神经干细胞静脉移植治疗脊髓损伤的实验研究   总被引:3,自引:0,他引:3  
[目的]观察神经干细胞静脉移植对损伤大鼠脊髓功能的治疗作用。[方法]取孕14—16dSD胎鼠的脑室下区组织,体外培养后鉴定细胞。制作脊髓全切模型,伤后1周将Brdu标记好的神经干细胞通过尾静脉注射移植到大鼠体内,移植后及8周行皮层体感诱发电位(CSEP)检测和BBB功能评分,并留损伤脊髓处作病理切片及免疫组化染色。[结果](1)移植后8周BBB评分损伤组、移植组都有所恢复,但都未达到正常水平,移植组恢复较好;(2)模型制作后,CSEP波均消失,细胞移植后8周移植组的波形有不同程度的恢复,但潜伏期延长;(3)移植组大鼠脊髓损伤处存在大量Brdu染色阳性细胞,表明移植的细胞在体内可到达损伤脊髓处并能存活;脊髓损伤部位NF-200及GFAP染色阳性的细胞表明移植的细胞可以分化为具有神经元和胶质细胞特性的细胞。[结论]静脉移植的神经干细胞能到达损伤区代替受损的神经元及神经胶质细胞,使损伤的脊髓功能得到一定程度的恢复。  相似文献

We studied the trigeminal and facial motor nuclei of the hagfish by the retrograde HRP method. We distinguished 4 components in a single column of the motor nuclei of the trigeminal nerve and the facial nerve, viz., the pars magnocellularis of the trigeminal motor nucleus (mVm), the anterior part of the pars parvocellularis of the trigeminal motor nucleus (mVp1), the posterior part of the pars parvocellularis of the trigeminal motor nucleus (mVp2) and the facial motor nucleus (mVII). Although in Nissl preparations only the mVm could be distinguished from the rest of the nucleus, the boundaries of the other 3 components were clearly demarcated in HRP preparations. Intramuscular injections into two representative antagonistic jaw muscles revealed that there was no apparent topological organization of the neurons pertaining to the opening and closing muscles in the mVm and mVp1, but both antagonistic muscles were innervated bilaterally. Although the hagfish does possess a cartilaginous jaw, the organization pattern of the motor nuclei of the jaw muscles seems to be the most primitive of all living vertebrates.  相似文献

目的探讨急性脑血管病患者早期肾损害的机制及早期监测指标。方法对194例急性脑血管病患者(包括脑梗死129例,脑出血65例)及60例正常对照者同时测定血半胱氨酸蛋白酶抑制剂C(Cys-C)和血肌酐(SCr)浓度,对其中血Cys-c浓度增高但SCr浓度正常的48例急性脑血管病肾早期损害患者用甘露醇治疗,并检测治疗前后血Cys-c浓度。结果在早期肾损害组,血Cys-c浓度明显高于对照组(P<0.001),而SCr浓度2组无显著性差异(P>0.05)。48例急性脑血管病肾早期损害者经甘露醇治疗后,血Cys-c浓度较治疗前明显降低(P<0.001)。结论血清Cys-c测定有助于早期发现肾功能损害,治疗脑血管病的同时应注意监测肾功能。  相似文献

目的探讨中药当归、川芎、黄芪、丹参提取液对大鼠放射性骨髓损伤的防护作用.方法雄性SD大鼠60只,随机分为正常对照组、模型组、中药组,每组6只,在相同条件下饲养2周,模型组和中药组大鼠给予6.0 Gy 60Co γ射线一次性全身照射,继续饲养1周处死全部动物并取材.用骨髓细胞计数法计数骨髓有核细胞数,用Western blot法测定血管内皮生长因子(vascularendothelialgrowth factor,VEGF)、血小板衍生生长因子(ptatelet derived growth factor,PDGF)的蛋白含量.结果与正常对照组比较,模型组大鼠的骨髓有核细胞数、VEGF、PDGF蛋白含量明显减少(P<0.01),与模型组比较,中药组大鼠的骨髓有核细胞数、VEGF、PDGF蛋白含量明显增加(P<0.01或P<0.05).结论中药当归、黄芪、川芎、丹参对大鼠放射性骨髓损伤有一定防护作用.  相似文献

