目的 :构建丙型肝炎病毒 (HCV)包膜糖蛋白 (E1E2 )基因的原核表达载体 ,为研究HCVE1E2蛋白属性及基因免疫奠定基础。方法 :设计HCVE1E2区基因上、下游引物 ,分别引入BglⅡ及EcoRI酶切位点 ,以含有HCVH株基因序列的质粒pBRTM/HCV1- 30 11为模板 ,通过PCR扩增获得HCVE1E2区基因片段 ,将其克隆入原核表达载体pRSETA ,然后通过酶切和序列测定对其进行鉴定 ;将此表达载体转化入表达菌BL2 1(DE3)pLySs诱导表达 ,并采用SDS -PAGE对表达产物进行鉴定。结果 :经酶切及序列测定分析表明 ,成功地构建了重组原核表达载体pRSETA -HCVE1E2 ;SDS -PAGE显示目的蛋白大量表达 ,且呈非溶状态。结论 :HCVE1E2融合蛋白可以在大肠杆菌中大量表达 ,为HCVE1E2蛋白的制备及基因疫苗的研制奠定了基础。 相似文献

BackgroundEmerging research has suggested a plausible relationship may exist between lower limb coordination and musculoskeletal injury. A small number of studies have investigated the link between coordination and anterior cruciate ligament (ACL) injury during sidestep cutting. While prior work has shown unanticipated sidestep cutting to exhibit a more ‘at risk’ kinematic profile compared to anticipated tasks, a detailed understanding of the coordination between multiple joints and how they differ during unanticipated actions is lacking, particularly in females.Research questionThe purpose of this study was to observe the difference in trunk, pelvis and lower limb coordination and coordination variability during a dynamic, sidestep cutting task under anticipated and unanticipated conditions in a healthy female cohort.MethodsThree-dimensional motion analysis data were recorded during anticipated and unanticipated sidestep cutting for nineteen healthy female participants (age, 24 ± 3yrs; height, 164 ± 5 cm; and weight, 58 ± 6 kg). Vector coding methodology was used to calculate coordination and coordination variability values and statistical parametric and non-parametric mapping was used to comprehensively determine differences between anticipated and unanticipated conditions.ResultsDifferences were observed between anticipated and unanticipated conditions in the hip flexion – knee abduction angle (89 % of stance), hip rotation – knee abduction angle (55 % of stance), knee flexion – knee abduction angle (81–83 %, 86 % and 88–89 %) and knee flexion – ankle flexion angle (14–18 %) coupling angles. Differences in coupling angle variability were also observed with only one cluster of significance seen in hip abduction – knee abduction variability (27–30 % of stance).SignificanceHealthy females exhibit significant differences in lower limb coupling angles and coupling angle variability between anticipated and unanticipated sidestep cutting. Interventions aimed at reducing ACL injury risk may need to consider that anticipated and unanticipated sidestep cutting tasks present unique demands, and therefore should both be trained specifically. 相似文献

目的 克隆人源脯氨酸-谷氨酸-亮氨酸富集蛋白1(proline-,glutanic acid-,leucine-rich proteinl,PELP1)基因全长,构建PELP1基因全长过表达载体,研究PELP1基因表达特点,为PELP1基因在不同组织细胞中的功能性研究奠定基础.方法 采用反转录PCR法从MCF-7细胞总RNA中克隆PELP1基因全长,与含有EcoR I与Xho I双酶切位点的真核过表达载体pIRES2-EGFP质粒载体连接,构建重组质粒pIRES2-EGFP-PELP1.经双酶切及测序鉴定重组质粒中PELP1基因的完整性及正确性.运用脂质体将重组质粒转染至人牙周膜干细胞中,运用实时定量PCR(quantitative real-time PCR,qRT-PCR)及蛋白质印迹法分别从基因和蛋白水平对转染细胞中PELP1的表达进行检测.结果 成功克隆出PELP1基因全长,并将其插入至pIRES2-EGFP过表达载体中,经双酶切鉴定和测序鉴定证实目的质粒片段插入无误.重组质粒pIRES2-EGFP-PELP1转染48 h后PELP1基因水平是转染空质粒人牙周膜干细胞的(148.0±1.3)倍,二者差异有统计学意义(P< 0.005).蛋白质印迹法结果显示,与转入pIRES2-EGFP的人牙周膜干细胞中PELP1蛋白相对表达量(0.3300±0.0117)相比,转入pIRES2-EGFP-PELP1重组质粒的人牙周膜干细胞中PELP1蛋白相对表达量(0.6200 ±0.0045)显著增高,二者差异有统计学意义(P< 0.005).结论 本项研究成功运用qRT-PCR反应从MCF-7细胞中克隆出PELP1全长基因,并成功构建pIRES2-EGFP-PELP1真核过表达载体. 相似文献

