Parotid glands of 5-day-old rats were maintained in DM-153 synthetic, serum-free medium in organ culture for 24 to 48 hr. Differentiation of acinar cells in vitro, as revealed by an increase in amylase activity in the gland and in the culture fluid, as well as by the immunocytochemical demonstration of amylase in the acinar cells, was accelerated by insulin, prednisolone, and l-thyroxine added to the culture medium. The potent tumor promoter 12-O-tetradecanoylphorbol-13-acetate (10?7M) inhibited both the unstimulated and hormone-stimulated differentiation of the gland without causing cellular degeneration or necrosis. 相似文献

Transformation of primary rat kidney cells with specific genome fragments of oncogenic human adenovirus type 12 provides a suitable model for the study of separate phenotypic manifestations of (tumorigenic) transformation. In this model, oncogenic transformation by specific virus fragments was accompanied by the appearance of a new, high-molecular-weight class of glycoprotein-bound carbohydrates similar as observed in a variety of spontaneous and experimental tumors. These tumor-associated carbohydrate changes were absent on the surface of nontumorigenic cells, morphologically transformed by other Ad 12 genome fragments or on untransformed control cells. It is concluded that tumor-associated changes in membrane carbohydrates are expressed simultaneously with a 60kD T antigen required for oncogenicity. In Ascaris muscle mitochondria the major respiratory chain-linked phosphorylation activity is accomplished by a NADH-linked reduction of fumarate to succinate. Oxygen can also be employed as a terminal electron acceptor via a cyanide- and salicyl-hydroxamate-resistant terminal oxidase. As in fumarate-dependent electron transport this process appears to be coupled to energy conservation at phosphorylation site I. The branchpoint from which electrons are taken from the main respiratory chain to either the alternative oxidase or fumarate reductase is likely to be on the oxygen side of the NADH dehydrogenase segment.Malate and succinate are the only substrates which appreciably support respiration in the mitochondrion of the nematode. Regardless of the presence or absence of oxygen malate is utilized by an oxidation-reduction reaction resulting in the formation of pyruvate, acetate, succinate, propionate and CO2. In addition, aerobically, hydrogen peroxide is formed as the product of oxygen reduction. Succinate accumulation was found to be significantly higher in the anaerobic as compared to the aerobic incubation mixtures. This effect was accompanied by an increase in anaerobic malate consumption. ATP generation and the formation of pyruvate, acetate and propionate were found to be similar in the presence and absence of oxygen.In malate-supported respiration of intact Ascaris mitochondria reducing equivalents (NADH) are produced exclusively through pyruvate and acetate formation. These enzymatic reactions are functionally coupled to the electron transport-linked reductions of fumarate to succinate and oxygen to hydrogen peroxide, respectively. In accordance with the position of the redox potentials of the fumarate/succinate and O2/H2O2 couples, anaerobic and aerobic respiration was found to be associated with relatively low energy conservation efficiencies. Thus one molecule of ATP was conserved per 2e? transferred to fumarate or oxygen, respectively. No evidence could be obtained for a significant activity of energy conservation sites II and III and electron transfer through the alternative oxidase pathway was shown not to be coupled to phosphorylation. 相似文献

An analysis of core particles of 1.23 g/cm3 density of the endogenous feline type C virus (RD-114) showed the presence of all of the major low molecular weight structural proteins, namely p30, p12, p10, and the phosphoprotein pp15, in relative proportions similar to those detected in intact RD-114 virions. The protein kinase activity of the virus was also found to be associated with the core. A comparison of the content of multiple phosphorylated species of the pp15 derived from the core with that isolated from the whole virion demonstrated that all major phosphorylated species of RD-114 pp15 were located in the core structure. 相似文献

