The current study was carried out with an attempt to separate similarly structured title drugs by liquid chromatography. Spectrophotometric techniques were generally insufficient under these conditions because of the spectral overlapping of drugs with similar functional groups. The pharmaceutical drugs prazosin, terazosin, and doxazosin contain the same parent quinazoline nucleus, thus making it especially difficult to separate the former two drugs because of their very similar structures. A simple and sensitive method for the routine determination of these drugs in pharmaceutical formulations was attempted. We found that the mobile phase consisting of A: ACN–diethylamine (0.05 ml), B: methanol, and C: 10 mM Ammonium acetate separated these drugs effectively. Separations were carried out on a new Kromasil C18 column (250 × 4.6 mm, 5.0 μm) at 254 nm wavelength. The calibration curve was found to be linear in the range of 2–500 μg/ml. The stated method was then validated in terms of specificity, linearity, precision, and accuracy. Additionally, the proposed method reduced the duration of the analysis.  相似文献

Background and aims

This study compared the accuracy of the FreeStyle Libre (Abbott, Alameda, CA) and Dexcom G4 Platinum (DG4P, Dexcom, San Diego, CA) CGM sensors.

Methods and results

Twenty-two adults with type 1 diabetes wore the two sensors simultaneously for 2 weeks. Libre was used according to manufacturer-specified lifetime (MSL); DG4P was used 7 days beyond MSL. At a clinical research center (CRC), subjects were randomized to receive the same breakfast with standard insulin bolus (standard) or a delayed and increased (delayed & increased) bolus to induce large glucose swings during weeks 1 and 2; venous glucose was checked every 5–15 min for 6 h. Subjects performed ≥4 reference fingersticks/day at home. Accuracy was assessed by differences in mean absolute relative difference (%MARD) in glucose levels compared with fingerstick test (home use) and YSI reference (CRC). During home-stay the Libre MARD was 13.7 ± 3.6% and the DG4P MARD 12.9 ± 2.5% (difference not significant [NS]). With both systems MARD increased during hypoglycaemia and decreased during hyperglycaemia, without significant difference between sensors. In the euglycaemic range MARD was smaller with DG4P [12.0 ± 2.4% vs 14.0 ± 3.6%, p = 0.026]. MARD increased in both sensors following delayed & increased vs. standard bolus (Libre: 14.9 ± 5.5% vs. 10.9 ± 4.1%, p = 0.008; DG4P: 18.1 ± 8.1% vs. 13.1 ± 4.6%, p = 0.026); between-sensor differences were not significant (p = 0.062). Libre was more accurate during moderate and rapid glucose changes.

Conclusions

DG4P and Libre performed similarly up to 7 days beyond DG4P MSL. Both sensors performed less well during hypoglycaemia but Libre was more accurate during glucose swings.

目的 研究胶体金免疫层析试纸条检测鼠疫F1抗原的敏感性和特异性.方法通过鼠疫F1抗原单克隆抗体(F1MAb)捕捉F1抗原的胶体金免疫层析(GICA)试纸条,检测308只强毒鼠疫耶尔森菌(鼠疫菌)感染鼠脏器标本和225份对照鼠(达乌尔黄鼠217只,小白鼠5只,豚鼠3只)标本,同时用鼠疫反向间接血球凝集试验(RIHA)微量法和细菌培养做平行检测.结果 225只对照鼠鼠疫细菌学检验及GICA和RIHA的F1抗原检测均为阴性,GICA和RIHA在1×108 cfu/ml水平上未发现与近缘假结核耶尔森菌有交叉反应.GICA、RIHA对鼠疫菌检测灵敏性为2.5×105、2.0×105cfu/ml;检测F1抗原灵敏性为1、10 μg/L.GICA与细菌学检验符合率为97.94%(522/533),Kappa值=0.959,阳性检出率组间比较,差异无统计学意义(x2=0.36,P>0.05);与RIHA符合率为97.94%(522/533),Kappa值=0.959,阳性检出率组间比较差异有统计学意义(x2=9.09,P<0.05).308只实验鼠样本细菌培养阳性284只,284份细菌培养阳性标本中,GICA有280份检测阳性,阳性率为98.59%,高于RIHA[阳性率为96.13%(522/533)],两组比较差异有统计学意义(x2=5.14;P<0.05).GICA的敏感度为98.59%(280/284),特异度为97.19%(242/249),阳性预测值为97.56%(280/287),阴性预测值为98.37%(242/246),Youden指数为0.9578.结论 GICA检测鼠疫F1抗原敏感特异,快速简便,是鼠疫早期快速诊断中有应用价值的检测技术.  相似文献

