目的通过观察和血生络方对心肌梗死大鼠血清中中性粒细胞集落刺激因子(G-CSF)、碱性成纤维细胞生长因子(bFGF)等的影响。方法以雄性Wistar大鼠制作心肌梗死大鼠模型,随机分为硝酸异山梨酯(消心痛)组、和血生络高、低剂量组及模型组,同时建立假手术组,每组8只。药物治疗组分别给予和血生络水溶液(9.9,19.8 g.kg-1.d-1)及消心痛水溶液(2.7 mg.kg-1.d-1)每天灌胃1次,其他组给予等量蒸馏水,连续用药3 w后处死大鼠,取血清检测G-CSF与bFGF的含量。结果和血生络高剂量组、消心痛组血清中G-CSF与bFGF含量较模型组显著增高(P<0.05),和血生络高剂量组血清中G-CSF较消心痛组显著增高(P<0.05)。结论和血生络法的作用机制可能与两个方面有关,一方面促进内皮细胞增殖和迁移以利于血管的新生,另一方面动员骨髓中的内皮祖细胞迁移到梗死局部并分化成为内皮细胞促进新血管形成。 相似文献

IntroductionPostoperative dysphonia is mostly caused by vocal fold scarring, and careful management of vocal fold surgery has been reported to reduce the risk of scar formation. However, depending on the vocal fold injury, treatment of postoperative dysphonia can be challenging.ObjectiveThe goal of the current study was to develop a novel prophylactic regenerative approach for the treatment of injured vocal folds after surgery, using biodegradable gelatin hydrogel microspheres as a drug delivery system for basic fibroblast growth factor.MethodsVideoendoscopic laryngeal surgery was performed to create vocal fold injury in 14 rabbits. Immediately following this procedure, biodegradable gelatin hydrogel microspheres with basic fibroblast growth factor were injected in the vocal fold. Two weeks after injection, larynges were excised for evaluation of vocal fold histology and mucosal movement.ResultsThe presence of poor vibratory function was confirmed in the injured vocal folds. Histology and digital image analysis demonstrated that the injured vocal folds injected with gelatin hydrogel microspheres with basic fibroblast growth factor showed less scar formation, compared to the injured vocal folds injected with gelatin hydrogel microspheres only, or those without any injection.ConclusionA prophylactic injection of basic fibroblast growth factor -containing biodegradable gelatin hydrogel microspheres demonstrates a regenerative potential for injured vocal folds in a rabbit model. 相似文献

目的 探讨垂体瘤转化基因(PTTG)和碱性成纤维细胞生长因子(bFGF)在人脑星形细胞肿瘤中的表达及其意义.方法 免疫组化SP法检测55例人脑星形细胞肿瘤中PTTG、bFGF的表达.结果 各级别肿瘤中均不同程度发现PTTG、bFGF阳性表达,二者表达强度有随肿瘤恶性程度增高而增强的趋势(rS分别为0.8757、0.7557,P<0.01),PTTG、bFGF的表达呈正相关(rS=0.7520,P<0.01).结论 PTTG-bFGF通路存在于人脑星形细胞肿瘤中,与肿瘤恶性程度密切相关,二者在胶质瘤的发生及恶性转化中具有重要作用. 相似文献

Wound healing involves the interactions of many cell types, and is controlled in part by growth factors. Intercellular communication
mediated by gap junctions is considered to play an important role in the coordination of cellular metabolism during the growth
and development of tissues and organs. Basic fibroblast growth factor (bFGF), known to be important in wound healing, has
been found to increase Cx43 expression and intercellular communication in endothelial cells and cardiac fibroblasts. It has
been proposed that an increased coupling is necessary for the coordination of these cells in wound healing and angiogenesis,
and that one of the actions of bFGF is to modulate intercellular communication. The aim of our study was to evaluate the effects
of bFGF on gap junctional intercellular communication (GJIC) in vitro, and the presence of gap junctional proteins connexin
(Cx) 26, Cx32, and Cx43 in fibroblasts of diabetic and nondiabetic individuals. Fibroblast cell lines (n=10) were cultured for 3 d in serum-free media with or without bFGF (3 ng/mL). Cells were evaluated for the rate of GJIC by
using laser cytometry, and for the presence of Cx26, Cx32, and Cx43 by immunohistochemical and Western analyses. All cell
types communicated via contact-dependent mechanisms. The rate of GJIC was greater (p<0.01) for diabetic than for nondiabetic fibroblasts (4.1±0.01 vs 3.3±0.01 %/min). bFGF increased (p<0.01) the rate of GJIC for diabetic (4.9±0.01 vs 4.1±0.01%) and nondiabetic (4.1±0.01 vs 3.3±0.01%) fibroblasts. Immunohistochemistry
identified Cx26 in the cytoplasm, Cx32 was not detected, and Cx43 was present on the cellular borders in all cultures. Image
analysis of immunofluorescent staining demonstrated that bFGF increased (p<0.05) Cx43 expression in diabetic and nondiabetic fibroblasts. Western immunoblot analysis revealed bands at 43–46 kD that
were similar in volume for diabetic and nondiabetic fibroblasts. Thus, gap junctions involving Cx43 and GJIC among fibroblasts
appear to be targets for bFGF. Fibroblasts of diabetic individuals appear to have an increased rate of cell-cell coupling,