背景:研究表明,力生长因子(mechano-growth factor,MGF)能激活卫星细胞,促进成肌细胞增殖,在治疗肌损失、预防心肌损伤和修复神经损伤等方面有重要的作用。机械拉伸可使成骨细胞表达MGF,但是MGF对骨组织生理生化行为的影响机制尚不清楚。
目的:应用携带MGF基因的重组腺病毒载体转染成骨细胞,观察MGF在成骨细胞中的表达。
方法:将MGF基因插入到腺病毒穿梭质粒pAdTrack-CMV中,构建pAdTrack-MGF重组体。pAdTrack-MGF与腺病毒骨架质粒pAdEasy-1在BJ5183菌中进行同源重组,生成重组腺病毒质粒pAdEasy-MGF。脂质体介导pAdEasy-MGF转染293A细胞,包装成整的重组腺病毒Ad/MGF。用Ad/MGF感染原代培养的大鼠成骨细胞,荧光示踪计数法测定感染效率,RT-PCR法对重组腺病毒感染结果进行鉴定。
结果与结论:实验构建的重组腺病毒载体Ad/MGF滴度可达8.5×108 pfu/mL。Ad/MGF能高效感染体外培养的Wistar大鼠成骨细胞并表达目的基因,当感染复数为100时,感染效率最高。说明实验构建的Ad/MGF重组腺病毒可在大鼠成骨细胞中有效表达。 相似文献

目的:探讨人Tip30全长基因的真核表达载体pAAV-TIP30的构建,并观察其转染肝癌细胞株HepG2后的表达。方法:以正常人肝组织mRNA为模板,用RT-PCR方法扩增Tip30,利用限制性内切酶BamH I和Xba I,将Tip30基因克隆到真核表达载体pAAV-MCS中,并经酶切和测序鉴定。重组质粒转染HepG2细胞,运用Western blot法检测Tip30蛋白表达。结果:RT-PCR方法正确地扩增出全长Tip30基因。限制性内切酶酶切和测序结果证实Tip30基因克隆完全正确。重组质粒转染肝癌细胞系HepG2后,Western blot证实Tip30蛋白表达增高。结论:成功构建了真核表达重组质粒pAAV-TIP30并转染HepG2细胞,为进一步研究Tip30基因对肝癌的影响及其机制打下了基础。 相似文献

Foamy virus vectors are potent alternatives to lenti- and gamma-retroviral vectors for gene therapy. To construct and optimize gutless feline foamy virus (FFV) replication-deficient (RD) vectors, viral elements essential for optimal efficient marker gene transduction were characterized and fine-mapped and packaging clones constructed. For these purposes, new Gag and Pol expression clones which allow efficient expression of packaging proteins and vectors carrying deletions in coding and non-coding regions of the genome were constructed and functionally evaluated. These studies demonstrate that the 5′ major splice donor (5′ SD) is indispensable for RD vectors while defined mutations introduced to inactivate the gag start codon improve transgene delivery efficiency. Based on these findings, new gutless FFV vectors were generated yielding un-concentrated vector titers above 105 transducing units (TU)/ml. By minimizing the second cis-acting sequence in the pol gene, only 3.8 kb viral sequences are maintained in the novel gutless FFV RD vectors. 相似文献

ObjectiveTo assess whether ictal electric source imaging (ESI) on low-density scalp EEG can approximate the seizure onset zone (SOZ) location and predict surgical outcome in children with refractory epilepsy undergoing surgery.MethodsWe examined 35 children with refractory epilepsy. We dichotomized surgical outcome into seizure- and non-seizure-free. We identified ictal onsets recorded with scalp and intracranial EEG and localized them using equivalent current dipoles and standardized low-resolution magnetic tomography (sLORETA). We estimated the localization accuracy of scalp EEG as distance of scalp dipoles from intracranial dipoles. We also calculated the distances of scalp dipoles from resection, as well as their resection percentage and compared between seizure-free and non-seizure-free patients. We built receiver operating characteristic curves to test whether resection percentage predicted outcome.ResultsResection distance was lower in seizure-free patients for both dipoles (p = 0.006) and sLORETA (p = 0.04). Resection percentage predicted outcome with a sensitivity of 57.1% (95% CI, 34–78.2%), a specificity of 85.7% (95% CI, 57.2–98.2%) and an accuracy of 68.6% (95% CI, 50.7–83.5%) (p = 0.01).ConclusionIctal ESI performed on low-density scalp EEG can delineate the SOZ and predict outcome.SignificanceSuch an application may increase the number of children who are referred for epilepsy surgery and improve their outcome. 相似文献
