目的确立、验证Sysmex XE-2100全自动血液分析仪(简称XE-2100)白细胞(WBC)高、低值的最佳复检范围。方法将WBC〈4.0×109/L及WBC〉18×109/L的663例非血液病住院患者的样本分为2组,即高值组和低值组。其中高值组又分为A组(18.00~22.00×109/L,共148份)、B组(22.01~30.00×109/L,共164份);C组(〉30.00×109/L,共74份)。低值组分为A'组(1.00~2.00×109/L,共90份);B'组(2.01~3.00×109/L,共92份);C'组(3.01~4.00×109/L,共95份)。对上述各组进行检测分析,以中华人民共和国卫生行业标准——白细胞分类计数参考方法为标准,通过受试者工作特征(ROC)curve曲线找出仪器检测WBC最佳复检范围,并对其进行验证。结果 A组ROC曲线下面积为0.820,当WBC为21.05×109/L时其灵敏度为1.0、特异度为0.536。B组曲线下面积为0.749,C组曲线下面积为0.329。A'组曲线下面积为0.689,B'组曲线下面积为0.825,当WBC为2.45×109/L时其灵敏度为1.0、特异度为0.317。C'组曲线下面积为0.983。验证结果:当WBC在2.45×109/L时无仪器警示标志的共28例,其中2例为假阴性,均为嗜碱粒细胞〉2,无重要临床意义;有警示标志的7例,其中1例为假阳性。当WBC为21.0×109/L时无仪器警示标志的共33例,其中只有1例假阴性也是嗜碱粒细胞〉2,有警示标志的7例其中3例为假阳性。结论 XE-2100的警示系统可以为复片提供有价值的信息,但血细胞计数范围的高低值也是不容忽略的复检因素。XE-2100在WBC〈2.50×109/L及WBC〉21.00×109/L时分类误差较大,即便无警示标志的出现依然需进行血细胞显微镜复检,以免造成漏诊。  相似文献

Penetrating eye injuries caused by bird pecking are uncommon with less than 40 cases reported in the literature. In this article, we present a case of penetrating ocular injury caused by a bird along with pertinent literature review. An otherwise healthy 56-year old man presented to the emergency department complaining of right eye severe pain and decreased vision following trauma caused by a bittern bird. The patient was diagnosed with penetrating ocular injury & he underwent primary repair. On follow up, he was found to have traumatic cataract, which was operated, however, the surgery was complicated with IOL dislocation into the vitreous cavity. The patient was referred to retina service where he underwent pars plana vitrectomy with IOL repositioning in the sulcus. After two months, the patient’s uncorrected visual acuity improved from counting fingers at 2 feet to 20/30?2. This case serves as a reminder that bird pecking is one of the causes of penetrating eye trauma.  相似文献

The thyroid receptor interactor protein 6 (TRIP6) has emerged as a key regulator for the proliferation and migration of various cells. However, whether TRIP6 is involved in regulating the proliferation and migration of airway smooth muscle (ASM) cells in the progression of pediatric asthma remains undetermined. The present study investigated the function of TRIP6 in regulating the proliferation and migration of fetal ASM cells induced by tumor necrosis factor (TNF)-α in vitro. The results revealed that TRIP6 expression was significantly upregulated in TNF-α-stimulated ASM cells. Loss-of-function experiments demonstrated that the knockdown of TRIP6 markedly suppressed TNF-α-proliferation and migration of ASM cells. By contrast, overexpression of TRIP6 had the opposite effect. In-depth research uncovered that TNF-α stimulation promoted the activation of yes-associated protein (YAP), which could be significantly reversed by TRIP6 silencing. Moreover, inactivation of YAP significantly reversed the promotion effect of TRIP6 overexpression on TNF-α-induced ASM cell proliferation and migration. Overall, these results reveal that upregulation of TRIP6 contributes to the proliferation and migration of fetal ASM cells by enhancing YAP activation, highlighting the importance of the TRIP6/YAP axis in the airway remodeling of pediatric asthma.  相似文献

BackgroundsTriptolide (TP) exhibits effective activity against colon cancer in multiple preclinical models, but the mechanisms underlying the observed effects are not fully understood. Sphingosine-1-phosphate (S1P) is a potent bioactive sphingolipid involved in the regulation of colon cancer progression. The aim of this study was to investigate the effect of TP on the sphingosine kinase (SPHK)-S1P signaling pathway in colitis-associated colon cancer.MethodsAn azoxymethane (AOM)/dextran sulfate sodium (DSS) mouse model and the THP-1 cell line were used to evaluate the therapeutic effects and mechanisms of TP in colitis-associated colon cancer (CACC). Various molecular cell biology experiments, including Western blotting, real-time PCR and immunofluorescence, were used to obtain relevant experimental data. A liquid chromatography–tandem mass spectrometry (LC-MS/MS) method was also established to detect the levels of S1P in tissue and plasma.ResultsIn the AOM/DSS mouse model, TP treatment induced a dose-dependent decrease in tumor incidence and inhibited macrophage recruitment and M2 polarization in the tumors. TP also efficiently decreased the S1P levels and SPHK1/S1PR1/S1PR2 expression and significantly inhibited activation of the S1P-mediated phosphorylation of ERK protein in macrophages.ConclusionsThe results indicated that TP might influence the recruitment and polarization of tumor-associated macrophages by suppressing the SPHK-S1P signaling pathway.  相似文献

