不同工艺对九味羌活颗粒中欧前胡素及异欧前胡素含量的影响

目的研究不同制备工艺对九味羌活颗粒中成分欧前胡素、异欧前胡素含量的影响.方法采用高效液相法对自制和市售九味羌活颗粒样品中欧前胡素、异欧前胡素的含量进行测定.以Agilent C18(4.6mm×250mm,5μm)为固定相,乙腈-水(55∶45,v/v)为流动相,流速为1.0mL?min-1,检测波长为250nm.结果自制含挥发油与不含挥发油的九味羌活颗粒样品中欧前胡素、异欧前胡素含量差异非常明显,前者含量大约是后者的1/10,且含挥发油样品与市售样品含量相当.结论该实验所得结论,对九味羌活颗粒的制备工艺的改进具有借鉴意义.     

The scaffold protein JSAP1 regulates proliferation and differentiation of cerebellar granule cell precursors by modulating JNK signaling

Cerebellar granule cell precursors (GCPs) proliferate in the outer part of the external granular layer (EGL). They begin their differentiation by exiting the cell cycle and migrating into the inner part of the EGL. Here we report that JSAP1, a scaffold protein for JNK signaling pathways, is expressed predominantly in the post-mitotic GCPs of the inner EGL. JSAP1 knockdown or treatment with a JNK inhibitor enhances the proliferation of cultured GCPs, but the overexpression of wild-type JSAP1 leads to increased proportions of p27^Kip1- and NeuN-positive cells, even with saturating concentrations of Sonic hedgehog (Shh), a potent GCP mitogen. However, these differentiation-promoting effects on GCPs are attenuated significantly in cells overexpressing a mutant JSAP1 that lacks the JNK-binding domain. Together, these data suggest that JSAP1 antagonizes the mitogenic effect of Shh on GCPs and promotes their exit from the cell cycle and differentiation, by modulating JNK activity.     

菊葛天麻颗粒挥发油的包结及表征

目的确定菊葛天麻颗粒挥发油与β-环糊精的包结工艺,分析测定包合物。方法以包结率和收得率为指标,采用正交L9(34)试验,优选包结条件;以X射线衍射法,对挥发油与β-环糊精形成包合物进行分析。结果挥发油与β-环糊精用量比为1∶10,包结温度为40℃,搅拌时间为3h,为挥发油与β-环糊精最佳包结条件,形成β-环糊精包合物。结论按实验确定的挥发油包结方法,可成功的将挥发油包结起来。     

Granulation of Acetaminophen by a Rotating Fluidized-Bed Granulator

The purpose of this research was to evaluate the use of a rotating fluidized-bed granulator to produce acetaminophen granules with sufficient binding force between particles and good plasticity in tablets. Ethenzamide and ascorbic acid were used to compare the relationship between granulation and the sample wetness. It was revealed that a blade rotation rate of 300 rpm, inlet air flow rate of 42 m³/hr, and spraying pressure of 1.5 kg/cm³ produced tablets with the best properties. The granule and tablet properties of ethenzamide and ascorbic acid were compared to those of acetaminophen. These compounds showed different wetting behaviors with water and different compression behaviors. With an increase in medicament content, tablet hardness increased except for the ascorbic acid formulation. Capping and sticking were observed in acetaminophen and in ascorbic acid, respectively, and acetaminophen and ethenzamide showed prolonged disintegration time.     

Expression of transient receptor potential (TRP) channel mRNAs in the mouse olfactory bulb

Transient receptor potential (TRP) channels are a large family of cation channels. The 28 TRP channel subtypes in rodent are divided into 6 subfamilies: TRPC1-7, TRPV1-6, TRPM1-8, TRPP2/3/5, TRPML1-3 and TRPA1. TRP channels are involved in peripheral olfactory transduction. Several TRPC channels are expressed in unidentified neurons in the main olfactory bulb (OB), but the expression of most TRP channels in the OB has not been investigated. The present study employed RT-PCR as an initial survey of the expression of TRP channel mRNAs in the mouse OB and in 3 cell types: external tufted, mitral and granule cells. All TRP channel mRNAs except TRPV5 were detected in OB tissue. Single cell RT-PCR revealed that external tufted, mitral and granule cell populations expressed in aggregate 14 TRP channel mRNAs encompassing members of all 6 subfamilies. These different OB neuron populations expressed 7–12 channel mRNAs. Common channel expression was more similar among external tufted and mitral cells than among these cells and granule cells. These results indicate that a large number of TRP channel subtypes are expressed in OB neurons, providing the molecular bases for these channels to regulate OB neuron activity and central olfactory processing.     