目的 应用蛋白质组学技术筛选并鉴定肝癌患者血清中的差异蛋白质.方法 采用表面增强激光解析及电离飞行时间质谱技术,对33例肝癌患者和33例正常对照的血清蛋白质进行了检测分析,筛选肝癌的差异蛋白;用等电点沉淀法、十二烷基硫酸钠-聚丙烯酰胺凝胶电泳及高效液相色谱串联质谱等一系列方法分离鉴定差异蛋白质.肝癌组与正常组蛋白质峰差异比较采用两样本t检验. 结果 肝癌组与正常人组血清蛋白质图谱在相对分子质量为2 000~10 000范围内,有65个蛋白质峰差异有统计学意义(P<0.05),由此组建的诊断模型灵敏度为100%,特异度为96.97%其中包括8706.5 M/Z和8579.2M/Z(t值分别为2.562和2.783,P值均<0.05),经分离鉴定其为阿朴脂蛋白AⅡ. 结论 阿朴脂蛋白AⅡ是肝癌的差异蛋白之一,可能与肝癌的发生有关.  相似文献

目的 探讨人MTB标准株H37Rv与牛分枝杆菌标准株卡介苗株的亚细胞蛋白质组学差异.方法 应用密度梯度离心法分离H37Rv和卡介苗株的细胞壁、细胞膜和细胞质蛋白,利用二维液相色谱分离技术进一步获得H37Rv和卡介苗株亚细胞蛋白质组,采用酶联免疫吸附试验分别检测H37Rv和卡介苗各组分与结核病患者和健康人各5例血清的免疫学反应.组间比较采用t检验.结果 共获得26个特异性引发免疫反应的H37Rv菌株蛋白组分.细胞膜组分M3Fr18与结核病患者的血清免疫学反应的吸收光度(A450值)为(721±3)×10-3,明显高于健康人[(356±6)×10-3],也明显高于卡介苗株细胞膜相应组分与健康人的血清免疫学反应强度[(414±7)×10-3],差异均有统计学意义(t值分别为1.852和1.037,均P<0.01).细胞膜组分MI0Fr21与结核病患者的血清免疫学反应的A450值为(954±6)×10-3,明显高于健康人[(415±6)×10-3],也明显高于卡介苗株细胞膜相应组分与健康人的血清免疫学反应强度[(315±4)×10-3],差异均有统计学意义(t值分别为2.113和2.550,均P<0.01).结论 二维液相色谱和酶联免疫吸附试验联合应用,有助于检测人MTB感染的相关抗原蛋白.M3Fr18和M10Fr21蛋白质组分可引起结核病患者的特异性免疫反应,提示其可能是人MTB感染的特异抗原.
Abstract:
Objective To compare the proteome of Mycobacterium tuberculosis (MTB) H37 Rv strain with Bovine mycobacterium (Bacillus Calmette-Guerin, BCG) strain at the sub-cellular level. Methods Proteins of the cell wall, membrane and cytolymph of H37 Rv and BCG were extracted by density gradient centrifugation. Sub-cellular proteome of H37 Rv and BCG were analyzed using 2-dimensional liquid chromatography. The immunity reactions of H37 Rv fractions with sera from patients ( n = 5 ) and healthy controls ( n = 5 ), as well as BCG fractions with sera from healthy controls ( n = 5 ) were analyzed by ELISA.Data was analyzed by t test. Results Twenty-six fractions of H37 Rv were found to elicit specific antibody response. Fraction M3Fr18 of H37 Rv reacted with sera from patients. The A450 [ (721 ± 3 ) × 10-3] was higher than that with sera from healthy controls [ ( 356 ± 6) × 10 - 3 ] , as well as the A450 of the corresponding fractions of BCG with sera from healthy controls [ (414 ± 7) × 10-3 ]. The differences between the patient group and the 2 healthy control groups were significant ( t = 1. 852 and 1. 037, all P < 0. 01 ). Moreover,fraction M10Fr21 of H37Rv reacted with sera from the patients. The A450[ (954 ±6) × 10 -3 ] was higher than that with sera from the healthy controls [ (415 ± 6 ) × 10-3 ], as well as the A450 of the corresponding fractions of BCG with sera from the healthy controls [ ( 315 ± 4 ) × 10-3 ]. The differences between the patient group and the 2 healthy control groups were significant ( t = 2. 113 and 2. 550, all P < 0. 01 ).Conclusions The combination of 2-dimensional liquid chromatography and immunology technology is useful in finding antigens associated with MTB infection. Fractions M3Fr18 and M10Fr21 can elicit specific immune reaction among MTB patients, suggesting that they may be specific antigens of MTB infection.  相似文献

