The prevalence of human papillomavirus (HPV) in paired cervical scrape and urine specimens from 144 women attending a clinic for genitourinary medicine was determined by polymerase chain reaction (PCR) and nested PCR, using degenerate and general primer pairs localized within the L1 region. HPV typing was by restriction fragment length polymorphism (RFLP), type-specific PCR (HPV 6, 11, 16, 18, 33), and partial DNA sequencing of PCR products. HPV DNA was detected in 114 (84%) women. HPV DNA was detected in the specimens of 58 patients after amplification with MY09/MY11 primers and in a further 54 patients after nested PCR with the GP5 + /GP6 + primers. A total of 106/136 (78%) of women had HPV DNA positive cervical scrapes and 89 (65%) had HPV DNA positive urine specimens. Both the urine and cervical specimens of 81 women were positive. In 25 women HPV DNA was detected in the cervical specimen only, and in 8 women HPV DNA was detected in the urine specimens only. A total of 108 specimens from 75 patients were typed. For 33 patients HPV typing was achieved in both the cervical and the urine specimens and 19 women had identical types in paired specimens. Multiple HPV infections could be detected in 15 (20%) of 75 women where either the cervical and urine specimen or both of the specimens could be typed. More then one HPV type was found in 8 specimens and from multiple sites (cervix and urinary tract) in the same patients on 7 occasions. The results of this study indicate that the detection of HPVs in the urogenital tract can be maximised through the testing of both cervical scrapes and urine specimens in conjunction with the use of a nested PCR to increase the sensitivity of HPV DNA detection. DNA was extracted from an odontogenic keratocyst and assayed for the presence of human papilloma virus (HPV) type 16 DNA sequences using high stringency Southern blot hybridization. HPV type 16 homologous DNA sequences were detected at a copy number of 50-100 genome copies per diploid cell. The oral HPV DNA was not identical to the prototype HPV 16 when cleaved with the restriction enzyme Pst-I, since it appeared to lack the Pst-I C fragment (L2/L1 ORFs) and contained "off-sized" high molecular weight fragments suggestive of integration events into the host cell chromosome. 相似文献

目的 :探讨人乳头瘤病毒 (humanpapillomavirus ,HPV)与宫颈癌前病变 (cervi calintraepithelialneoplasia ,CIN)和宫颈癌的关系以及与它们的预后关系。方法 :采用第 2代杂交捕获试验法检测我院门诊和住院患者 994例随访手术治疗患者 99例 ,随访患者同时行宫颈细胞学、阴道镜检查 ,以病理结果为金标准 ,按宫颈病变严重程度比较高危型HPV的检出率 ,以及比较术前术后宫颈癌前病变、宫颈癌感染高危型HPV变化情况。结果 :以HPVDNA≥ 1 0pg/mL为阳性标准 ,慢性宫颈炎、CIN各组及宫颈癌分别为 46 92 % ( 2 0 6/4 3 9)、65 71% ( 2 3 /3 5 )、81 82 % ( 3 6/4 4)、 98 41%( 62 /63 )、82 3 2 % ( 3 40 /4 13 )。随访患者细胞学 ,阴道镜检查除 1例CIN3患者为阳性 ,其余均为阴性 ,术后HPV持续阳性率为 2 1 18% ,其中 1例诊断为宫颈原位癌。结论 :高危型HPV感染与宫颈癌前病变和宫颈癌有明显相关性 ;宫颈锥切术后HPV持续阳性患者为复发的高危人群 ,应严密随访 相似文献

目的:研究人乳头瘤病毒(HPV)16型和18型感染与宫颈癌的关系。方法:采用荧光探针PCR技术对62例患的宫颈管分泌物中HPV病原体分别进行检测,其中宫颈癌22例(包含CINⅢ级10例);慢性宫颈炎20例;正常宫颈20例。结果:上述宫颈分泌物中的HPVl6、HPVl8总阳性率分别是59.1%、20.0%和0%,三差异具有显性,且CINⅢ级与宫颈癌I~Ⅲ期之间差异显。结论:HPVl6、HPVl8感染是宫颈癌致病的重要因素。HPVl6、HPVl8荧光探针PCR技术可以作为检测HPVl6、HPVl8感染及宫颈癌早期诊断标记之一。 相似文献

OBJECTIVE: Our purpose was to recognize special features of women with both vestibulitis and constant vulvar pain (vestibulodynia) and to determine whether vestibulodynia is a separate disease entity or a subset of vestibulitis. STUDY DESIGN: Ninety-one women with severe vulvar vestibulitis underwent perineoplasty by a single surgeon during 1992 to 1995. Twenty-five (27%) of them who had vestibulodynia were compared with 66 (73%) who had dyspareunia alone. With univariate and then multivariate analysis, the two groups were compared with regard to demographic, social, and medical variables, the presence of human papillomavirus deoxyribonucleic acid, physical findings in the vestibule, and surgical outcome. RESULTS: The vestibulodynia and vestibulitis groups differed in age (30.5 ± 10.6 years vs 24.7 ± 6.2 years respectively), as well as in marital status (married, 44% vs 18%, respectively), nonuse of contraception (20% vs 1%, respectively), presence of human papillomavirus deoxyribonucleic acid in the vestibule (80% vs 48%, respectively), and failure of surgical treatment (incomplete response rate, 64% vs 2%, respectively). With multivariate analysis, patients with human papillomavirus deoxyribonucleic acid present in the vestibule have an odds ratio of 5.44 (95% confidence interval, 1.45 to 20.33) of belonging to the group with vestibulodynia, whereas dysuria and age have odds ratios of 3.70 (95% confidence interval 1.09 to 12.55) and 1.14 (95% confidence interval 1.04 to 1.24), respectively. Human papillomavirus (HPV) DNA sequences were detected by Southern blot hybridization and polymerase chain reaction (PCR)
in 10 out of 19 patients (52.7%) with adenocarcinoma [15] and adenosquamous [4] carcinoma of the uterine cervix. HPV 18 DNA
was detected in 8 of these 19 patients (42.1%), HPV 16 DNA in 1 patient (5.3%) and HPV type X (unknown) in another (5.3%).
Of the 10 HPV positive samples HPV 18 was found in 6 out of 6 pure adenocarcinomas (100%), and in 2 of 4 (50%) adenosquamous
carcinomas. HPV 16 and HPV X were each detected in 1 out of 4 (25%) adenosquamous carcinomas. The physical state of the viral
DNA was investigated in 5 of the 10 HPV-positive cases. All the specimens from these 5 cases showed HPV to be integrated into
the host genome, except for one adenosquamous specimen, which showed both episomal and integrated forms of HPV 16. Six of
8 HPV 18 DNA positive specimens were from cases of pure adenocarcinoma and it was found by PCR that five of these 6 specimens

应用聚合酶链反应(PCR)技术制备非放射性探针标记物——地高辛素标记人类乳头瘤病毒(HPV)共有引物探针,对146例喉不同病变的新鲜组织标本(喉癌68例,喉其它病变48例,正常喉组织30例)进行HPV6,11,16,18,31,33,35,42,58共9型HPVSDNAS感染的检测。结果表明,喉癌组HPV感染阳性率45.6%(31/68),喉癌颈转移淋巴结组阳性率21.0%(3/15),喉癌前病变阳性率11.8%(2/17),声带息肉组阳性率6.3%(1/16),15例癌旁及15例癌周正常喉组织均为HPVDNA阴性。文中还对HPV在喉癌中的致病作用及应用PCR技术制备地高李标记HPV共有引物探针的敏感性及特异性进行了讨论。 相似文献
