目的研究PTTG和MMP-9在食管癌中的表达及其相关性,探讨它们对食管癌临床分期的价值。方法应用免疫组织化学SP法检测84例食管癌组织中PTTG和MMP-9蛋白表达,结合临床病理参数并将其纳入食管癌临床分期进行预后分析。结果PTTG和MMP-9在食管癌组织中均呈阳性表达,且两者表达呈正相关。PTTG、MMP-9蛋白在食管癌临床分期较晚的患者中阳性表达率明显高于临床分期早的患者。结论PTTG和MMP-9蛋白在食管癌组织中呈高表达,且与食管癌临床分期显著正相关,提示两者在食管癌发生、发展及侵袭转移过程中可能起重要作用。  相似文献

目的探讨重组载体介导的Survivin小干扰RNA(siRNA)对人结肠癌细胞株Lovo中PTTG蛋白的影响。方法运用靶向Survivin siRNA真核表达载体转染结肠癌Lovo细胞,western blot检测PTTG蛋白的表达水平。结果 Survivin基因沉默后24-72h,与正常对照组相比,Survivin和PTTG蛋白表达均明显降低,差异具有统计学意义(P<0.01)。结论 Survivin基因沉默后结直肠癌细胞PTTG蛋白表达显著下调,提示Survivin和PTTG可能相互促进共同参与结直肠癌的发生发展。  相似文献

目的:探讨垂体肿瘤转化基因(Pituitary Tumor Transforming Gene,PTrG)蛋白在舌癌细胞株Tca8113中的表达及其与碱性成纤维细胞生长因子(basic Fibroblast Growth Factor,bFGF)间的关系.方法:应用免疫细胞化学方法,检测加入bFGF抗体后孵育的癌细胞中PTTG蛋白表达的变化.结果:Tca8113中PTTG蛋白呈阳性表达,并受bFGF影响,加入bFGF抗体的培养液中的癌细胞PTTG蛋白荧光明显减弱,随着时间延长和bFGF抗体滴度增加,减弱趋势明显.结论:舌癌细胞株中PTTG蛋白的表达受bFGF制约,对二者的联合治疗可能是一种治疗舌癌的有效方法.  相似文献

目的:探讨食管鳞癌组织中PTTG mRNA与蛋白的表达及其二者间的相互关系。研究它们的表达与肿瘤临床资料之间的联系。方法:采用原位杂交技术检测30例食管鳞癌及相应的癌旁组织中PTTGmRNA的表达,同时采用免疫组织化学SP法检测相应标本中PTTG蛋白的表达。结果:在食管鳞癌和相应的癌旁组织中PTTG mRNA表达的阳性率分别为66.7%(20/30),10%(3/30)。PTTG蛋白表达的阳性率分别为76.7%(23/30),16%(5/30)。食管鳞癌组远高于癌旁组,有显著性差异(P<0.01)。PTTG mRNA与蛋白在食管鳞癌组织的阳性表达具有一致性(P>0.05),其阳性率高低与性别、年龄、肿瘤大小无关,而与组织学分级、淋巴结转移、TNM分期有关。结论:PTTG基因的异常表达可能对食管鳞癌的发生和发展起重要作用,它可能为食管鳞癌的早期诊断和基因治疗提供新的靶点。  相似文献

Human pituitary tumor-transforming gene (PTTG) is a proto-oncogene involved in the development, invasion, and metastasis of many types of cancer, including ovarian cancer. However, little is known about the role of PTTG in the metabolic shift of ovarian cancer cells. In our study, we show that PTTG expression was positively correlated with the differentiation degree of ovarian cancer tissue. In addition, PTTG suppression by specific shRNA could inhibit the proliferation of ovarian cancer cells A2780 and SKOV-3. Furthermore, aerobic glycolysis was suppressed and oxidative phosphorylation was increased in ovarian cancer cells after PTTG suppression. We further found that the expression of c-myc and several crucial enzymes involved in aerobic glycolysis (e.g., PKM2, LDHA, and glucose transporter 1 (GLUT-1)) were downregulated by PTTG knockwown. Overexpression of c-myc could prevent the metabolic shift induced by PTTG knockwown. Together, our findings suggest that the oncogene PTTG promotes the progression of ovarian cancer cells, and its loss resists tumor development, in part, by regulating cellular metabolic reprogramming that supports cell growth and proliferation via c-myc pathway.  相似文献

