大鼠局灶性脑缺血再灌注后Hsp70的表达及与细胞凋亡的关系

目的：研究大鼠局灶性脑缺血再灌注后热休克蛋白（ｈｅａｔ ｓｈｏｃｋ ｐｒｔｅｉｎ,Ｈｓｐ）７０的表达及与细胞凋亡的关系。方法：用线栓法制备ＭＣＡＯ模型,用免疫组化法测定Ｈｓｐ７０阳性细胞及用ＴＵＮＥＬ法测定凋亡细胞。结果：缺血２ｈ即有Ｈｓｐ７０阳性细胞,６～２４ｈ明显,４８ｈ减弱,３ｄ消失。ＴＵＮＥＬ阳性细胞于再灌注后２ｈ出现,２４～４８ｈ达高峰,７２ｈ减弱。结论：脑缺血再灌注后早期即有Ｈｓｐ７０表达及细胞凋亡发生。     

Effects of benzodiazepine receptor ligands on the performance of an operant delayed matching to position task in rats: opposite effects of FG 7142 and lorazepam

The effects of a series of benzodiazepine (BZ) receptor ligands, ranging from a full agonist through to partial inverse agonists, were examined on short term working memory in the rat. The behavioural paradigm used was a discrete trial, operant delayed matching to position task, as originally described by Dunnett (1985), with delays of 0, 5, 15 and 30 s. The benzodiazepine receptor (BZR) full agonist lorazepam (0.25, 0.375 and 0.5 mg/kg) dose and delay dependently impaired matching accuracy. Lorazepam also increased the latency to respond and decreased the number of nose pokes made into the food tray during the delays. In contrast, the BZR partial agonist ZK 95 962 (1, 3, 10 mg/kg) did not affect matching accuracy, but did increase the speed of responding. The BZR antagonist ZK 93 426 (1.25, 5, 25 mg/kg) had no effects in this paradigm. The BZR weak partial inverse agonists Ro 15-4513 (0.1, 1 and 10 mg/kg) and ZK 90 886 (1, 3 and 10 mg/kg) did not affect accuracy of performance. However, both of these drugs increased the latency to respond and decreased nose poke responses. These motoric effects were particularly strong following 10 mg/kg Ro 15-4513. This shows that the effects of drugs on the accuracy of responding and on the speed of responding can be dissociated. The BZR partial inverse agonist FG 7142 had effects on matching accuracy that were dependent upon dose. The lowest dose of FG 7142 (1 mg/kg) significantly improved accuracy, whereas the highest dose (10 mg/kg) impaired accuracy. This impairment induced by FG 7142 (10 mg/kg) was accompanied by an increase in the latency to respond and a decrease in the number of nose pokes. Taken together, these results show that the accuracy of delayed matching performance can be modulated in opposite ways by the BZR full agonist lorazepam and a low dose of the BZR partial inverse agonist, FG 7142.     

肿瘤标志物DR-70在恶性肿瘤诊断中的应用

目的：评价肿瘤标志物ＤＲ－７０在恶性肿瘤中的诊断应用。方法：应用美国ＡＭＤＬ公司的ＤＲ－７０＾ＴＭ ＥＬＩＳＡ试剂盒,测定对２０例正常人和５９例恶性肿瘤病人血清ＤＲ－７０含量。结果：各种恶性肿瘤ＤＲ－７０的阳性率为６６．７￣１００％,均较正常对照组有非常显著性差异（Ｐ〈０．００１）。结论：血清ＤＲ－７０是诊断恶性肿瘤有价值的标志物。     

藏药七十味珍珠丸对动物出血和凝血时间的影响

藏药七十味珍珠丸（RNSP）灌胃给药（ig）给药0．5g／kg和1g／kg对大、小鼠的出血时间和凝血时间无影响;对“血瘀”模型大鼠的凝血时间和血小板数也无统计学差异。而ig给药2g／kg对延长小鼠出血时间的作用非常明显。     

HSP70高表达对K_(562)细胞热耐力的影响

用硫酸镉诱导细胞ＨＳＰ７０ 高表达,ＲＴ—ＰＣＲ 检测ＨＳＰ７０ｍ ＲＮＡ 水平并观察ＨＳＰ７０ 水平对细胞热耐力的影响。发现硫酸镉作用于细胞１ｈ 后,细胞的ＨＳＰ７０ 水平显著增高, ＨＳＰ７０ 水平高的细胞其热耐力明显强于ＨＳＰ７０ 水平低的细胞组;ＨＳＰ７０ 水平与细胞热耐力间正相关。提示化学因素诱导的ＨＳＰ７０ 表达增高可以保护热暴露细胞,提高细胞热耐力     

