采用细胞生物学方法测定类风湿性关节炎( R A)14 例血清 I L- 6 、 T N F 水平,均显著高于正常对照组( P< 0 .001) 。相关分析显示 I L- 6 与 T N F 水平变化呈正相关( P< 0 .01) , I L- 6 、 T N F 与血沉亦呈正相关( P<0 .05) 。认为, I L- 6 、 T N F 参与 R A 的发病,两者在致病中有协同相同;其水平变化与 R A 的活动性及血沉相关。  相似文献

We hypothesized that hypothalamic NPYergic mechanisms mediate the blood pressure lowering effect of caloric restriction in hypertensive rats. Aortic coarctation-induced (AC) hypertensive rats (n=25) were assigned to either an ad libitum fed control group (AL) or food restricted group (FR; 60% of AL consumption) for 3 weeks. Rats were instrumented chronically with vascular catheters and bilateral guide cannulae directed at the paraventricular hypothalamic nuclei (PVN). Blood pressure (BP) and heart rate (HR) responses to bilateral PVN microinjection of saline (200 nl) or the putative NPY receptor antagonists [D-Trp32]NPY(1-36) (3.3 micrograms/200 nl) and [D-Tyr27,36 Thr32]NPY(27-36) (D-NPY(27-36); 3.3 micrograms/200 nl) were determined. The FR rats were then refed and cardiovascular responses to PVN injections of NPY receptor antagonists were again determined. FR rats had significantly reduced resting BP (159+/-4 vs. 129+/-4 mmHg) and HR (360+/-11 vs. 326+/-9 bpm) compared to AL controls. Refeeding restored BP and HR of FR rats to levels similar to AL (BP=153+/-4 mmHg, HR=359+/-11 bpm). PVN administration of [D-Trp32]NPY produced foraging behavior and concurrent increases in BP and HR in FR, AL and Re-fed rats. The behavioral activation suggests that [D-Trp32]NPY(1-36) produced activation of NPY receptors. In contrast, D-NPY (27-36) did not produce any behavioral response or affect BP or HR in AL or Re-fed rats. In FR rats, D-NPY (27-36) produced significant increases in BP (peak=15+/-3 mmHg) which partially reversed the effect of FR on BP. Thus, in FR rats with reduced BP, PVN administration of an NPY receptor antagonist increases BP. NPY blockade in the PVN accounted for about 50% of the BP effect of food restriction, thus other mechanisms are likely to be involved. These findings are consistent with the hypothesis that NPYergic mechanisms may contribute to the reduction of BP produced by food restriction.  相似文献

The phenomenon of artificially induced local leucocyte reactions during the supravital period could be of practical importance, but has not yet been comprehensively investigated. For a more detailed evaluation, experiments with the chemotactic agents interleukin-1 (IL-1) and N-formyl-methionyl-leucyl-phenylalanine (FMLP) were performed by subcutaneous injection into various anatomical regions (back, abdomen, limbs) of NMRI-mice (National Medical Research Institute) and pigs 0–5 min after circulatory arrest. Phosphate buffered saline (PBS) without effective components was administered to equivalent areas of the animals as a control. Tissue specimens were collected at 6 h postmortem (mice) and 12–14 h postmortem (pigs), cut into serial sections, stained with H & E and examined under the microscope. A leucocyte reaction did not develop in pigs (n = 10, 30 tissue samples) following injection of FMLP, however, dermal, subcutaneous and perivascular infiltration of leucocytes (in particular mononuclear cells and a few granulocytes) was found in 3 out of 30 tissue specimens in murine experiments. In addition intravascular cell accumulations were detected in 2 out of 30 samples. The injection of IL-l to mice gave similar results, i.e. aggregations of leucocytes and intravascular cell accumulations in 4 out of 30 and 3 out of 30 tissue samples, respectively. In negative controls no leucocyte reaction was detectable. This shows that potent chemotactic factors such as IL-1 and FMLP administered in the early supravital period can induce moderate local leucocyte reactions in animal models in at least some cases. A clear morphological differentiation between vital and supravital chemotaxis does not seem to be possible. The supravitally stimulated accumulations of leucocytes are interpreted as an aggregation of resident macrophages in combination with a slight migration of blood leucocytes. Presumably, these alterations are restricted to the very early supravital period as long as sufficient energy reserves are available. It must be stated that the observed changes are reactions, not spontaneous actions, so that the general validity of the phenomenon of leucocyte infiltration as a vital parameter is not affected.  相似文献

