Abstract
Cilostazol is a selective inhibitor of phosphodiesterase III with anti-platelet-aggregatory and vasodilating properties. Randomised, double-blind, placebo-controlled trials in 2702 patients with intermittent claudication demonstrated that cilostazol significantly increased walking distances compared with placebo. Furthermore, the agent has beneficial effects on the serum lipid profile and fatty acid composition in plasma. Consequently, cilostazol may be useful to prevent atherosclerosis from progressing by ameliorating lipid and fatty acid metabolism. 相似文献

Metabolic pathways leading to lipid biosynthesis in four different developmental stages of Schistosoma mansoni were explored and quantified by incubation in the presence of labeled precursors in a chemically defined medium. At the schistosomulum stage and in male, female, or paired worms, glycerol and oleate incorporation into neutral lipids, mainly in the form of triacylglycerols, was greater than into phospholipids, whereas in 11-and 15-day-old worms, synthesis mainly led to phospholipids. Incorporation into phospholipids was recovered largely in phosphatidylcholine, and distribution into other phospholipids depended on the developmental stage. Incorporation of choline and ethanolamine into their respective phospholipids represented up to 15% of the parasitic phospholipid content. The formation of phosphatidylcholine by phosphatidylethanolamine methylation occurred mainly in the immature parasitic stages. Inositol incorporation was also measurable, whereas [14C]serine incorporation was low or undetectable. Addition of 1-palmitoyl-2-[14C]oleyl phosphatidylcholine revealed a very high uptake of this phospholipid by the immature stages but further metabolism was not detectable. In contrast, adult S. mansoni were completely unable to take up or absorb this exogenous phospholipid. The most striking aspect of this study was the relatively high metabolic activity in 11-day-old worms and the lower but sustained activity on day 15 and at the schistosomulum stage. By comparison, biosynthetic activity in adult S. mansoni, on which research studies have been focused until now, was very low. We present a mathematical model for pre-fusion interaction between an influenza virus and a healthy cell. Our model describes the role played by hemagglutinin (HA) protein clusters in bringing the viral membrane into close contact with the host cell membrane as a first step of the fusion process between the two membranes. The viral membrane is modelled as a lipid bilayer with bending rigidity. Using the calculus of variations, we compute the deformation of the viral membrane under the influence of HA protein clusters. Our numerical results support the hypothesis of dimple formation in the fusion site proposed in the literature. The asymmetric nature of the protein molecules due to various reasons such as tilting is the primary cause for the dimple formation. We discuss the effects of spontaneous curvature, the protein cluster radius, fusion-site size and the bending moment exerted by the protein cluster. We also examine the effects of membrane tension and the presence of a host cell on the dimple shape. Our results support previous experimental observations. 相似文献

Restoration of bile flow after 9-day cholestasis in rat liver normalized the content of lipid peroxidation products. The removal
of the cholestatic factor after 12-day cholestasis was not followed by recovery of these parameters. We showed that measurement
of serum concentration of lipid peroxidation products in patients with cholelithiasis during the preoperative period holds
promise for selection of the optimum time for surgical treatment and prediction of the risk of postoperative complications.

BACKGROUND: Allergy to plant-derived foods is associated with birch pollinosis in central and northern Europe. Symptoms elicited are usually limited to the oropharyngeal system. By contrast, in the Mediterranean area, allergy to the same foods manifests more frequently with systemic reactions caused by nonspecific lipid transfer proteins (nsLTP), independently of an associated pollinosis. OBJECTIVE: We sought to investigate the pattern of immunoglobulin E (IgE) binding protein bands implicated in lettuce allergy, in particular the presence of an nsLTP. METHODS: Consecutive lettuce allergic patients were selected. Determination of serum-specific IgE, immunoblot, and inhibition experiments were performed in order to study the pattern of IgE binding proteins and the potential cross-reactivity to pollens. Inhibition studies with recombinant allergens were conducted to identify the lettuce allergens. The major allergen was subjected to N-terminal amino acid sequencing. RESULTS: Fourteen patients were diagnosed as being allergic to lettuce. All were sensitized to Platanus pollen. Ten of them showed specific IgE to a lettuce protein of 9-kDa. The IgE binding to this protein was completely inhibited by the cherry-LTP and peach extract. The N-terminal sequence of the 9-kDa protein showed a high degree of amino acid sequence identity to other nsLTPs. A clear partial cross-reactivity was observed between lettuce-LTP and Platanus-pollen extract. CONCLUSIONS: An LTP has been demonstrated to be a major allergen in patients suffering from lettuce allergy. 相似文献

目的:探讨核转录因子NF-κB在单磷酰脂A预处理的延迟阶段的作用。 方法: 建立大鼠心肌缺血/再灌注(I/R)损伤模型。分别应用单磷酰脂A(MLA)、NF-κB的特异性抑制剂DDTC加MLA预处理心脏, 24 h对心脏进行较长时间缺血再灌注,检测心肌梗死范围、LDH释放及心肌细胞凋亡率。另外分组检测MLA预处理0 min、15 min、30 min、60 min、NF-κB抑制剂DDTC加MLA预处理后30 min时NF-κB活性。 结果: MLA预处理减小再灌注心肌梗死范围、降低血浆LDH活性、减少细胞凋亡(P<0.05 vs I/R)。同MLA预处理组相比, DDTC+MLA组心肌梗死范围增大、LDH活性增高, 细胞凋亡增加(P<0.05)。与预处理前相比,NF-κB活性在预处理后15 min明显升高、30 min达到高峰、60 min下降(P<0.01)。与MLA预处理后15 min、30 min、60 min相比, DDTC加MLA预处理后30 min时,NF-κB活性明显降低(P<0.01),而与MLA预处理0 min无显著差别(P>0.05)。 结论: (1) MLA预处理对24 h后缺血/再灌注心肌有保护作用。(2) 核转录因子NF-κB参与心肌预处理后的延迟保护作用,MLA预处理后NF-κB快速的核转移并激活可能是药物预处理延迟保护作用的重要机制之一。 相似文献

目的:探讨小陷胸汤加味中药方对血管内皮细胞的保护作用。方港:建立ox-LDL损伤人脐静脉内皮细胞株(ECV-304)模型,用小陷胸汤加味含药血清处理模型,并用放免和硝酸酶还原法在药物干预6h和24h后检测细胞上清液中ET-1和NO含量。结果:100 μg/ml的ox-LDL可损伤血管内皮细胞并导致其分泌NO和ET-1功能失调,小陷胸汤加味含药血清通过影响NO/ET-1的分泌而明显改善此失调状态。结论:小陷胸汤加味中药通过调节NO/ET-1水平显著拮抗ox-LDL对血管内皮细胞损害,具有防治AS的作用。 相似文献