Background
Several methods have been developed to detect allergen-specific IgE in sera. The passive IgE sensitization assay using human IgE receptor-expressing rat cell line RBL-2H3 is a powerful tool to detect biologically active allergen-specific IgE in serum samples. However, one disadvantage is that RBL-2H3 cells are vulnerable to high concentrations of human sera. Only a few human cultured cell lines are easily applicable to the passive IgE sensitization assay. However, the use of human induced pluripotent stem cells (iPSCs) to generate human mast cells (MCs) has not yet been reported.Methods
The nuclear factor-kappa B (NF-κB)-responsive luciferase reporter gene was stably introduced into a human iPSC line 201B7, and the transfectants were induced to differentiate into MCs (iPSC-MCs). The iPSC-MCs were sensitized overnight with sera from subjects who were allergic to cedar pollen, ragweed pollen, mites, or house dust, and then stimulated with an extract of corresponding allergens. Activation of iPSC-MCs was evaluated by β-hexosaminidase release, histamine release, or luciferase intensity.Results
iPSCs-MCs stably expressed high-affinity IgE receptor and functionally responded to various allergens when sensitized with human sera from relevant allergic subjects. This passive IgE sensitization system, which we termed the induced mast cell activation test (iMAT), worked well even with undiluted human sera.Conclusions

目的:探讨前列散瘀胶囊对实验大鼠前列腺增生的影响。方法:将48只雄性大鼠随机分为正常对照组、损伤模型组、癃闭舒组和前列散瘀胶囊低、中、高3个剂量组。除正常对照组外,其余各组采用丙酸睾丸酮皮下注射引起大鼠前列腺增生模型。连续给药30天,称取动物体重,采用心脏采血,将动物处死,摘除前列腺各叶,称取前列腺湿重。计算大鼠前列腺指数,每例标本进行常规HE染色,检测大鼠血清bFGF含量。结果:大鼠体重、前列腺重量及前列腺指数比较:损伤模型组大鼠体重较正常对照组明显轻(P<0.05),而前列腺重量明显增加(P<0.01),前列腺指数明显升高(P<0.01);前列散瘀胶囊高剂量组与损伤模型组相比,前列腺重量明显减轻(P<0.01),前列腺指数明显下降(P<0.01)。bFGF浓度比较:损伤模型组大鼠血清bFGF含量与正常对照组相比,差异具有统计学意义(P<0.01);与损伤模型组相比,前列散瘀胶囊高、中剂量组和癃闭舒组大鼠血清bFGF浓度降低,差异具有统计学意义(P<0.01)。结论:前列散瘀胶囊能降低实验大鼠前列腺湿重、前列腺指数及血清bFGF浓度。 相似文献

目的 探讨20(s)-原人参二醇(Ppd)对肝癌血管内皮生长因子(VEGF)及碱性成纤维细胞生长因子(bFGF)表达的影响.方法 建立肝癌动物模型,将实验动物分为五组,每组10只,分对照组、环磷酰胺组(CTX)、20(s)-原人参二醇25mg/kg、50mg/kg、100mg/kg给药组,给药二周后处死动物.称肿瘤重量及肿瘤体积,制成组织切片,以备免疫组化应用.结果 与对照组相比,给药组VEGF、bFGF表达受到抑制,肿瘤的重量及体积明显减轻,组间有显著性差异(P<0.01).结论 提示Ppd抑制了VEGF、bFGF蛋白的表达,抑制了肿瘤生长. 相似文献

BACKGROUND & AIMS: Angiogenesis is a critical component of neoplastic and chronic inflammatory disorders, but whether angiogenesis also occurs in inflammatory bowel disease (IBD) has yet to be established. We assessed mucosal vascularization, expression of endothelial alphaVbeta3 integrin, angiogenic factors, and their bioactivity in Crohn's disease (CD) and ulcerative colitis (UC) mucosa. METHODS: Mucosal endothelium was immunostained for CD31 and factor VIII and quantified by digital morphometry. alphaVbeta3 expression was studied in vivo by confocal microscopy and in vitro by flow cytometric analysis of human intestinal microvascular endothelial cells (HIMECs). Vascular endothelial growth factor (VEGF), interleukin (IL)-8, and bFGF levels were measured in mucosal extracts and cells and angiogenic bioactivity shown by induction of HIMEC migration and the corneal and chorioallantoic membrane angiogenesis assays. RESULTS: Microvessel density was increased in IBD mucosa. Endothelial alphaVbeta3 was strongly expressed in IBD but only sporadically in normal mucosa and was up-regulated in HIMECs by VEGF, tumor necrosis factor alpha, and bFGF. IBD mucosal extracts induced a significantly higher degree of HIMEC migration than control mucosa, and this response was mostly dependent on IL-8 and less on basic fibroblast growth factor or vascular endothelial growth factor. Compared with normal mucosa, IBD mucosal extracts induced a potent angiogenic response in both the corneal and chorioallantoic membrane assays. CONCLUSIONS: These results provide morphological, phenotypic and functional evidence of potent angiogenic activity in both CD and UC mucosa, indicating that the local microvasculature undergoes an intense process of inflammation-dependent angiogenesis. Thus, angiogenesis appears to be an integral component of IBD pathogenesis, providing the practical and conceptual framework for anti-angiogenic therapies in IBD. 相似文献