Summary A 73-year-old man with chronic renal failure of undetermined aetiology had received haemodialysis for 12 years when he died of acute purulent peritonitis due to caecal perforation. Amyloid deposits detected in a cystic bone lesion in the left hip had caused a pathological fracture 17 days before death. At autopsy, extensive amyloid deposits were found in the osteoarticular system, in the cartilaginous surface and the capsular tissue of joints, ligaments, vertebral discs and bone. In addition, vascular amyloid deposits were diagnosed in the heart, kidneys, testes, lungs, skin and in the gastrointestinal tract. A special feature of this case were interstitial amyloid deposits forming a fine-meshed structure in the myocardium and plate-like deposits in the gastrointestinal tract. Immunohistochemically, all these deposits reacted strongly with antibody to human 2-microglobulin but showed no reaction with antibodies to AA, Alambda, A-kappa and AF. The present case demonstrates that extra-osteoarticular manifestations of AB-amyloidosis can cause serious complications.  相似文献

目的 探讨 p2 7kip1和 p5 3基因蛋白在恶性卵巢上皮性肿瘤中的表达及其与预后的关系。 方法 1992年 11月至 1999年 12月应用免疫组化检测 5 6例恶性卵巢上皮性肿瘤石蜡切片中 p2 7kip1和p5 3基因蛋白表达情况 ,并应用Kaplan -Mier法及多变量Cox比例风险回归模型分析患者预后。 结果  5 6例恶性卵巢上皮性肿瘤患者的 p2 7kip1基因蛋白表达率为 4 1 1% ;p5 3基因蛋白表达率为 4 8 2 % ,两种基因蛋白表达比较 ,差异无显著性 (P >0 0 5 )。p2 7kip1及 p5 3基因蛋白表达与肿瘤的病理分级有关 ( P <0 0 5 ) ,与其他临床病理特征无关。单因素分析 :p2 7kip1及 p5 3基因蛋白表达与患者的中位生存时间、生存曲线各时点生存率密切相关。 结论 p2 7kip1基因蛋白表达对恶性卵巢上皮性肿瘤的预后有显著影响 ,而 p5 3基因蛋白表达与预后关系不显著  相似文献

目的 探讨雷公藤多甙 (TW )对狼疮性肾炎 (LN)患者血浆白介素 - 13(IL - 13)含量的影响及治疗价值。方法 分别将激素诱导缓解治疗后的LN患者 ,按其活动指标分为活动期和非活动期 2组 ,活动期患者加用TW治疗 ,观察治疗 3个月后血浆IL - 13水平的变化和肾炎的缓解情况。结果 血浆IL - 13水平明显下降 ,尿蛋白减少 ,狼疮活动性免疫指标、血尿和肾功能改善。结论 TW可抑制血浆IL - 13的产生 ,对控制LN的活动有一定的治疗价值。  相似文献

目的 为了开发我国珍稀动物——吉林双阳梅花鹿的基因资源,在分子水平上揭示鹿药材的药理作用。方法 构建了梅花鹿脾脏细胞cDNA质粒文库,克隆了一些看家基因并进行了序列分析。结果 其中的一个序列在核酸及推导的氨基酸序列水平上与编码人、犬、大鼠、小鼠核蛋白体蛋白L27(ribosomal protein L27)cDNA序列和对应的氨基酸序列高度同源,并具有完整的开放阅读框架(ORF)。结论 发现了双阳梅花鹿核蛋白体蛋白L27全长cDNA序列,此序列已经在(Genbank中登录,登录号为:AF373231。  相似文献

Fibronectin (a glycoprotein which modulates inflammation) may decrease mortality in systemic infection. Children with meningococcal disease (MCD) may have low fibronectin levels. We aimed to compare plasma fibronectin levels in children with MCD and controls, correlate fibronectin levels with interleukin-6 (IL-6), shock and death, and assess fibronectin as an aid to early diagnosis in MCD. Samples were taken on admission from 99 children with MCD and 49 controls. Plasma fibronectin was measured using a turbidimetric immunoassay. Plasma fibronectin was significantly lower in MCD compared to controls (57 μg/ml vs 105 μg/ml; P < 0.005). Children who died had significantly lower levels than survivors (29 μg/ml vs 62 μg/ml; P = 0.01). Fibronectin levels were negatively correlated with IL-6 levels. Fibronectin was a poor predictor of MCD. Conclusion Plasma fibronectin levels are decreased in children with MCD, especially in shock and death. This decrease is associated with high IL-6 levels. 本文研究了从正常人精浆中分离纯化的酸性磷酸酶(ACP)对BALB/c小鼠脾脏细胞来源的T淋巴细胞产生IL-2的影响。本文采用MTT生物学测定法测IL-2活性。实验证明:人糖浆ACP对小鼠T淋巴细胞产生IL-2有明显的抑制作用(P<0.05),并且抑制作用随ACP剂量的增加而增强。这说明了精浆中大量ACP是构成精浆免疫抑制功能的重要组成成份之一。  相似文献