Enumerating the number of items in a set accurately and quickly is a basic mathematical skill. This ability is especially crucial in the more real-life situations, where relevant items have to be discriminated from irrelevant distracters. Although much work has been done on the brain mechanisms and neural correlates of the enumeration and/or discrimination process, no agreement has been reached yet. We used event-related potentials (ERPs) to show the time course of brain activity elicited by a task that involved both enumeration and discrimination at the same time. We found that even though the two processes run to some extent in parallel, discrimination seems to take place mainly in an earlier time window (from 100 ms after the stimulus onset) than enumeration (beyond 200 ms after the stimulus onset). Moreover, electrophysiological evidence based on the N2 and P3 components make it reasonable to argue for the existence of a dichotomy between subitizing (for sets of less than four items) and counting (for sets of four and more items). Source estimation suggests that subitizing and counting, though being distinct brain processes, do recruit similar brain areas.  相似文献

目的通过调查国产乳酸环丙沙星滴眼液抑菌剂的使用情况,对本品抑菌剂使用的合理性进行分析。方法参考ChP2015四部通则1105,采用菌数计数法,通过方法适用性试验确定各时间各浓度点的存活菌数。参照ChP2015四部通则1121,评价各企业样品的抑菌效力是否满足控制要求。结合抑菌效力结果及各企业处方,讨论抑菌剂使用的合理浓度。结果 3个企业提供的处方中,2家企业使用苯扎溴铵,1家企业使用羟苯乙酯。抑菌剂含量均达标80.0%~120.0%的要求。使用苯扎溴铵为抑菌剂两企业抑菌效力均能达到A级,但存在添加过量的问题,使用羟苯乙酯作为抑菌剂的企业抑菌效力仅能达到B级,存在安全隐患。结论国产乳酸环丙沙星滴眼液中的抑菌剂存在处方不合理问题,有较大的优化空间,抑菌剂的使用应满足最低有效原则。  相似文献

目的 探索快速有效的内镜清洗操作流程,简化现有清洗步骤,缩短清洗时间.方法 将180条待清洗肠镜随机分为6组进行不同处理,分别为不同清洗流程、清洗流程中不同刷洗次数:即现有清洗流程、简化清洗流程刷洗1~3次,处理后采样做细菌学检测.结果 90条处理前内镜的平均菌落计数对数值为7.4909,处理后为0.4843,不同清洗流程之间的清洗效果相似、不同刷洗次数之间的效果相似,差异均无统计学意义;简化清洗流程较现有清洗流程节约了时间和一个清洗槽.结论 使用简化的清洗流程对内镜管腔刷洗1次即可达到规范要求,较现行的清洗流程更为适用;提高了工作效率和内镜使用率,为新的内镜清洗消毒规范的制定提供依据.  相似文献

目的 基于OSL原理,测量和分析在激发光下剂量片中沉积的剂量与激发出来的荧光光子数目的关系,为以后相关科研提供一个可靠的依据。方法 使用一个1.48 GBq(40 mCi)-90Sr/90Y β源对α-Al2O3:C剂量片进行不同时间的辐照,再用设计好的激发光装置将沉积在α-Al2O3:C剂量片的信号进行光激发,通过光电倍增管对荧光光子进行收集,最后通过单光子计数器输出计数。结果 通过上面的实验方法得到了剂量片上沉积剂量与荧光光子数的关系,通过线性拟合后可以得到线性关系。结论 从结果上可以看出剂量片上的本底剂量与其产生的荧光光子数存在线性关系,同时沉积剂量与荧光光子数也呈线性关系。  相似文献

BackgroundIntrahepatic cholangiocarcinoma (ICC) is a latent and malignant tumor with a dismal prognosis. This study was to evaluate the clinical relevance and therapeutic potential of SOX9-AS1 expression in ICC.MethodsThe cancerous tissues and adjacent normal tissues were collected from ICC patients. Blood samples from ICC, hepatocellular carcinoma (HCC) group, the extrahepatic cholangiocarcinoma (ECC) group and the healthy controls were collected. SOX9-AS1 levels were evaluated in tissues (versus normal tissues) and plasma samples (versus plasma from HCC and ECC by quantitative real-time RT-PCR. The diagnostic value of SOX9-AS1 for ICC was estimated using receiver operating characteristic (ROC) curves. The relevancy between SOX9-AS1 expression and overall survival or recurrence-free survival was assessed by Kaplan-Meier curves multivariate analyses. The overexpression and knockdown of SOX9-AS1 on cell behavior were assessed by CCK-8 and transwell assay.ResultsSOX9-AS1 levels were increased in ICC, both in the tissues and the cell lines. The upregulation of SOX9-AS1 showed a highly discriminative profile, distinguishing ICC patients from healthy subjects or HCC or ECC patients. Upregulation of SOX9-AS1 was related to shorter overall survival and recurrence-free survival. Muli-variate analysis revealed that SOX9-AS1 expression was an independent prognostic purpose factor of worst overall survival and recurrence-free survival.ConclusionsSOX9-AS1 drives tumor growth and metastasis in ICC. SOX9-AS1 may be applied as a new diagnostic and prognostic purposed marker, in addition to a promising therapeutic target in ICC.  相似文献
