为探讨金属硫蛋白(MT)对豚鼠乳头肌缺血再灌注损伤所致心律失常的影响,利用标准玻璃微电极技术,采用缺氧及复氧豚鼠乳头肌模型,模拟体内缺血再灌注损伤,观察不同浓度MT对豚鼠乳头肌电生理特性的影响。结果显示低浓度的MT(0.002mmol/L)对正常及缺氧和复氧豚鼠乳头肌的动作电位(AP)有关参数及自律性均无影响;中等浓度的MT(0.02mmol/L)仅使正常乳头肌的AP复极达50%时程(APD50)缩短24%(P<0.05),但使缺氧乳头肌的AP复极达20%时程(APD20)、APD50和AP复极达90%时程(APD90)分别缩短68%、56%和43%(P均<0.01),并使静息电位(RP)、AP幅值(APA)和0相最大上升速率(Vmax)分别增加30%、30%和45%(P均<0.01);高浓度的MT(0.1mmol/L)使正常豚鼠乳头肌的APD20、APD50和APD90分别缩短57%、54%和50%(P均<0.01),并且RP、APA及Vmax分别下降22%(P<0.05)、28%(P<0.01)和29%(P<0.05),而使缺氧豚鼠乳头肌的APD20、APD50和APD90分别延长92%、78%和50%(P均<0.01),对RP、APA及Vmax无明显影响。在复氧期间,0.02mmol/L的MT可使自律性的发生率从77.8%降至55.6%(P<0.05);而0.1mmol/L的MT则使自律性的发生率从77.8%降至22.2%(P  相似文献

Objective. To determine in a prospective study the normal MRI morphology of the sacroiliac joints (SIJs) in relation to age and sex during adolescence. Design and patients. A total of 98 children (63 boys, mean age 12.7±2.8 years; 35 girls, mean age 13.7±2.3 years), ranging in age from 8 to 17 years, with juvenile chronic arthritis (JCA) but without signs of sacroiliitis fulfilled the study prerequisites (no back pain and no pathologic changes of the SIJs on physical examination before MRI in a 1.5-year follow-up). An additional eight HLA-B27-negative boys and eight HLA-B27-negative girls without arthritis served as controls. The MRI protocol comprised a T1-weighted SE sequence, an opposed-phase T2*-weighted GE sequence, and a dynamic contrast-enhanced study in single-section technique. Results. Noncontrast MRI permitted differentiation of “open” from ossified segmental and lateral apophyses of the sacral wings, with a significant difference in age (P <0.05) between children with open and ossified apophyses. Ossification of the apophyses of the sacral wings was seen significantly earlier (P <0.05) in girls than in boys. Girls also had a significantly higher incidence of transitional lumbosacral vertebrae, pelvic asymmetries, and accessory joints. In the contrast-enhanced opposed-phase MRI study, normal cartilage of the SIJs showed no contrast enhancement whereas the joint capsule showed a moderate enhancement. Conclusion. There are significant age- and sex-related differences in the normal MRI morphology of juvenile SIJs. Our findings might serve as a standard of comparison for the evaluation of pathologic changes – in particular for the early identification of juvenile sacroiliitis.  相似文献

Bone mineral “density” (BMD) measured by dual-energy X-ray absorptiometry (DEXA) does not represent the volumetric density (grams per cubic centimeter), but rather the areal density (grams per square centimeter). This distinction is important during growth. The purpose of this study was to measure vertebral dimensions in cadavers of young pigtail macaques (Macaca nemestrina), and to derive equations to predict the volumetric bone density from noninvasive measurements. We measured the areal bone density by DEXA, vertebral volume by underwater weighing, mineral content by ashing, dimensions of lumbar vertebrae by calipers, and dimensions of vertebrae by radiography. Somatometric measurements of the female lumbar vertebral bodies showed that the shape changed during growth. The bone mineral content from the densitometer correlated significantly with the ash weight (r = 0.99, error 8.7%). The correlation coefficient between the volumetric bone mineral density and areal BMD measurement was significant (r = 0.68, p < 0.0001) with a 9.5% error; this improved significantly to 0.82 (7.2% error) when the BMD was divided by the vertebral depth from the radiograph. Areal BMD showed a strong correlation with age (r = 0.82, p < 0.0001), with an average increase of 7.4%/year. In contrast, volumetric mineral density showed a weak relationship with age (r = 0.43, p < 0.01), for an average increase of 1.5%/year. When studying bone mineral density during growth, the differences between volumetric and areal bone mineral density should be taken into consideration. (  相似文献

The effects of oestrogen administration on 1 h post‐exercise cardiac muscle myeloperoxidase (MPO) and calpain activities were determined in female rats. Rats were ovariectomized and implanted for 2 weeks with either oestrogen (25 mg 17‐oestradiol) or placebo pellets or left with ovaries intact. Rats were then run for 1 h at 21 m min–1, 12% grade, killed 1 h post‐exercise and cardiac muscle and blood samples were removed. Control animals from each group were killed without prior exercise. Serum oestrogen levels in the order of the highest to lowest were; ovariectomized oestrogen replaced rats > intact ovaries rats > ovariectomized placebo rats. Oestrogen induced significant (P < 0.05) elevations in cardiac MPO activity at rest and at 1 h post‐exercise in ovariectomized rats. No significant elevations in cardiac MPO activity were evident in placebo ovariectomized or normal ovary rats at rest or post‐exercise. Cardiac calpain activities were similar in all unexercised groups. Ovariectomized placebo and intact ovary rats had significantly (P < 0.05) elevated cardiac calpain activities 1 h post‐exercise while calpain activity was not significantly elevated in hearts from ovariectomized oestrogen rats. These results demonstrate that oestrogen supplementation in ovariectomized rats induces elevations in cardiac muscle MPO activities at rest and at 1 h post‐exercise. This is opposite to the effect of oestrogen in post‐exercise skeletal muscle and implies a greater neutrophil infiltration into cardiac muscle caused by oestrogen. This effect cannot be explained by changes in 1 h post‐exercise cardiac muscle calpain activity, the elevation of which was suppressed by oestrogen administration. Oestrogen influences cardiac calpain activity similarly to its effect in skeletal muscle. Thus, oestrogen administration to ovariectomized rats induces elevations in cardiac MPO activity while suppressing cardiac calpain activity.  相似文献

Abstract: This paper describes a device used to measure the isometric forces generated during electrical stimulation of the canine latissimus dorsi muscle in vivo with a preserved neurovascular supply. This device uses 2 strain gauge force sensors linked to a movable alignment frame to which the muscle is attached. The muscle length is controlled by the application of known weights to the system. The device has a frequency of response of 17.5 Hz and compliance of -0.1 mm N-1, and its experimental performance was tested in the anesthetized mongrel dog.  相似文献

Objective. To evaluate the impact of chronic inflammation on lipoprotein lipase (LPL) levels and triglyceride metabolism in patients with rheumatoid arthritis (RA). Design. Plasma levels of LPL activity and mass before and after heparin were determined in post-menopausal women with active RA and in controls. The results were related to lipid levels and inflammatory variables. The LPL activity and mass together with triglyceride levels were also measured before and 6 h after an oral fat load. Setting. The study was performed on in- and out-patients at a University Rheumatology clinic. The controls came from the same reference area. Subjects. Altogether 17 consecutive post-menopausal female patients with RA and 16 age and sex matched controls were enrolled for the initial determination of LPL. Fifteen of the patients and 15 of the controls agreed to take part in the fat load. Of these, one patient and one control were excluded. Main outcome measures. LPL determination: basal levels and post-heparin levels of LPL activity and mass. Correlations between LPL and blood lipids (cholesterol, triglycerides), lipoprotein levels (high density lipoprotein, HDL; low density lipoprotein, LDL), erythrocyte sedimentation rate (ESR) acute phase proteins (orosomucoid, haptoglobin, fibrinogen mass) and cytokines (tumour necrosis factor α, TNF-α; interleukin 1β, IL-1β; and interleukin-6, IL-6). Fat tolerance test: LPL activity, mass and triglyceride levels before and 6 h after a per oral fat load. Results. Pre-heparin LPL mass (P<0.01) and activity (P<0.01) were significantly lower in the rheumatoid patients. Pre-heparin LPL mass showed no correlation to the lipid levels, but an inverse correlation to several inflammatory parameters; it was significant for orosomucoid (rs=?0.63, P<0.05) and C-reactive protein (CRP) (rs=?0.54, P<0.05) and close to significant for haptoglobin (rs=?0.48, P=0.087) and IL-6 (rs=?0.52, P=0.061). Six hours after a lipid load the LPL activity and mass were significantly lower in RA (P<0.05 and P<0.01, respectively) but the triglyceride level was not significantly different compared to controls. Conclusion. An inverse relationship exists between inflammatory status and pre-heparin LPL mass. Pre-heparin LPL mass reflects mainly the inactive monomeric fraction of LPL. This has been shown to hinder the uptake of remnant lipoprotein particles through competition with lipoprotein bound dimeric LPL for the LDL receptor-related protein (LRP receptor) on hepatocytes and macrophages in culture. A decrease of the level of monomeric LPL in plasma may thus be beneficial for remnant catabolism. The same mechanism may on the other hand increase macrophage uptake of lipids. This may not affect global lipid metabolism but may be important in driving the atherosclerotic process in the vessel wall.  相似文献

Advances in combination chemotherapy, radiation therapy, surgery, and bone marrow transplantation have resulted in markedly improved survival rates for many children with cancer. Advancements in therapy, however, have led to new concerns, namely long-term consequences of effective treatments. Young adult and adult survivors of childhood cancer are at risk for a number of disorders related to therapy. Specifically, the young adult who has survived cancer, attendant treatments, and their complications is at risk for factors that can lead to suboptimal acquisition of peak bone mass. These factors include chronic illness, nutritional deficiencies, limited physical activity, and treatment with glucocorticoids, multiagent chemotherapy, and radiation. The long-term adverse effects of these therapies on endocrine systems, especially sex steroid and growth hormone deficiencies, are additional risk factors for some patients. After a brief review of the processes associated with acquisition of peak bone mass in the young adult, this article examines the impact of cancer and cancer therapy on bone mineral density in survivors of childhood cancer.  相似文献

目的 :观察大鼠骨骼肌钝挫伤后 ,不同干预方法对骨骼肌修复过程中肌电信号的影响 ,研究肌电图在骨骼肌损伤愈合过程中的评价作用。方法 :10 8只雄性SD大鼠 ,利用自制的重物坠落打击装置造成右侧腓肠肌钝挫伤。随机分成 3组 ,即胰岛素样生长因子 - 1(IGF - 1)组、中药组和自然愈合组 ,分别于损伤局部注射IGF - 1、黄芪丹参注射液、不作处理。于伤后第 2、5、7、10、14、2 1、2 8、35、5 6天观察并比较双侧腓肠肌的肌电图变化 ,记录纤颤电位和正尖波等自发活动 ,分析复合肌肉动作电位 (CMAP)并测量其潜伏期和波幅 ,观察伤后CMAP的变化。结果 :(1)各组大鼠均于伤后第 2~ 5天出现纤颤电位和正尖波 ,约至第 7~ 10天达到高峰 ,然后逐渐减少。以IGF - 1组最早消失 ,中药组次之。 (2 )各组大鼠的伤侧肌电图CMAP中的潜伏期逐渐缩短 ,波幅逐渐增高。以IGF - 1组最快 ,中药组次之 ,自然愈合组最慢。 (3)伤后第 8周 ,IGF - 1组和中药组大鼠损伤侧CMAP波幅接近正常 ,自然愈合组则显著低于其它两组。结论 :(1)肌电图检测证实 ,IGF - 1和黄芪丹参注射液均具有促进骨骼肌损伤愈合的作用 ,但以IGF - 1的作用更明显。 (2 )损伤肌肉局部肌电信号的变化 ,可以用来评价损伤骨骼肌的修复进程和愈合质量  相似文献

目的:对两例生长发育高峰期后的骨性下颌后缩患者成功矫正深覆盖后的面部软组织改变进行研究。方法:患者矫治前后的头颅侧位X线片进行描图和重叠,分析软组织的变化。结果:软组织主要变化有:下面高增加,下唇明显前移,上唇后移,颏唇沟变浅,唇肌紧张度改善。结论:固定矫治器配合Forsus矫治器和II类间牵引使两例严重深覆盖患者的侧貌得到了明显改善。但矫正效果主要为牙颌-牙槽代偿,临床中需谨慎应用,注意病例选择。  相似文献