Physiologic assessment of diseases of the motor unit from the anterior horn cells to the muscles relies on a combination of needle electromyography (EMG) and nerve conduction studies (NCS). Both require a unique combination of knowledge of peripheral nervous system anatomy, physiology, pathophysiology, diseases, techniques, and electricity is necessary. Successful, high‐quality, reproducible EMG depends on the skills of a clinician in patient interaction during the physical insertion and movement of the needle while recording the electrical signals. Objectives –  To assess long‐term functioning and disability after traumatic brain injury (TBI). Material and methods –  Individuals (n = 88) in Norrbotten, northern Sweden, who had been transferred for neurosurgical care were assessed with internationally established TBI outcome measures 6–15 years post‐injury. Results –  There was an improvement in overall outcome from discharge from inpatient rehabilitation to follow‐up. Many individuals had a high degree of motor and cognitive functioning, which enabled them to live independently in their own home without assistance, but there remained a disability related to community reintegration and social participation. This affected their productivity and to some degree their marital stability. The remaining disability and reduced productivity were related to the age at injury and the injury severity. Conclusions –  Our data showed that individuals with a TBI can achieve and maintain a high degree of functioning many years after the injury. Increasing age and a greater injury severity contributed to their long‐term disability.  相似文献

Some investigators have suggested that preoperative chemotherapy for hepatic colorectal metastases may cause hepatic injury and increase perioperative morbidity and mortality. The objective of the current study was to examine whether treatment with preoperative chemotherapy was associated with hepatic injury of the nontumorous liver and whether such injury, if present, was associated with increased morbidity or mortality after hepatic resection. Two-hundred and twelve eligible patients who underwent hepatic resection for colorectal liver metastases between January 1999 and December 2005 were identified. Data on demographics, clinicopathologic characteristics, and preoperative chemotherapy details were collected and analyzed. The majority of patients received preoperative chemotherapy (n = 153; 72.2%). Chemotherapy consisted of fluoropyrimidine-based regimens: 5-FU monotherapy, 31.6%; irinotecan, 25.9%; and oxaliplatin, 14.6%. Among those patients who received chemotherapy, the type of chemotherapy regimen predicted distinct patterns of liver injury. Oxaliplatin was associated with increased likelihood of grade 3 sinusoidal dilatation (p = 0.017). Steatosis >30% was associated with irinotecan (27.3%) compared with no chemotherapy, 5-FU monotherapy, and oxaliplatin (all p < 0.05). Irinotecan also was associated with steatohepatitis, as two of the three patients with steatohepatitis had received irinotecan preoperatively. Overall, the perioperative complication rate was similar between the no-chemotherapy group (30.5%) and the chemotherapy group (35.3%) (p = 0.79). Preoperative chemotherapy was also not associated with 60-day mortality. In patients with hepatic colorectal metastases, preoperative chemotherapy is associated with hepatic injury in about 20 to 30% of patients. Furthermore, the type of hepatic injury after preoperative chemotherapy was regimen-specific. 用5只猫于左侧颈部食管壁内注入 HRP 溶液,通过 HRP 逆行追踪法研究颈部食管的交感神经支配,结果表明:1.长轴突型交感节前神经元直接分布到食管壁内,其标记细胞位于双侧脊髓的胸1~3节段,以胸2节段最多(占标记细胞总数的66.45%),注射侧占优势。标记细胞主要位于中间带外侧核(约占95.02%),其次为侧索、中介核、前角腹后外侧核。其细胞形态不一,以中小型细胞为主(占标记细胞总数的90.4%)。2.支配颈部食管的交感节后神经元主要位于星状神经节(约占61.99%),余者位于双侧颈前、中和2~5胸交感节内、以小细胞最多。  相似文献