本文用超滤和肝素亲和层析法对培养的兔主动脉平滑肌细胞的无血清条件培养基进行纯化。用3H-TdR掺入细胞DNA法检测层析所得各组分对NIH3T3纤维母细胞的致有丝分裂活性。用NaDodSO4-聚丙烯酰胺凝胶电泳及等电聚焦电泳检测其分子量及等电点。结果表明,该条件培养基经肝素亲和层析,被1.0~1.6mol·L'NaCl洗脱下来的蛋白质成分对3T3细胞有致有丝分裂活性,其分子量范围为23.0~26.9kDa,等电点约为4.6。这些生物化学特性不同于PDGF、IGF及FGF,提示这种致有丝分裂蛋白质可能是一种独立的生长因子。  相似文献

采用毛细管柱气相色谱技术分离出结核杆菌特征组份并鉴定为结核杆菌硬脂酸。对59例脑脊液菌阴结核性脑膜炎患者与122名非结核性脑膜炎对照组进行分析。实验结果显示结核性脑膜炎组检出结核杆菌硬脂酸44例,对照组中检出5例,敏感性及特异性分别为74.6%和95.9%。表明结核杆菌硬脂酸的检测有助于菌阴结核性脑膜炎的早期诊断。  相似文献

AIM: To analyze the clinical characteristics of Chinese hereditary nonpolyposis colorectal cancer (HNPCC) families and to screen the germline mutations of human mismatch repair genes hMLH1 and hMSH2 in the probands.METHODS: Thirty-one independent Chinese HNPCC families were collected in Zhejiang Province. All of them met Chinese HNPCC criteria. Clinical data about patient gender, site of colorectal cancer, age of onset, history of multiple colorectal cancer, associated extracolonic cancer were recorded. PCR and denaturing high performance liquid chromatography (DHPLC) were employed to screen the mutations. Sequencing analysis was used to find out the exact mutation site and characteristics of the samples showing abnormal DHPLC profiles.RESULTS: One hundred and thirty-six malignant neoplasms were found in 107 patients including 14 multiple cancers. One hundred and six of the 136 neoplasms (77.9%) were diagnosed as colorectal cancer, with an average age of onset at 48.57 ± 29.00 years. Gastric cancer was the most common extracolonic cancer (10.3%) in these families. Twenty-three different sequence variations in hMLH1 and hMSH2 genes were detected in these 17 families. Fifteen sequence variations were located in the exons, including 5 SNPs, 3 silent mutations, 3 missense mutations, 2 nonsense mutations and 2 frameshift mutations. The latter seven mutations seemed to be pathogenic.CONCLUSION: Germline mutations of hMLH1 and hMSH2 genes are identified in about one-third HNPCC kindreds fulfilling Chinese HNPCC criteria. Chinese HNPCC families have some particular clinical characteristics, such as a left-sided predominance, less synchronous or metachronous colorectal cancer, and frequent occurrence of gastric cancer.  相似文献
