运用“317护”微信平台开展健康宣教。比较干预后两组患者健康教育内容掌握情况、依从性以及患者满意度指标。结果 干预后,观察组乳腺癌患者健康教育内容掌握情况、依从性、满意度评分均明显高于对照组,差异均有统计学意义(P<0.05)。结论 “互联网+”健康教育模式在乳腺外科的应用效果较传统健康教育方式有显著优势, 值得临床推广应用。  相似文献

卫生学是非预防医学专业医学生的必修课程之一,南京医科大学卫生学课程采用E-learning网络平台与课堂授课相结合的教学模式。为了探讨E-learning平台在卫生学教学中的应用效果,应用电子问卷调查1436名本科生参与E-learning平台学习卫生学课程的情况,结果显示大部分同学认可E-learning平台对理论课程学习有促进作用,但学习的主动性有待加强。提高平台使用便捷程度,激发学生学习兴趣,有利于提升卫生学教学效果。  相似文献

目的评价微信课堂在妇科医师绝经相关疾病诊治培训中的运用效果。方法选取2017年3月自愿参与北京世纪坛医院线下继续医学教育项目"绝经相关疾病诊治培训"的47位妇科医师作为对照组(传统教学),自愿参与线上微信课堂培训的52位妇科医师作为实验组(微信课堂教学)。通过比较培训前后妇科医师对绝经相关疾病知识的掌握情况及课程满意度,以评价教学效果。采用SPSS 20.0对组间数据行t检验或卡方检验。结果所有妇科医师均完成了研究,培训前后的测评问卷回收率均为100%。培训前实验组学员绝经相关疾病知识平均得分(63.65±21.42)、知识考核通过率为65.4%,与对照组[(60.85±24.83)分,63.8%]相比,差异无统计学意义(P>0.05)。两组学员培训后的绝经相关疾病知识得分和考核通过率均高于培训前,且差异有统计学意义(P<0.05)。培训后,实验组学员平均得分为(77.50±16.19)分、考核通过率为90.1%,优于对照组[(78.72±16.89)分,87.2%],但两组差异无统计学意义(P>0.05)。实验组学员对培训的满意度相较对照组更高,但两组差异无统计学意义(P>0.05)。结论微信课堂用于妇科医师绝经相关疾病诊治培训,具有与传统教学相同的效果且培训满意度更高,可进一步运用于妇科其他亚专业培训及向其他专科推广。  相似文献

Despite more than two decades of research and development on nucleic acid vaccines, there is still no commercial product for human use. Taking advantage of the recent innovations in systemic delivery of short interfering RNA (siRNA) using lipid nanoparticles (LNPs), we developed a self-amplifying RNA vaccine. Here we show that nonviral delivery of a 9-kb self-amplifying RNA encapsulated within an LNP substantially increased immunogenicity compared with delivery of unformulated RNA. This unique vaccine technology was found to elicit broad, potent, and protective immune responses, that were comparable to a viral delivery technology, but without the inherent limitations of viral vectors. Given the many positive attributes of nucleic acid vaccines, our results suggest that a comprehensive evaluation of nonviral technologies to deliver self-amplifying RNA vaccines is warranted.  相似文献

Aim: To validate the “platform switching” concept at oral implants with respect to the preservation of the alveolar crestal bone levels in an animal model. Material & methods: Five minipigs received three implants each with a 0.25 mm implant/abutment mismatch and were placed flush (T0), 1 mm below (T1) and 1 mm above (T+1) the alveolar bony crest, and as a control, one conventionally restored implant placed at the bone level. The implants were randomly inserted flapless into the mandible. Four months after implant insertion, the animals were sacrificed, and undecalcified block sections were obtained and used for histological analyses. Results: The mean values for peri‐implant bone resorption were 1.09±0.59 mm (Control), 0.51 (±0.27 mm, T0), 0.50 (±0.46 mm, T+1) and 1.30 (±0.21 mm, T?1), respectively. Statistically significant differences (P<0.05) were found among the test (T0, T?1) and the control sites. Control implants presented an average biologic width length of 3.20 mm (±0.33), with a connective tissue adaptation compartment of 1.29 mm (±0.53) and an epithelial attachment of 1.91 mm (±0.71). T0, T+1 and T?1 implants presented with a mean biologic width of 1.97mm (± 1.20), 2.70mm (± 1.36) and 2.84mm (± 0.90), respectively, with a connective tissue adaptation compartment of 1.21mm (± 0.97), 1.21mm (± 0.65) and 1.50mm (± 0.70) and an epithelial attachment of 0.84mm (± 0.93), 1.66mm (±0.88) and 1.35mm (± 0.44), respectively. Differences between the configurations were mainly associated with the length of the epithelial attachment. The epithelial attachment was significantly longer in the C sites than in T0 (P=0.014). However, no other differences between configurations were detected. Conclusion: If the implants are positioned at the level of the alveolar bony crest, the platform‐switching concept may have a minor impact on the length of the epithelial attachment (0.84 vs. 1.91 mm), while the connective tissue adaptation compartment remains relatively unaffected. Moreover, platform switching resulted in less resorption of the alveolar crest (0.58 mm). To cite this article:
