Comparison of arachidonic acid metabolism by bovine coronary artery endothelial cells, bovine aortic endothelial cells and human umbilical endothelial cells indicated potentially important differences in relative amounts of the different prostaglandins produced. Bovine coronary endothelial cells converted 14C-arachidonic acid to radioactive 6-keto PGF1 (the stable metabolite of PGI2) and to a lesser extent PGE2. Bovine aortic cells synthesized 6-keto PGF1 and 6,15-diketo PGF1 as the major products. PGE2, 6-keto PGE1, PGE2, and PGD2 were minor metabolites. By comparison, endothelia cells isolated from human umbilical artery or vein formed mainly 6-keto PGF1 and substantial amounts of PGF2, PGE2 and PGD2. Basal concentrations of 6-keto PGF1 were two-fold higher in bovine coronary cells than in bovine aortic endothelial cells, but seven-fold less than in endothelial cells cultured from human umbilical vessels. Histamine, bradykinin and thrombin stimulated PGI2 synthesis in both coronary endothelial cells and human umbilical cells, but only bradykinin stimulated PGI2 synthesis in bovine aortic cells. This comparative study indicates that endothelial cells vary in the metabolites of arachidonic acid that they produce depending upon the vascular origin of the cells. In this study, we employed 1α-hydroxylase knockout (1α-(OH)ase(-/-) ) mice to investigate the influence of 1,25-dihydroxy vitamin D(3) (1,25-(OH)(2) D(3) ) deficiency on the adult neurogenesis in the hippocampal dentate gyrus (DG). The numbers of both 24-hr-old BrdU(+) cells and proliferating cell nuclear antigen positive cells in 8-week-old 1α-(OH)ase(-/-) mice increased approximately twofold compared with wild-type littermates. In contrast, the numbers of 7- and 28-day-old BrdU(+) cells in 1α-(OH)ase(-/-) mice decreased by 50% compared with wild-type mice, while the proportion of BrdU(+) /NeuN(+) cells in BrdU(+) population showed no difference between 1α-(OH)ase(-/-) and wild-type mice. Apoptotic cells in the subgranular zone (SGZ) of DG markedly increased in 1α-(OH)ase(-/-) mice. Replenishment of 1,25-(OH)(2) D(3) , but not correction of serum calcium and phosphorus levels, completely prevented changes in the neurogenesis in 1α-(OH)ase(-/-) mice. The absence of 1,25-(OH)(2) D(3) led to an increase in the expression of L-type voltage-gated calcium channel (L-VGCC) and a decrease in the nerve growth factor (NGF) mRNA level. Treatment with the L-VGCC inhibitor nifedipine blocked the increased cell proliferations by 1,25-(OH)(2) D(3) deficiency. Severe shot peening (SSP) was used on additive manufactured 316L by laser powder bed fusion. The effect of the post processing on the surface features of the material was analyzed through residual stress measurements, tensile testing, hardness-depth profiles, and fatigue testing by flexural bending. The results showed that SSP can be utilized to form residual stresses up to −400 MPa 200 μm below the surface. At the same time, a clear improvement on the surface hardness was achieved from 275 HV to near 650 HV. These together resulted in a clear improvement on material strength which was recorded at 10% improvement in ultimate tensile strength. Most significantly, the fatigue limit of the material was tripled from 200 MPa to over 600 MPa and the overall fatigue strength raised similarly from a low to high cycle regime. 相似文献

通过对HPV L1序列进行比对,发现HPV L1 C-末端存在长30个氨基酸残基的保守序列短肽;出于检测短肽是否可以诱生HPV多种型别交叉抗体的目的,将该序列短肽加弗氏佐剂用于日本大耳白兔和BALB/c小鼠免疫,然后用ELISA方法检测此免疫动物血清及其分泌物中的IgG抗体滴度,发现此免疫动物体内已诱生出高滴度的血清IgG抗体(>1∶20000);再用ELISA、免疫组织化学和Western blot的方法对此诱生血清抗体与HPV阳性宫颈癌细胞株的反应情况进行检测,发现这些短肽抗血清可与16、18型HPV L1很好地进行反应,其对照组呈现阴性。这一研究结果表明短肽可以诱生HPV多种型别交叉抗体。它对后续研发HPV L1广谱疫苗或检测试剂盒具有重要意义。 相似文献

Oxaliplatin (Oxa)-associated adverse side effects have considerably limited the clinical use of the drug in colon cancer therapy. Mutant p53 has diverse mutational profiles in colon cancer, and it influences the potencies of various chemotherapeutic drugs, including Oxa. Thus, it would be highly beneficial to identify an alternative therapeutic strategy that not only reduces the toxicity of Oxa, but also exerts a synergistic effect against colon cancers, regardless of their p53 profiles. The present study was aimed at preparing and optimizing Teucrium polium L. essential oil nanoemulsion (TPO-NANO) and investigating its effect on the sensitivity of colon cancer cells with differences in p53 status (HCT116 wild-type and HT-29 mutant-type) to Oxa. The viability of treated cells was determined and the combination index (CI) was calculated. Morphological changes were determined under inverted microscopy, while percentage apoptosis was assayed using flow cytometry. Intracellular ROS and the protein levels of p53 and Bax were measured. The colony-forming potential of treated cells was determined using colony assay. The size of TPO-NANO was markedly increased from 12.90 ± 0.04 nm to 14.47 ± 0.53 nm after loading Oxa (p ≤ 0.05). The combination (Oxa + TPO-NANO) produced a synergetic effect in HCT116 and HT-29, with CI of 0.94 and 0.88, respectively. Microscopic examination and flow cytometric analysis revealed that cells treated with Oxa + TPO-NANO had a higher percentage of apoptosis than cells exposed to monotherapy. Cumulatively, Oxa exerted an apoptotic effect on wild or mutant p53 colon cancer cells when combined with TPO-NANO, through a mechanism involving ROS-mediated mitochondrial apoptosis. 相似文献

目的建立测定旱芹非淀粉多糖中各单糖组分含量的柱前衍生化GC法。方法非淀粉多糖经衍生化生成糖腈乙酯,采用GC法进行分析。色谱柱:DB-17柱(30 m×0.25 mm,0.25μm);柱温:程序升温;载气:N2;进样温度:270℃;进样方式:不分流;检测器温度:270℃;进样量:1.0μL。结果水不溶性非淀粉多糖(insoluble non-starch polysaccharides,INSP)水解后得到7种单糖,分别是:L-(+)-鼠李糖、L-(+)-阿拉伯糖、D-(+)-岩藻糖、D-(+)-木糖、D-(+)-甘露糖、D-(+)-葡萄糖和D-(+)-半乳糖;水溶性非淀粉多糖(soluble non-starch polysaccharides,SNSP)水解后得到6种单糖,除无D-(+)-岩藻糖外,其余均同INSP水解后单糖。结论本方法准确度高,重现性好,可用于测定旱芹水溶性和水不溶性非淀粉多糖中各单糖组分的含量。 相似文献

目的 为了增强青蒿的特异药理活性,探讨人β-趋化因子RANTES基因在青蒿细胞中表达的可能性。方法 将克隆的RANTES基因经Ti质粒衍生的双元表达载体pRPKII导入根癌农杆菌LBA4404菌株,通过叶盘共培养法已获得一批表现卡那霉素抗性的转基因青蒿植株。结果 PCR和Southern印迹杂交分析表明,RANTES基因已导入并整合到青蒿基因组中;RT-PCR、Northern印迹、Western印迹杂交结果证实,RANTES基因已在转基因青蒿植株中成功表达。结论 首次报道了RANTES基因在转基因青蒿植株中的表达,为基因工程改造青蒿打下了良好的基础。 相似文献

Dietary intake of fermented soymilk is associated with hypotensive effects, but the mechanisms involved have not been fully elucidated. We investigated the anti-hypertensive effects of soymilk fermented by L. rhamnosus AC1 on DOCA-salt hypertension from the point of view of oxidative stress, inflammatory response and alteration of the gut microbiome. The antioxidant assays in vitro indicated the ethanol extract (EE) of L. rhamnosus AC1 fermented soymilk showed better antioxidative effects than the water extract (WE). Those extracts displayed a hypotensive effect using a tail-cuff approach to measuring blood pressure and improved nitric oxide (NO), angiotensin II (Ang II), tumor necrosis factor-α (TNF-α) and interleukin factor-6 (IL-6) on DOCA-salt hypertensive rats. Furthermore, cardiac and renal fibrosis were attenuated by those extracts. The gut microbiota analysis revealed that they significantly reduced the abundance of phylum Proteobacteria, its family Enterobacteriaceae and genus Escherichia-Shigella. Moreover, metabolomic profiling revealed several potential gut microbiota-related metabolites which appeared to involve in the development and recovery of hypertension. In conclusion, fermented soymilk is a promising nutritional intervention strategy to improve hypertension via reducing inflammation and reverting dysbiotic microbiota. 相似文献
