Effect of interferon on two human choriocarcinoma-derived cell lines

Two cell lines derived from human choriocarcinomas (HCCM-5 and BeWo) are resistant to several biological effects of human interferon such as inhibition of VSV multiplication and inhibition of cell growth, but they develop a normal antiviral activity against EMCV. Nevertheless, in both cell lines, 2-5A synthetase and protein kinase are induced by IFN. 2-5A-dependent endonuclease can be measured by two independent methods and 2-5A itself is detected at least in poly(rI):poly(rC)- and IFN-treated BeWo cells. This is another example of two cell lines that are partially, with respect to the antiviral effect toward VSV, and totally, with respect to the anticellular effect, refractory to IFN treatment, although all the known elements of the 2-5A system are present.     

Sister chromatid exchanges in patients with retinoblastoma

Spontaneous and mitomycin C(MMC)-induced sisyer chromatid exchanges were studied in 11 patients with retinoblastoma and 7 normal controls. Spontaneous rates were similar in patients and in controls. The MMC-induced rate was found to be significantly higher in bilaterally affected patients than in controls. It is suggested that this increase may be due to a DNA repair deficiency. However, it is not possible to clarify wether this abnormality is associated with the retinoblastoma gene or with another factor acting on the degree of expressivity of the disease in gene carriers.     

Preferential involvement of chromosome 1q in a primary breast carcinoma

The cytogenetic findings by G- and C-banding in a primary breast carcinoma are reported. The tumor is characterized by a high modal number of chromosomes (79) and 14 markers of ten different origins. Chromosome #1 is more frequently involved in structural and numerical aberrations than is any other chromosome; nine copies of 1q were present in each cell examined. This observation supports the hypothesis that duplication of chromosome 1q may play an important role in the progression of malignant tumors.     

Relating cistrons and functions in bacteriophage PM2

An approach to correlate functions with cistrons in bacteriophage PM2 is presented. Two of the temperature-sensitive mutants obtained differed from the wild type in heat sensitivity and four differed in host range. Comparison of the electrophoretic patterns of DNA from cells infected at the restrictive temperature with those obtained in wildtype infection revealed that viral DNA bands were absent with three additional mutants. Complementation analysis assigned the four host range mutants to cistron I and the three mutants defective in DNA synthesis to cistron IV. Recombination frequencies were used to locate cistrons III and IV on the partial genetic map of PM2.

通过对母料载体树脂的筛选,无机填料的选择,添加性能优良的耐辐射助剂,制成了具有耐辐射性能、加工性能良好的母料,该母料的制备工艺简单。本文所述的填充级HDPE专用料是将HDPE与填充母料经过简单掺混制成的。经测试专用料满足国家医药标准YY 0114—93《医用输液、输血、注射器聚乙烯专用料》的要求。制品厂试验表明:该填充级的HDPE专用料加工性能良好,用该填充级HDPE专用料注射成型的注射器芯杆装配成一次性注射器满足国家专业标准ZBC 31009—87《一次性无菌注射器》的要求。该填充母料的加入可大大降低材料的成本,100份HDPE 7006A可添加67份填充母料,该填充级专用料的价格比HDPE树脂降低约1500元/吨,因此该填充级HDPE专用料的推广和使用具有良好的经济效益。     

Does Low FEV1 in Addition to Fixed Ratio and/or Lower Limit of Normal of FEV1/FVC Improve Prediction of Mortality in COPD? The NHANES-III-linked-mortality Cohort

Purpose

There is presently an ongoing debate on the relative merits of suggested criteria for spirometric airway obstruction. This study tests the null hypothesis that no superiority exists with the use of fixed ratio (FR) of forced expiratory volume in the first second (FEV1)/forced vital capacity (FVC) < 0.7 versus less than lower limit predicted (LLN) criteria with or without FEV1 <80% predicted in regards to future mortality.

Mapping of the influenza virus genome II. Identification of the P1, P2, and P3 genes

Polyacrylamide gel electrophoresis of the RNAs of influenza A viruses was performed under conditions in which each of the eight RNA segments of influenza A/PR/8/34 (H0N1) virus migrates differently from the equivalent segments of influenza A/Hong Kong/8/68 (H3N2) virus. As reported previously [Palese, P. and Schulman, J. L. Proc. Nat. Acad. Sci. USA73, 2142–2146 (1976)], analysis of RNA patterns of recombinant viruses derived from these two parents permitted the identification of the fourth RNA segment (the slowest-moving RNA segment is counted as #1) of each virus as the gene coding for hemagglutinin, and the fifth segment of Hong Kong virus and the sixth segment of PR8 virus as the genes for the respective neuraminidases. Three more genes have been identified by correlating migration patterns of RNAs with protein patterns of recombinant viruses on gradient polyacrylamide gels. The slowest-moving band (RNA 1) is the gene coding for the third largest influenza virus protein (P3). RNA segment 2 codes for the largest protein (Pl), and the P2 protein is coded for by RNA segment 3.     

Distribution of radioactive cortisol in the tissues and media of the eye

Incorporation of cortisol-³H and the dynamics of its accumulation in the tissues and media of the eye (sclera, ciliary body, cornea, iris, capsule of the lens, aqueous humor, vitreous body) were investigated. The intensity of incorporation of cortisol into the tissues and media of the eye and also the rate of its elimination from them were shown to differ. The sclera, cornea, ciliary body, and the capsule of the lens were shown to be target tissues for cortisol.Moscow Helmholtz Scientific-Research Institute of Eye Diseases. Purification and characterization of potyvirus helper component

Helper component (HC) was purified from tobacco vein mottling (TVMV)- and potato virus Y (PVY)- infected tobacco plants by sucrose gradient fractionation followed by affinity chromatography on oligo(dT)-cellulose and by gel electrophoresis. The subunit apparent molecular weights (M(r)) of the purified HCs were 53,000 (53K) and 58K for TVMV and PVY, respectively. Antisera to these purified polypeptides specifically blocked the activity of the homologous HC, as determined by aphid transmission assays, and specifically precipitated 75K products of the cell-free translation of the homologous RNA. The molecular weight of undissociated, biologically active TVMV or PVY HC, as determined by high-pressure liquid chromatography (HPLC)-gel permeation chromatography was found to be between 100K and 150K, suggesting that the active molecule is a dimer.