Heart rate (HR) and skin conductance (SC) were recorded while 17 psychopathic (P) and 17 nonpsychopathic (NP) inmates (referred to as A) were engaged in a mixed-motive game situation with another S (referred to as B). On each trial A had to choose the intensity of shock to be delivered to himself and to B. B then was given a chance to retaliate, although his choices were actually overridden by the experimenter. A 10 sec tone (CS) preceded delivery of shock to each S. There were no differences between Groups P and NP in the intensity of shock chosen for themselves and for the other (B) Ss. Compared with Group NP, Group P gave small unconditioned skin conductance (SC) responses to shock directly received and to shocks delivered to the other S. There were no differences between groups in the unconditioned HR response to either direct shock (acceleration) or to shocks delivered to the other S (slight deceleration). Group P gave small electrodermal orienting responses (ORs) and anticipatory responses (ARs) to the CS preceding shock to self and shock to other; Group NP gave relatively large ORs and ARs to the CS preceding shock to self, and small ones prior to shock to other. Both Groups gave a biphasic conditioned HR response–acceleration followed by deceleration; each component was larger in Group P than in Group NP, and the acceleratory component in Group P appeared on the first trial. The electrodermal data were consistent with the view that psychopaths experience little fear arousal prior to reception of aversive stimuli by themselves or by others. It was suggested that the anticipatory HR responses of the psychopathic Ss were part of an adaptive response that helped them to cope with stress. 相似文献

Summary The 275 Purkyn cells identified by the criteria of the previous paper have been investigated with respect to their role as units integrating the input to the anterior lobe from various limb nerves. The discharges from single Purkyn cells have been studied in lightly anesthetized (pentothal) or in decerebrate unanesthetized cats, there being averaging usually of 128 responses in the form of post-stimulus time histograms and cumulative frequency distributions.Single Purkyn cells exhibited a wide variation in their responses to the diverse inputs from the various afferent nerves. Attention was focussed on excitatory and inhibitory responses evoked by mossy fibers with a short latency, usually 10–15 msec for hindlimb afferents. With most Purkyn cells these responses were predominantly evoked from cutaneous nerves, low threshold fibers being particularly effective. A few Purkyn cells were preponderantly excited by afferent volleys from muscle nerves, but there was a large group with a mixed input from cutaneous and muscle nerves. Graded strengths of stimulation of muscle nerves showed that sometimes group I volleys were prepotent, but other Purkyn cells were selectively excited by group II volleys. Though sometimes the afferent volleys from antagonistic muscles had a reciprocal action on a Purkyn cell, as on a motoneurone, it was more common to find similar actions. Also convergence of inputs from forelimb and hindlirnb nerves, both cutaneous and muscular, was not uncommon, particularly in marginal areas between hindlimb and forelimb zones. A special design feature is the convergence onto a Purkyn cell of mossy fiber and climbing fiber inputs evoked by the same afferent volley. This convergence was of particular interest along the parasagittal strip of hindlimb climbing fiber distribution in lobule V.It was not possible to translate the observations into some map of the cerebellar cortex on which are marked the territorial distributions from the various limb afferent nerves. Rather, there was an ill-defined patchy character, closely adjacent Purkyn cells often receiving very different subsets of the total input from the various limb nerves. For highly diffusive solutes the kinetics of blood–tissue exchange is only poorly represented by a model consisting of sets of independent parallel capillary–tissue units. We constructed a more realistic multicapillary network model conforming statistically to morphometric data. Flows through the tortuous paths in the network were calculated based on constant resistance per unit length throughout the network and the resulting advective intracapillary velocity field was used as a framework for describing the extravascular diffusion of a substance for which there is no barrier or permeability limitation. Simulated impulse responses from the system, analogous to tracer water outflow dilution curves, showed flow-limited behavior over a range of flows from about 2 to 5 ml min–1 g–1, as is observed for water in the heart in vivo. The present model serves as a reference standard against which to evaluate computationally simpler, less physically realistic models. The simulated outflow curves from the network model, like experimental water curves, were matched to outflow curves from the commonly used axially distributed models only by setting the capillary wall permeability–surface area (PS) to a value so artifactually low that it is incompatible with the experimental observations that transport is flow limited. However, simple axially distributed models with appropriately high PSs will fit water outflow dilution curves if axial diffusion coefficients are set at high enough values to account for enhanced dispersion due to the complex geometry of the capillary network. Without incorporating this enhanced dispersion, when applied to experimental curves over a range of flows, the simpler models give a false inference that there is recruitment of capillary surface area with increasing flow. Thus distributed models must account for diffusional as well as permeation processes to provide physiologically appropriate parameter estimates. © 2000 Biomedical Engineering Society.