复方泽漆冲剂对不同证型寻常性银屑病患者IL-23/Th 17细胞轴的影响

目的观察复方泽漆冲剂对血热、血瘀、血燥三型寻常性银屑病(PV)患者血清中IL-17,IL-22和IL-23的影响,探讨复方泽漆冲剂对三型PV患者IL-23/Th17细胞轴的影响。方法三型PV患者均口服复方泽漆冲剂,每型患者均为20例,疗程4周,采用ELISA法对60例PV患者治疗前后和10例健康对照组的血清中IL-17,IL-22和IL-23水平进行检测。结果 60例PV患者血清中的IL-17,IL-22和IL-23的水平治疗前、后均明显高于健康对照组(P均<0.01)。三型患者治疗后的IL-17,IL-22和IL-23的水平与治疗前相比,均明显下降,(P均<0.01)。血热型治疗前、后的IL-17,IL-22和IL-23分别为23.369±2.402pg/mL,12.753±2.014pg/mL;200.694±20.087pg/mL,135.772±10.551pg/mL;23.124±1.886pg/mL,16.156±2.481pg/mL。血瘀型治疗前后的IL-17,IL-22和IL-23分别为23.456±2.142pg/mL,14.944±1.912pg/mL;200.554±19.725pg/mL、146.112±16.523pg/mL;22.562±1.768pg/mL,14.512±2.111pg/mL。血燥型治疗前后的IL-17,IL-22和IL-23分别为24.556±1.653pg/mL,16.836±2.161pg/mL;204.754±19.428pg/mL,155.702±11.489pg/mL;23.362±2.462pg/mL、12.876±2.462pg/mL。结论复方泽漆冲剂能降低三型PV患者血清中IL-17,IL-22和IL-23的水平,其中对血热型的作用最强,对血瘀型次之,对血燥型最弱。     

Calbindin D28K expression in relation to granule cell dispersion, mossy fibre sprouting and memory impairment in hippocampal sclerosis: a surgical and post mortem series

Reorganisation of the dentate gyrus, including granule cell dispersion (GCD) and mossy fibre sprouting, typically accompany hippocampal sclerosis (HS) in temporal lobe epilepsy. Calbindin (CB) expression in granule cells increases during infancy, influences granule cell excitability, vulnerability to excitotoxicity in addition to important physiological functions in memory. Our aim was to study CB patterns in relation to dentate gyrus re-organisation, epilepsy history and memory function. Forty-five surgical cases and 11 post mortems were examined from patients with drug-resistant epilepsy in addition to three post mortem controls. In the surgical cases, CB expression, and the degree of GCD and mossy fibre sprouting were measured. In post mortem cases, CB expression was assessed in relation to the pattern of HS along the rostral-caudal axis of the hippocampus, and compared to PM controls. Three patterns were identified. In Group 1 (40%), the most dispersed granule cells were CB-positive and basal cells negative imitating developmental patterns. In Group 2 (47%), normal CB expression was retained and in Group 3 (13%), CB was predominantly lost in granule cells. These patterns correlated with GCD, the presence of mossy fibre sprouting and the pattern of HS. Group 1 was associated with early onset of seizures but not independently predictive of outcome. In post mortem cases, altered CB expression lateralised to the side of HS and persisted despite seizure remission in some patients. No significant correlation between CB expression and memory function was identified. CB expression patterns in HS may indicate developmental dysmaturation and are associated with the extent of GCD and mossy fibre sprouting in HS. The functional significance of CB loss, in terms of epileptogenesis and effects on memory, remain uncertain.     

参芪清热颗粒联合复方丹参注射、生脉注射液治疗急性白血病临床研究

[目的]探究参芪清热颗粒联合复方丹参注射液、生脉注射液治疗急性白血病(Acute leukemia,AL)的疗效.[方法]将193例AL患者分为治疗组106例,对照组87例.对照组单纯给予国际标准化疗方案治疗;治疗组在此基础上采用复方丹参、生脉注射液逆转耐药并中药分阶段干预治疗,观察并比较两组疗效.[结果]治疗组初治患者有效率81.43％,与对照组初治有效率74.36％比较,差异无统计学意义(P＞0.05);难治患者有效率69.44％,与对照组难治有效率38.46％比较,差异有统计学意义(P＜0.05).治疗组白细胞最低值及感染率与对照组比较,变化无统计学意义(P＞0.05);白细胞＜2.0×109/L的持续时间及感染时间比对照组缩短(P＜0.05);血制品的输注亦比对照组减少(P＜0.05),心脏副作用减轻.[结论]采用益气养阴、活血化瘀中药制剂联合生脉、复方丹参注射液配合化疗治疗AL,能提高疗效,降低毒副作用,改善患者生存质量.     

来氟米特片联合肾易通颗粒治疗系膜增生性肾小球肾炎临床疗效分析

目的 分析来氟米特片联合肾易通颗粒治疗系膜增生性肾小球肾炎的临床效果。方法 采用病例对照分析研究,将65例患者随机分为来氟米特组（25例）、来氟米特加肾易通组（20例）、肾易通组（20例）。观察临床症状、体征以及尿常规、尿红细胞计数、24 h尿蛋白定量、血清白蛋白（ALB）、血常规、血尿素氮、血肌酐（Ser）、谷丙转氨酶、谷草转氨酶的变化以及用药期间不良反应。结果 来氟米特组各疗程有效率虽较肾易通组高,但差异无显著性（P＞0.05）;来氟米特加肾易通组治疗6个月后的疗效（100%）不仅优于肾易通组（P＜0.05）,而且优于来氟米特组（P＜0.05）。结论 来氟米特联合肾易通颗粒对系膜增生性肾小球肾炎患者具有疗效好、不良反应少的特点,值得临床推广。
