In 200 adult autopsy specimens, the arterial supply to the pancreas and spleen was studied radiologically and by manual dissection. The splenic artery divided into two or three lobar arteries, which supplied its corresponding lobe; each lobar artery subsequently divided into two to four lobular branches. Six to twelve lobular branches were observed entering the splenic substance at the hilum. Lobar arteries did not anastomose with each other, hence, the lobes of the spleen are also termed segments. The lobules, however, were not found to be independent segments and the arteries of one lobule anastomosed with those of other lobules. The branching pattern of the splenic artery varied from one specimen to another, so much so that a prevailing pattern could not be identified. Polar arteries, particularly to the superior pole, arose quite proximal to the hilum in 51% of cases and were occasionally missed. In 45% of males and 40% of females, the posterior gastric artery arose from about the middle of the splenic artery. The splenic artery was not found to be tortuous in fetuses, newborns, and young children. Tortuosity was seen in only 10% of adults; thus, the characteristic tortuosity of the splenic artery appears to develop with age. 相似文献

We investigated the influence of organ-specific parameters on tolerance and immunity to human MUC1. C57Bl/6 mice (wild-type) and C57Bl/6 transgenic for MUC1 (MUC1.Tg) were challenged in the pancreas with Panc02-MUC1, a C57Bl/6-syngeneic pancreatic cancer cell line expressing human MUC1. Wild-type mice produced immune responses to MUC1 when presented on tumor cells growing in the pancreas; however, the responses to tumors in the pancreas were less effective than responses produced by tumor challenge at the s.c. site. Tumor immunity specific for MUC1 was produced in wild-type mice by two different procedures: (i) s.c. immunization of wild-type mice with a low dose of Panc02-MUC1 or (ii) adoptive transfer of spleen and lymph node cells harvested from wild-type mice previously immunized s.c. with Panc02-MUC1. This demonstrates that immune responses to MUC1 presented at the s.c. site can be detected and adoptively transferred. MUC1.Tg mice were immunologically tolerant to MUC1; however, some immunological protection against orthotopic challenge with Panc02-MUC1 was conferred by adoptive transfer of CD4+ and CD8+ T cells from wild-type mice. These results show that it is more difficult to produce immune responses to tumors growing at the pancreatic site than the s.c. site. Panc02-MUC1 cells growing in the pancreas were accessible to the immune system, and immune responses evoked by s.c. presentation of this molecule in wild-type mice were effective in rejecting tumor cells in the pancreas of both wild-type and MUC1.Tg mice. Nonfunctioning islet cell tumors or pancreatic endocrine tumors are the most common type of malignant islet cell tumor. Although
previously detected usually at an advanced stage because of mass effect, the early detection rate of small localized disease
has been increasing. To date it has been difficult to predict the clinical behavior in localized regional nonfunctioning tumors.
To investigate potential markers predicting malignancy and poor prognosis in nonfunctioning pancreatic endocrine tumors, we
analyzed the expression of Ki-67, topoisomerase IIα (Topollα), and p27, as well as a variety of clinicopathologic parameters
in 76 cases of nonfunctioning islet cell tumors (23 benign cases and 53 malignant cases). Ki-67, Topollα, and p27 labeling
indices were significantly different between benign and malignant tumors. Expression of Ki-67, Topollα, and p27 were associated
with survival in patients with a malignant tumor in a univariate setting. However, only p27 and Topollα were jointly associated
with survival in multivariate analysis. Immunohistochemical staining for p27, Topollα, and Ki-67 can be helpful in the diagnosis
of nonfunctioning pancreatic endocrine tumor. Analysis of p27 and Topollα may also have potential utility as prognostic factors

The poor outcome of intraportal islet transplantation may be explained by the instant blood-mediated inflammatory reaction (IBMIR), characterized by islet entrapment in blood clots, leucocyte infiltration and disruption of islet morphology. Here we employ a newly developed in vitro system to identify the blood cells involved in this process. Islets were mixed with ABO-compatible blood in heparinized tubes and incubated for various times up to 6 h. Clots were analysed immunohistochemically for detection of platelets (CD41a), leucocytes/lymphocytes (CD11b), granulocytes (CD16, lysozyme), neutrophilic granulocytes (neutrophil elastase), eosinophilic granulocytes (NaCN + H(2)O(2)), macrophages (CD68), dendritic cells (CD209/DC-SIGN), B cells (CD20) and T cells (CD4, CD8). Platelets were rapidly deposited around the islets in contact with the blood, reaching a maximum by 30 min. The first neutrophilic granulocytes appeared in the islets after 15 min, increased at 1 h and peaked at 2 h. Small numbers of macrophages were found infiltrating the islets already after 5 min, with a slight increase over time. However, control stainings of cultured islets and pancreas biopsies identified these cells as being largely of donor origin. No T cells, B cells, dendritic cells or eosinophilic granulocytes were detected during the 6 h observation time. Neutrophilic granulocytes were identified as the main infiltrating blood cell in islets exposed to blood, implying that these cells play a key role in clinical islet transplantation. Because islets are known to be exquisitely susceptible to oxidative stress, development of drugs targeting neutrophilic cytotoxicity could markedly improve the outcome of islet transplantation. 相似文献

目的:研究Nestin、CK19、胰岛素、胰高血糖素及生长抑素在胚胎胰腺发育中的表达,探讨胰岛细胞分化发育的机制。方法:采用免疫组化SABC法及免疫组化双染法(LAB SA),对30例6~14wk人胚胎胰腺中,Nestin、CK19、胰岛素,胰高血糖素及生长抑素阳性的细胞进行定位。结果:(1)胚胎胰腺发育中Nestin阳性细胞存在于胰腺的间质,数量极少;CK19在胰腺导管上皮分化中持续表达,呈强阳性;(2)7wk胰腺导管上皮开始分化出胰岛细胞,胰岛素、胰高血糖素及生长抑素表达阳性,三者的表达并无时段差异性;随着胎龄的增加,阳性细胞数增加,14wk时胰岛逐渐形成,表达达到高峰。结论:胚胎胰腺的发育中,胰腺间质存在胰腺干细胞;胚胎早期胰腺导管上皮细胞开始分化并分泌胰岛素,其自分泌的激素可能参与调节胰岛细胞的迁移和胰岛的形成。 相似文献

目的探索Opa相互作用蛋白5(OIP5)在胰腺癌中的表达及其对PANC-1细胞增殖的影响。方法通过数据库分析OIP5在胰腺癌组织及癌旁组织中的表达;用实时定量PCR(RT-qPCR)和蛋白印迹法(Western blot)分别检测人胰腺癌细胞系MIAPaCa-2、PANC-1、KP-3、BxPC-3细胞中OIP5 mRNA和蛋白表达;构建OIP5基因沉默质粒的慢病毒(pGCSIL-shOIP5)和对照质粒慢病毒(pGCSIL-shCtrl),分别感染PANC-1细胞,分为OIP5基因沉默组和shCtrl对照组,5 d后采用RT-qPCR和Western blot测定慢病毒敲低效率,流式细胞计量术检测细胞凋亡;OIP5基因沉默组和shCtrl对照组连续5 d进行MTT检测和细胞计数;OIP5基因沉默组和shCtrl对照组孵育10 d形成集落,Giemsa染色分别集落总数。结果胰腺癌中OIP5 mRNA表达显著高于正常胰腺组织(P<0.05),OIP5高表达患者的总存活率显著低于OIP5低表达患者(P<0.05),且其无病生存率也显著降低(P<0.05);OIP5在MIAPaCa-2、PANC-1和KP-3中表达较高,而在BxPC-3细胞系中的表达较低;MTT检测结果显示OIP5沉默在第4和第5天显著降低了PANC-1细胞的增殖速率(P<0.01);OIP5沉默后细胞集落数(平均为9个)显著低于shCtrl对照组中的数量(平均为40个)(P<0.01);OIP5沉默后PANC-1细胞凋亡比例为8.3%显著高于shCtrl的4.5%(P<0.01)。结论OIP5在胰腺癌细胞系中异常高表达,OIP5基因可调控胰腺癌PANC-1细胞的增殖、凋亡以及集落形成,提示OIP5可能在胰腺癌发病机制中作为癌基因发挥作用,从而为胰腺癌的靶向治疗提供了潜在的生物标志物。 相似文献

目的 探索用静电法制备小微囊包裹成年猪胰岛细胞。方法 用自制的静电微囊发生装置、制备海藻酸钠 -多聚赖氨酸 -海藻酸钠 (APA)微囊 (微囊直径 <35 0 μm) ,包裹成年猪胰岛 ,体外检测APA小微囊猪胰岛生物活性及微囊膜的通透性 .结果 静电法制备的APA微囊直径 30 0~ 35 0 μm ,大小相对均一 .小微囊包裹成年猪胰岛后 ,每个微囊内可见 1~ 2个胰岛团 ,表面光滑 .囊内胰岛组织学结构完整 ,体外培养微囊化胰岛素分泌良好 ,葡萄糖刺激释放明显 ,显示了良好的细胞活力及微囊膜通透性 .结论 用我们自制的静电微囊发生装置能制备APA微囊包裹成年猪胰岛细胞 ,微囊直径 30 0~ 35 0 μm ,表面光滑 ,囊内猪胰岛生物活性良好 相似文献

The indirect immunofluorescence technique was used to demonstrate the coexistence of somatostatin together with avian pancreatic polypeptide-like immunoreactivity within certain neurons of the rat forebrain. Numerous neurons containing these peptides were observed in the neocortex, hippocampus, olfactory tubercle, striatum, nucleus accumbens and lateral septum. In studies of serial sections stained alternately for these two peptides, and in restaining experiments, It could be determined that in many neurons in these areas these two peptides coexisted. In other brain areas such as the anterior periventricular hypothalamus, somatostatin cells were never found to contain avian pancreatic polypeptide-like immunoreactivity. Also, within the pancreas these two peptides were never found to coexist in the same cells. Pancreatic somatostatinoma is a rare pancreatic endocrine neoplasm representing as little as 1% of pancreatic endocrine neoplasms (PENs). The histologic features of this tumor are like those of other PENs, except that it commonly forms acinar structures and often has cells with abundant, granular cytoplasm. We have recently encountered two of these neoplasms sampled by endoscopic ultrasound (EUS)-guided fine-needle aspiration (FNA). We discuss the cytologic and immunohistochemical findings of these two cases and the cytologic similarities these neoplasms share with pancreatic acinar-cell carcinoma (PACC). We review the cytologic features of PEN and PACC and discuss the importance of cell block immunohistochemistry in the diagnosis of pancreatic neoplasia sampled by EUS-guided FNA. 相似文献