目的:通过检测垂体腺瘤中垂体瘤转化基因(PTTG)、周期素依赖激酶抑制因子P27 mRNA及其蛋白的表达,以及与增殖细胞核抗原(PCNA)蛋白表达之间的相关关系,探讨PTTG、P27 mRNA及其蛋白的表达与垂体腺瘤增殖活性之间的关系。方法:30例垂体腺瘤手术标本分为侵袭组和非侵袭组,RT—PCR检测PTTG和P27 mRNA的表达,免疫组化检测PTTG、P27和PCNA蛋白的表达,并分析PTTG、P27蛋白表达与PCNA蛋白表达之间的相关性。结果:侵袭组PTTG mRNA的表达显著高于非侵袭组,P27 mRNA在侵袭组和非侵袭组的表达无显著差异,侵袭组PTTG蛋白表达显著增高,与PCNA蛋白表达呈正相关,P27蛋白表达显著减低,与PCNA蛋白表达呈负相关。结论:PTTG mRNA和蛋白在侵袭性垂体腺瘤的高表达导致垂体腺瘤细胞过度增殖,P27对垂体腺瘤细胞增殖的影响是通过翻译或翻译后机制调控的,二者对细胞增殖过程的影响呈细胞周期依赖性。  相似文献

Objective:To investigate the expressions and their relationship of pituitary tumor transforming gene(PTTG)and proliferating cell nuclear antigen (PCNA)in glioma.Methods:The protein expressions of PTTG and PCNA were detected by immunostaining assay using streptavidin-peroxidase(SP)method in 80 cases of glioma.Result:The positive rates of PTTG in grades Ⅰ-Ⅳ gliomas were 56.3%,68.2%,80.8%,and 100.0%respectively,and the protein expression of PTTG increased with the increasing of the pathological grade(X2=9.602,P<0.05):The positive rates of PCNA protein were 37.5%,54.5%, 69.2%.and 93.8%respectively,and the protein expression of PCNA increased with the increasing of the pathological grade (X2=12.147.P<0.01).The expression of PTTG had positive correlation with the expression of PCNA protein(Y8=0.557,P<0.01).Conclusion:The expressions of PTTG and PCNA proteins were related to malignant degree of gtioma.and may cooperate with each other in the tumorigenesis and progression and can be considered as the indicators of the biological behaviors in glioma.  相似文献

Pituitary adenomas (PAs) are noncancerous tumors, and about 35% of those reported to be invasive have been classified as “invasive pituitary adenomas (IPAs)”. In clinical, operative complications, total resection failures, and high relapse rates result from invasive features during the therapeutic process. Invasive mechanism is a complex process, including metalloproteases, inhibitors and tumor microenvironment factors etc. Thus, studying invasive mechanism of PAs might contribute to understanding its biological behavior. In our research, three type tissue samples of human, pituitaries, PAs, IPAs, their mRNA expression of MMP1, MMP2, MMP9, MMP14 and MMP15 were measured using real-time PCR. MMP2 and MMP14 protein levels also were measured with immunohistochemistry in same samples. We confirmed that elevated matrix metalloproteinase-14 expression correlates with invasive characteristics of IPAs. To investigate molecular mechanism of how MMP14 contributes to invasiveness, an ATT20 cell was used in this study. After transient-transfection of the MMP14-shRNA expression vector into ATT20 cells, we observed that mRNA expression of PTTG, VEGF, and TGFβ was significantly suppressed in interference groups. Meanwhile, ATT20 cells in high concentration TIMP-1 environment exhibit reduced PTTG, VEGF, and TGFβ expression accompanied with the down-regulation of MMP14. Thus, we propose that MMP14 plays an important role in tumor invasion and angiogenesis and that a novel regulatory pathway for MMP14 may exist through VEGF and PTTG. In brief, MMP14 may be a target for therapeutic treatment.  相似文献

目的:研究PTTG与垂体瘤的各亚型的相关性.方法:用免疫组化方法检测92例垂体肿瘤病人PTTG.其中29例GH,20例PRL,10例ACTH,11例FSH/LH,9例TSH和13例无功能腺瘤.结果:PTTG在细胞质内显著表达,各亚型中GH型是表达最高,PRL腺瘤表达最低,各亚型的阳性表达率与PRL亚型阳性表达率对比均有统计学意义.结论:重体腺瘤不同垂体亚型中PTTG的表达阳性率及表达强度不同,PTTG检测,可为早期诊断垂体腺瘤( GH、PRL、ACTH、FSH/LH、TSH)亚型和无功能型提供参考.  相似文献
