Predictive value of early HCV RNA quantitation for sustained response in nonresponders receiving daily interferon and ribavirin therapy

The prognostic value of early hepatitis C virus (HCV)-RNA load was evaluated among nonresponder patients to previous interferon (IFN) therapy treated with daily IFN and ribavirin. One hundred-six nonresponders (83 men), mean age 44.8 +/- 11 years, were treated with IFN-alpha 2b 3 MU/day for 24 weeks, followed by 3 MU x 3/week for 24 weeks plus ribavirin 1-1.2 g/day for 48 weeks. HCV RNA was quantified by Versant HCV RNA 3.0 assay (Bayer). The predictive values of the baseline and the change in viral load at week 1, 4, and 12 for sustained virological responses were analyzed using receiver operating characteristic (ROC) curves, as well as predictive values of >2 log(10) drop from baseline by weeks 1, 4, and 12 in combination with undetectable HCV RNA for sustained virological response. Thirty-two patients (30.2%) were sustained virological responders. The highest area under the curve was obtained at week 4. The unquantifiable HCV RNA level, in combination with at least a 2 log(10) drop in viral load by week 4 and week 12, had a negative predictive value of 96% and 97%, respectively. Nonresponse can be predicted as early as week 4 or week 12 in nonresponders treated with daily IFN and ribavirin.     

Nucleotide sequence of RNA segment 3 of the avian influenza A/FPV/Rostock/34 and its comparison with the corresponding segment of human strains A/PR/8/34 and A/NT/60/68

The nucleotide sequence of RNA segment 3 of A/FPV/Rostock/34 (H7N1), an avian strain of influenza A virus, has been determined from a cloned DNA copy. Segment 3 codes for the PA polypeptide and the sequence specifies an acidic polypeptide of 716 amino acid residues. Comparison of the sequence with the corresponding segment of two human strains A/PR/8/34 and A/NT/60/68 indicates significant divergence of the avian sequence from the human sequences at the nucleotide level. At the amino acid level there is considerably greater homology between the avian and human strains. This presumably reflects a constraint on divergence of the PA polypeptide imposed by a common functional requirement of PA in all influenza virus strains.

Metastatic potential of human CX-1 colon adenocarcinoma cells is dependent on the expression of sialosyl Le a antigen

Several lines of evidence indicate that sialosyl Le a , tumor-associated carbohydrate antigen present on human colon carcinoma cells, is involved in formation of metastases. To study the role of this carbohydrate structure in development of metastases, we have used the clone of human colon carcinoma CX-1 cells transfected with antisense expression vector containing fragment of cDNA for a1,3/4-fucosyltransferase (FT III), which is involved in synthesis of sialosyl Le a tetrasaccharide. It has been reported previously that, in contrast to the parental cells, the antisense-transfected CX-1.1AS5 cells do not express sialosyl Le a and do not adhere to E-selectin-expressing CHO cells. In the present work we have studied the formation of liver metastases by CX-1.1AS5 cells after their orthotopic or intrasplenic implantation into athymic nu/nu mice. After orthotopic implantation of sialosyl Le a -negative colon carcinoma CX-1.1AS5 cells, the number of mice with liver metas-tases was markedly lower (21% of mice) in comparison with their number after implantation of the parental CX-1.1 cells (86% of mice). However, no differences in ability to form colonies in liver were observed between parental CX-1.1 cells and antisense-transfected CX-1.1AS5 cells after intrasplenic inoculation. The liver metastases were formed in 89% and 84% of mice, respectively. Our data support the thesis on the importance of sialosyl Le a antigen expression in the development of liver metastases by colon cancer cells, and indicate the role of transplantation route and primary tumor localization in formation of metastases.© Kluwer Academic Publishers 1998     

Differential RNA fingerprinting as a tool in the analysis of spermatozoal gene expression

The apparent decline in human male fertility and the concomitantincrease in testicular pathology have prompted discussion ofthe underlying molecular mechanisms which may underpin theseobservations. While monitoring the expression of protamlne-2genes in the human ejaculate, we found a representative complementof sperm mRNAs following sequence-independent amplificationof reverse-transcribed cDNAs with the polymerase chain reaction(RT-PCR). The revelation of unique sperm-derived PCR productsusing this method suggests that it should now be possible toinvestigate gene expression in human spermatogenesis by differentialRNA fingerprinting of ejaculate spermatozoa. The Identificationof molecular markers and the corresponding genes associatedwith male Infertility will be considerably enhanced by theseinvestigations while obviating the requirement for invasivebiopsy.     

