By employing neoglycoproteins (NGP) and glycosaminoglycans, the detection of endogenous glycoligand-binding sites has become possible. Monitoring specific binding of 11 of these sugar receptor-specific tools, 13 trypanosomatids of monogenetic genera Blastocrithidia, Crithidia, Herpetomonas , and Leptomonas and digenetic genera Endotrypanum , Leishmania , and Sauroleishmania were analysed by agglutination and fluorescence assays. NGP showed agglutination reactions only with the digenetic but not with the monogenetic species. Sensitive flow cytofluorimetric investigations revealed that the apparently different reactivity to NGP is due to a pronounced quantitative difference in expression of binding sites between mono- and digenetic flagellates. Moreover, flow cytofluorimetry was used to demonstrate the occurrence of receptor sites for heparin on the cell surfaces of all trypanosomatids. An indication for a correlation of the binding capacity for the NGP N-acetyl-β-d-glucosamine and heparin to differences in the pathogenicity of parasites was observed for Leishmania donovani as well as Leishmania enriettii . Infective populations of these species contained a significantly higher number of cells which had bound N-acetyl-β-d-glucosamine and heparin than noninfective (long-term in vitro -cultured) populations. The results of the present report additionally support the hypothesis that lectin–carbohydrate interactions with neutral sugar moieties and heparin or heparin-like molecules participate in the interactions between trypanosomatids and host (cells), and that the detected binding sites for carbohydrates and heparin may thus be referred to as potential virulence factors.  相似文献

采用无抗凝作用剂量肝素皮下注射治疗小儿肾病综合征25例,观察了近期疗效与12~24月后对皮质激素或环磷酰胺(CTX)治疗反应的关系.结果表明,近期对肝素治疗显效但复发以及有效者,今后用激素或CTX治疗均可达完全缓解,无效者则效果较差.本文结果表明,肝素治疗的近期疗效可用于预测肾病患儿的远期预后.  相似文献

目的联合应用肝素及血管内皮细胞生长因子(VEGF)预处理以提高人工血管移植物内皮细胞(EC)生长及贴附率.方法联合应用肝素及血管内皮细胞生长因子预处理人工血管移植物,比较各组EC生长曲线和贴附率.结果肝素在体外与VEGF联合应用,可显著增加EC在人工血管内壁的贴附率,并对EC的增殖有一定的持续影响.结论肝素与VEGF联用,能促进EC在人工血管的增殖和贴附率.  相似文献

目的:观察危重型肾功能衰竭合并高危出血患者选择抗凝药物进行血液透析治疗的效果。方法:应用吉派林(低分子肝素钠)作为抗凝剂进行血液透析。结果:129例病人进行血液透析386次,均没有出血发生。结论:吉派林在高危出血患者血液透析中使用方便,安全,效果良好。  相似文献

This study shows that treatment of rats with exogenous glycosaminoglycans stimulates peripheral nerve regeneration, increases the abundance of mRNAs for myelin proteins and promotes muscle reinnervation. After the sciatic nerve had been crushed the number of regenerating axons in the distal stump was markedly and highly significantly increased by glycosaminoglycan treatment throughout the experimental period. The increased number of axons was correlated with increased axon and fibre (axon + myelin) diameter. The abundance of mRNAs for Po protein and myelin basic protein of regenerating nerves was also affected by treatment with glycosaminoglycans. The increase in mRNA was also observed in the contralateral unlesioned nerve. Such a phenomenon did not occur in saline-treated rats. Glycosaminoglycan treatment markedly increased the number of muscle fibres reinnervated and accelerated the restoration of muscle twitch tension elicited by nerve stimulation. The effect was particularly evident during the early stages (16 and 21 days after nerve crush) of muscle reinnervation.  相似文献

Fibrinolytic therapy substantially reduces mortality from acute myocardial infarction. Patient selection is, however, important. The patient must present within 12 hours of the onset ofischaemic symptoms, have definite ECG changes of ST elevation or left bundle branch block and no contraindications. The major contraindications are those for risk of an intracerebral bleed, recent stroke, intracranial tumour or risk of a major systemic bleed. Age and hypertension are not contraindications but may modify the regimen used.
Heparin is required with recombinant tissue plasminogen activator but is optional with streptokinase.
The recent COBALT trial suggests that the accelerated weight related t-PA regimen given over 90 minutes is more satisfactory than double bolus t-PA. However, inpatients under 75 years of age, the two regimens were equivalent.
For patients suffering acute myocardial infarction, practitioners should now individualise choice of therapy, rather than give the same cocktail to all patients. The choice of regimen will depend on the cardiac risk, the stroke risk, the bleeding risk and the cost.  相似文献

Glycosaminoglycan administration has favourable effects on morphologicaland functional renal abnormalities in different models. Thepossibility that exogenous glycosaminoglycans modulate glomerularmatrix synthesis was explored in both primary and SV40-MES13murine mesangial cell cultures. On both cell types, both low-molecular-weightheparin and different glycosaminoglycans showed dose-dependentinhibition of proliferation and increase of 35SO2–4 uptake.After 36 h the cell compartment contained a spectrum of 35S-moleculesof less than 200 kDa; under heparin treatment, the two main35SO2–4 components (high and medium MW) increased by 16and 37% respectively. Susceptibility to glycosidases revealedthat heparin promotes the expression of heparan sulphate andincreases that of chondroitin sulphate. Moreover, heparin modifiesthe expression of decorin and bigly-can, involved in adhesionand fibrillogenesis, while not affecting perlecan. The extracellularmatrix modulation in renal cells, for which the sulphation typeand ratio of heparin are crucial, may thus explain the beneficialrenal effects of heparin.  相似文献

目的 评测3种离子键肝素涂层体外循环管道的抗凝血性能和稳定性。方法 用PT和APTT对不同浓度肝素—氯烃基二甲基代苯甲胺(HBC)复合物涂层体外循环管道的凝血性能进行评测,同时测试体外转流对3种肝素涂层管道抗凝血性能的影响。结果 3种肝素涂层方法均能够将肝素分子结合于材料表面并具有抗凝活性,其中HBC复合物和肝素—聚乙烯亚胺复合物处理的体外循环管道经体外转流96h后仍具有较佳的抗凝血活性。结论 离子键肝素涂层因结合物质不同其稳定性也不同;HBC复合物和肝素—聚乙烯亚胺复合物处理的体外循环管道肝素分子结合较牢固,能够满足临床短期使用需要。  相似文献

The thromboresistant properties of hydrophilic gels based on copolymers of nitrogen-containing heterocyclic vinyl compounds with vinyl monomers were investigated. The hydrophilic gels were shown to prevent adsorption of fibrinogen, activation of procoagulants, and adhesion of platelets. Hydrogen surfaces possess selective affinity for plasma albumin. The authors consider that the thromboresistant effect of hydrophilic gels is due to the competitive action of plasma albumin.