Nephrosialidosis: ultrastructural and lectin histochemical study

Summary The neuropathological findings in a Japanese male with nephrosialidosis are reported. Clinically, coarse face, psychomotor retardation, macular cherryred spot and proteinuria were noted at 1 year and 7 months. He was diagnosed to have nephrosialidosis on the basis of a deficiency of -neuraminidase activity in both lymphocytes and cultured skin fibroblasts, and of severe glomerular and tubular involvement on renal biopsy. He died of multiple organ failure at 8 years and 6 months. There were numerous vacuoles and storage materials in visceral organs, particularly in the glomerular and tubular epithelial cells of the kidney and Kupffer cells as well as hepatocytes in the liver. Neuropathological examination revealed severe neuronal storage in the selected part of the central nervous system; lower motor neurons of the brain stem and spinal anterior horn cells, as well as neurons in the basal nucleus of Meynert. In the peripheral nervous system, sympathetic ganglia were severely affected. There was little or no neuronal storage in the basal ganglia, cerebral cortex or cerebellum, and demyelination was not found. Electron microscopic examination showed fine wavy multilamellar structures in the spinal anterior horn cells or Zebra body-like structures in the neurons of the Meynert's basal nucleus. Lectin histochemistry was positive for wheat germ agglutinin, Ricinus communis agglutinin-1 and peanut agglutinin within distended neurons. We conclude that the neuropathological feature in nephrosialidosis is not specific except for the selectiveness of the anatomical sites of involvement. It shares some aspects found in other types of sialidosis or galactosialidosis.     

Human pharmacokinetics of an analogue of atrial natriuretic factor

Summary The pharmacokinetics of ANP-270, a 26 amino acid analogue of alpha human natriuretic factor (-hANF) with a prolonged effect on isolated arterial preparations has been studied in 40 healthy males, in a doubleblind placebo controlled investigation. Placebo or ANP-270 0.3, 1.5 or 3.0 g/kg were given by intravenous bolus injection, each to groups of 10 subjects. Blood samples were assayed for ANP-270 by a specific sandwich ELISA.The disappearance of ANP-270 from plasma followed a two-compartment decay, with mean distribution and elimination half-lives of 2.6 min (n = 30) and 10.6 min (n=20), respectively. These estimates were similar to those obtained by other investigators for -hANF. Their brevity explains the lack of a prolonged effect of ANP-270 in vivo compared to -hANF.     

HSP70与FasL在子宫内膜异位症中的表达

目的 :探讨热休克蛋白 70 (HSP70 )和Fas配体 (FasL)在子宫内膜异位症(EM)发病中的作用。方法 :采用免疫组化链霉菌抗生物素蛋白 -过氧化物酶染色法 (S P法 )检测子宫内膜异位症 5 6例的异位内膜与在位内膜 (30例 )中HSP70和FasL蛋白的表达 ,并以正常子宫内膜为对照 (2 5例 )。结果 :异位内膜组织中HSP70与FasL均呈高表达 ,与正常内膜组差异有高度显著性 (P <0 .0 1) ,而且二者失去在正常内膜组织中表达的周期性变化。但在卵巢子宫内膜异位症 (OEM)与子宫腺肌症 (AM)的表达差异无显著性 (P>0 .0 5 )。结论 :异位内膜组织中HSP70和FasL蛋白均过度表达 ,可能在EM的发病中起重要作用     

HSP70的肿瘤免疫作用及其基因表达调节

热休克蛋白70能够与肿瘤特异性抗原多肽结合,形成热休克蛋白70-多肽复合物,它本身没有抗原性,其抗原性由它结合的多肽所决定,而它是作为"分子伴侣"参与肿瘤免疫作用过程.将这一特性加以应用,可能为肿瘤的治疗和消灭开辟新的途径.     

热休克蛋白70对星形细胞瘤凋亡调控的研究

目的 :研究热休克蛋白 70 (HSP70 )在星形细胞瘤 (astrocytoma)中的表达与意义。方法 :采用鼠抗人 HSP70单克隆抗体及免疫组织化学 SABC法。结果 :在 99例星形细胞瘤中 ,HSP70表达 70例 (70 .7% ) ;用于对照的正常脑组织中 ,HSP70阳性仅 1例 (10 % )。HSP70在星形细胞瘤中的表达强于正常对照组 (P<0 .0 5) ,HSP70在 级和 级、 级和 级、 级和 级星形细胞瘤相互之间表达无显著性差异 (P>0 .0 5) ,但在 50例低度恶性星形细胞瘤 (WHO 级～ 级 )中表达强于在49例高度恶性星形细胞瘤 (WHO 级～ 级 )中的表达 (P<0 .0 5)。结论 :HSP70在大部分星形细胞瘤中有不同程度表达 ,HSP70可能参与星形细胞瘤的凋亡调变 ;其表达与星形细胞瘤分化有关