Argonaute蛋白家族在RNAi中的作用

RNA干扰是一种双链RNA诱导的基因沉默现象。Argonaute蛋白组成了一个高度保守的家族,该蛋白家族包括许多成员,它们组成了RNA诱导的沉默复合体的核心元件,是RNA干扰所必须的。Argo-naute蛋白选择性地与miRNA和siRNA结合,并与Dicer酶相互作用,其PIWI盒子与Dicer的RNaseⅢ结构域直接相互作用,PIWI与Dicer之间的相互作用可能会促进miRNA/siRNA的释放。Argonaute蛋白很可能是RNA干扰中核酸内切酶活性的执行者。     

CD40靶向小干扰RNA抑制大鼠异体肢体移植急性排斥反应后细胞凋亡及P53、P21表达

目的探讨CD40靶向小干扰RNA（即短发夹RNA，shRNA）对大鼠异体肢体移植急性排斥反应及细胞凋亡的影响。方法以纯系SD大鼠为供体，纯系Wistar大鼠为受体，行同种异体右后肢移植。27只大鼠肢体移植后随机分为3组：实验组．注射梭华一Sofast（15μl）-siCD40—2，pSilencer（100μg）载体复合物600μl；空载体对照组，在肢体移植后，即注射Sofast（15μl）-pSilencer4．1-CMVneo（100μg）空载体复合物600μl；生理盐水对照组，在肢体移植注射生理盐水600μl。观察移植物排斥反应征象及存活情况，并于第7天对产生免疫耐受大鼠进行混合淋巴细胞反应（MLR），同时进行组织学检查。结果与其他组相比．实验组移植物发生排斥反应的时间及存活时间均显著延长（P〈0．01）（〉13d），未见排斥反应征象，其他组均于术后近期发生排斥反应；实验组大鼠对供体的淋巴细胞呈现低反应性，移植的供体同系大鼠的肢体得以存活。实验组移植物细胞凋亡率低于其他组。结论在术后不应用免疫抑制剂的情况下，CD40靶向的shRNA干扰可以抗大鼠异体肢体移植急性排斥反应。     

Effect of leukocytic pyrogen on the phagocytic properties of macrophages in tissue culture

The effect of rabbit leukocytic pyrogen (LP) on the phagocytic activity of peritoneal macrophages from albino mice toward shigellas was investigated. The effect was found to depend on dose: LP in a large dose inhibited phagocytosis whereas small doses of LP were not very effective; the addition of LP to macrophages in average doses, after administration of kanamycin, stimulated both the phase of ingestion and the phase of digestion of the shigellas. Stimulation of phagocytosis by LP was accompanied by an increase in the activity of the lysosomal enzyme acid phosphatase in the macrophages; changes in the RNA content were not significant.Department of General Pathology, Institute of Experimental Medicine, Leningrad. Simple two-electrode biosignal amplifier

A simple, cost effective circuit for a two-electrode non-differential biopotential amplifier is proposed. It uses a ‘virtual ground’ transimpedance amplifier and a parallel RC network for input common mode current equalisation, while the signal input impedance preserves its high value. With this innovative interface circuit, a simple non-inverting amplifier fully emulates high CMRR differential. The amplifier equivalent CMRR (typical range from 70–100 dB) is equal to the open loop gain of the operational amplifier used in the transimpedance interface stage. The circuit has very simple structure and utilises a small number of popular components. The amplifier is intended for use in various two-electrode applications, such as Holter-type monitors, defibrillators, ECG monitors, biotelemetry devices etc.
