BACKGROUND: Graft-vs.-host disease (GVHD) is the major cause of morbidity and mortality in patients undergoing allogeneic Bone Marrow Transplantation (BMT). The aim of our study was to identify the most relevant histological features for diagnosis of chronic Graft-vs.-Host Disease (cGVHD) in oral mucosa and minor salivary glands of 25 patients, as well as to evaluate the immunophenotype of the inflammatory cells. METHODS: Sixteen patients that were submitted to allogeneic BMT but did not present cGVHD were selected as a control group. The sections were studied on H & E and CD68, CD45, CD4, CD8, CD20 staining. RESULTS: The most frequent histologic findings in oral mucosa at the day of diagnosis of cGVHD were: hydropic degeneration of the basal layer of the epithelium, apoptotic bodies, lymphocytic infiltration, and focal or total cleavage between the epithelial and connective tissue. In the labial salivary glands (LSG), lymphocytic infiltration, acinar loss and fibrosis were the main alterations. Cytotoxic CD8-T cells and macrophages were predominant both in the epithelium and connective tissue, as well as in minor salivary glands. CONCLUSIONS: Histological features were useful in the diagnosis of oral cGVHD. It is suggested that CD8-T cells and macrophages play important role in the pathogenesis of the disease. 相似文献

Previously, we reported that allogeneic skin grafts were rapidly rejected by CD28 and CD40 ligand double deficient mice mediated by CD8+ T cells. These results indicated that some elements in addition to CD28- and CD40-mediated costimulation provide stimulatory signals for the activation of donor-specific CD8+ T cells. In this report, we investigated the role of inflammation associated with transplantation on costimulation-independent priming of CD8+ T cell during graft rejection. B6 RAG1 KO mice were transplanted with BALB/c-skin and adoptively transferred with syngeneic CD8+ T cells the same day or 50 days after transplantation. When blockade of CD28- and CD40-mediated costimulation failed to prevent acute rejection of freshly transplanted skin grafts, it efficiently delayed rejection of well-healed skin grafts. These results showed that factors associated with transplantation have essential roles in inducing costimulation blockade-resistant allograft rejection. Costimulation blockade failed to prevent acute graft-infiltration of NK cells and increasing expression of intragraft IL-12 and IL-15. These factors may trigger the graft-infiltration and priming of CD8+ T cells to induce costimulation blockade-resistant allograft rejection. 相似文献

目的探讨蛋白酶激活受体1(PAR1)激动肽和凝血酶对人肺上皮细胞白细胞介索-8(IL-8)分泌的影响。方法人肺上皮细胞系A549细胞分别接种于12孔培养板各孔内,并分别用不同浓度的PAR1激动肽SFLLR和反PAR1激动肽RLLFS以及不同浓度的凝血酶和/或凝血酶抑制剂水蛭索进行刺激,刺激时间为2和16h。用ELISA方法检测上清液中的IL-8水平。结果经过16h的培养,SFLLR可引起浓度相关性IL-8的释放增加,增加到300μmol/L时诱导IL-8的释放量比基础分泌量增加了近16倍,RLLFS不能引起IL-8的释放增加。凝血酶也可引起浓度相关性IL-8释放.凝血酶在浓度1kU/L时就可引起IL-8释放量增加,10kU/L时诱导IL-8释放量达高峰,为基础分泌量的7.5倍。水蛭索可以抑制凝血酶对IL-8的释放作用。时间相关曲线表明,PAR1介导的IL-8释放从2h起即可引起增加,16h达高峰。结论PAR1激动肽和凝血酶可促进人肺上皮细胞分泌IL-8,PAR1拮抗剂和凝血酶抑制剂可能具有抗炎作用。 相似文献

用AR CM MIC阳离子测定系统,测量单个细胞内游离钙浓度([Ca2+]i),研究8-(N,N-二乙胺)-n-辛基 3,4,5-三甲氧基苯甲酸酯(TMB-8)对培养乳牛基底动脉平滑肌[Ca2+]i的作用。在细胞外钙浓度为1.3mmol·L-1时,TMB-8(30μmol·L-1)可明显抑制BHQ,NE及KCl引起[Ca2+]i的升高。在细胞外钙为零+EGTA 0.1mmol·L-1时,TMB-8(10,30及100μmol·L-1)可浓度依赖性地降低静息[Ca2+]i,TMB-8(30μmol·L-1)可几乎完全阻断BHQ及NE引起[Ca2+]i的增加。研究表明TMB-8降低培养乳牛基底动脉平滑肌[Ca2+]i的机制,主要是抑制肌浆网Ca2+的释放,或增加肌浆网对Ca2+的摄入,并由此间接地抑制细胞外钙的内流。 相似文献

目的:研究重组人内皮细胞衍生的白细胞介素-8(IL8)对失血性休克的作用.方法:大鼠股动脉放血至MABP532kPa,维持90min,复制晚期失血性休克模型.输血后,静脉注射IL8250μg·kg-1.放免法测定血浆ET1和6KPGF1α含量.结果:给予IL8后,MABP显著提高,休克状态改善,2h存活率相应提高;休克晚期血浆ET1水平比正常明显升高(21±4vs82±18ng·L-1,P<001),血浆6KPGF1α含量明显降低(107±12vs157±11ng·L-1,P<001).IL8显著降低血浆ET1水平(10±4ng·L-1,P<001),提高血浆6KPGF1α含量(368±16ng·L-1,P<001).结论:IL8具有较好的抗休克作用. 相似文献

A gene similar to lef-8 of the Autographa californica (Ac) nucleopolyhedrovirus (MNPV) was identified in the Spodoptera littoralis (Spli) MNPV. The SpliMNPV lef-8-like gene was localized on the genomic map between 26.9 and 29 map units and is flanked by a chitinase gene and p47 gene.
This gene arrangement differs from that of similar genes in the AcMNPV genome, where the lef-8 gene is located about 62 kbp from the chitinase gene and about 7 kbp from the p47 gene. Sequence analyses of the lef-8 gene revealed an ORF of 2730 nucleotides, predicted to encode a protein with M
r 104876. The putative protein is 60.9% identical to the AcMNPV LEF-8 and contains an identical sequence of a conserved motif
of DNA-dependent RNA polymerases. Sequences downstream of the lef-8 gene contain two sequence repeats which resemble a repeated motif of the SpliMNPV enhancer element and other repetitive sequences from the viral genome.

Trisomy 8 in acute promyelocytic leukaemia: an interphase study by fluorescence in situ hybridization 总被引:1,自引:0,他引:1
Summary. Acute promyelocytic leukaemia (APL) is characterized by t(15;17)(q24;q21). Trisomy 8 is the commonest accompanying karyotypic aberration. We investigated 14 APL patients for trisomy 8 using fluorescence in situ hybridization (FISH). Conventional cytogenetic analysis showed trisomy 8 in two of nine successfully karyotyped cases. With FISH, a possible third case showing a subclone (1-2-5%) with trisomy 8 was found. The trisomy 8 clone size defined by karyotyping and FISH was concordant in one case and discordant in another, in which trisomy 8 was found in 100% of metaphases but only in 48% of leukaemic prbmyelocytes by FISH. Therefore trisomy 8 was mosaic in all the cases, suggesting that it had arisen from clonal evolution.
