目的 :建立测定血浆内瑞巴匹特浓度的高效液相色谱法。方法 :KromasilC18色谱柱 ,甲醇 -0 .0 5mol·L- 1磷酸二氢钾 (55∶45,pH3.0 )为流动相 ,并由序贯单纯形法确定此为最佳配比点 ,流速 1 .0ml·min- 1,检测波长 2 30nm。结果 :瑞巴匹特在 1 5~ 1 2 0 0 μg·L- 1范围内 ,血浆药物浓度C与色谱峰高H呈现良好线性相关 ,血浆中瑞巴匹特平均相对回收率为(97.5± 0 .5) %(n =5) ,平均绝对回收率为 (90 .3± 1 .8) %(n =5) ;平均日内和日间RSD分别为 4.6 8%和 5.6 6 %(n =5) ;方法对血浆瑞巴匹特最低检出浓度为 2 μg·L- 1。结论 :本方法适于血浆中瑞巴匹特的测定。 相似文献

A case of acute myeloblastic leukemia is reported in which, at diagnosis, 50% of the cells in bone marrow and blood contained double minutes (DMs). These cells disappeared from the marrow and blood following chemotherapy. A specific and sensitive radioimmunoassay has been developed to monitor plasma levels of captopril, the first orally active angiotensin-converting enzyme inhibitor. Because of the reactive nature of the captopril thiol group, captopril was measured as the captopril-N-ethylmaleimide complex (captopril-NEM). Accuracy studies, using samples with known added concentrations of captopril, were satisfactory (r = 0.95, p less than 0.001, y = 0.97x - 2.02), and the radioimmunoassay results compared well with those determined by a gas chromatography/mass spectrometric method (r = 0.98, p less than 0.0005, y = 1.2x - 19). Objective To assess the activities of biapenem against multidrug-resistant and extensively drug-resistant Mycobacterium tuberculosis. Methods Biapenem/clavulanate(BP/CL) was evaluated for in vitro activity against Mycobacterium tuberculosis(Mtb) multidrug-resistant(MDR) isolates, extensively drug-resistant(XDR) isolates, and the H37 RV strain. BP/CL activity against the H37 Rv strain was assessed in liquid cultures, in macrophages, and in mice. Results BP/CL exhibited activity against MDR and XDR Mtb isolates in liquid cultures. BP/CL treatment significantly reduced the number of colony forming units(CFU) of Mtb within macrophages compared with control untreated infected macrophages. Notably, BP/CL synergized in pairwise combinations with protionamide, aminosalicylate, and capreomycin to achieve a fractional inhibitory concentration for each pairing of 0.375 in vitro. In a mouse tuberculosis infection model, the efficacy of a cocktail of levofloxacin + pyrazinamide + protionamide + aminosalicylate against Mtb increased when the cocktail was combined with BP/CL, achieving efficacy similar to that of the positive control treatment(isoniazid + rifampin + pyrazinamide) after 2 months of treatment.
