目的:建立基于Rotor-Gene的链融解曲线法的甲基化标记筛选技术,分析各种影响因素,评估其实用性.方法:克隆Hela细胞雄激素受体基因外显子-1的Bisulfite-PCR产物,挑出经测序证实的可以代表不同甲基化程度的一组质粒,并以之为模板,在Rotor-Gene 3 000实时定量PCR仪进行扩增和链融解,分析融链温度、链融解曲线与甲基化程度、甲基化模式的相关性及各种影响因素.结果:基于Rotor-Gene 的链融解曲线法,不仅可以区分甲基化与非甲基化,而且能够鉴别片段内细微的甲基化差异;主要的影响因素有升温幅度、缓冲液离子强度、产物浓度等.结论:基于Rotor-Gene 的链融解曲线法,可以集扩增和链融解于一体,是一种经济简便的甲基化分析技术,可用于样品的甲基化筛选.  相似文献

液相连续反应釜反应效果可如下进行预测。通过实验室装置测定并绘制拟定条件下的反应动力学曲线。把生产釜模拟为带死角和短路的理想釜,用水作介质在生产釜中进行流动模型试验,测量一系列停留时间分布密度函数,得到两个模型参数,并建立操作线方程。反应动力学曲线和操作线的交点为此液相连续反应釜的预测结果。  相似文献

Eucommia ulmoides Oliv. Bark (EUE) is commonly used for the treatment of hypertension, rheumatoid arthritis, lumbago, and ischialgia as well as to promote longevity. In this study, we tested the effects of EUE aqueous extract in graded doses to protect and enhance cognition in scopolamine-induced learning and memory impairments in mice. EUE significantly improved the impairment of short-term or working memory induced by scopolamine in the Y-maze and significantly reversed learning and memory deficits in mice as measured by the passive avoidance and Morris water maze tests. One day after the last trial session of the Morris water maze test (probe trial session), EUE dramatically increased the latency time in the target quadrant in a dose-dependent manner. Furthermore, EUE significantly inhibited acetylcholinesterase (AChE) and thiobarbituric acid reactive substance (TBARS) activities in the hippocampus and frontal cortex in a dose-dependent manner. EUE also markedly increased brain-derived neurotrophic factor (BDNF) and phosphorylation of cAMP element binding protein (CREB) in the hippocampus of scopolamine-induced mice. Based on these findings, we suggest that EUE may be useful for the treatment of cognitive deficits, and that the beneficial effects of EUE are mediated, in part, by cholinergic signaling enhancement and/or protection.  相似文献

Purpose: To investigate the effect of the cumulative exposure to estradiol (E2) during the follicular phase on IVF outcome. Methods: Patients were stimulated with recombinant FSH after GnRH agonist suppression and had a day 3-embryo transfer. Estrogen exposure was determined as the area under the curve (AUC) for serum E2 levels measured from the first day of stimulation through the day after hCG administration. Results: E2 AUC thresholds for 10th and 90th percentiles were 4704 pg/ml and 16338 pg/ml, respectively. The pregnancy and implantation rates were highest in the 10th–90th percentile group, and were statistically higher in this group than in the >90th percentile group (54.6% vs. 33.3% and 24.8% and 12.9%, respectively, for pregnancy and implantation rates, P < 0.05). Recovered mature oocytes, fertilization, and number and mean score of transferred embryos were similar. Conclusions: High cumulative E2 exposure during the follicular phase of IVF cycles has detrimental effects on implantation.  相似文献

Background Computer-aided diagnosis (CAD) of lung cancer is the subject of many current researches. Statistical methods and artificial neural networks have been applied to more quantitatively characterize solitary pulmonary nodules (SPNs). In this study, we developed a CAD scheme based on an artificial neural network to distinguish malignant from benign SPNs on thin-section computed tomography (CT) images, and investigated how the CAD scheme can help radiologists with different levels of experience make diagnostic decisions.Methods Two hundred thin-section CT images of SPNs with proven diagnoses (135 small peripheral lung cancers and 65 benign nodules) were analyzed. Three clinical features and nine CT signs of each case were studied by radiologists, and the indices of qualitative diagnosis were quantified. One hundred and forty nodules were selected randomly to form training samples, on which the neural network model was built. The remaining 60 nodules, forming test samples, were presented to 9 radiologists with 3–20 years of clinical experience, accompanied by standard reference images. The radiologists were asked to determine whether a nodule was malignant or benign first without and then with CAD output. Diagnostic performance was evaluated by receiver operating characteristic (ROC) analysis.Results CAD outputs on test samples had higher agreement with pathological diagnoses (Kappa=0.841, P&lt;0.001). Compared with diagnostic results without CAD output, the average area under the ROC curve with CAD output was 0.96 (P&lt;0.001) for junior radiologists, 0.94 (P=0.014) for secondary radiologists and 0.96 (P=0.221) for senior radiologists, respectively. The differences in diagnostic performance with CAD output among the three levels of radiologists were not statistically significant (P=0.584, 0.920 and 0.707, respectively). Conclusions This CAD scheme based on an artificial neural network could improve diagnostic performance and assist radiologists in distinguishing malignant from benign SPNs on thin-section CT images.  相似文献

目的 探讨消旋-3-正丁基苯酞(NBP)对血管性痴呆大鼠学习记忆的影响.方法 用两血管法(2VO)建立血管性痴呆(vascular dementia,VD)模型,将60只3月龄雄性Wistar大鼠随机分成VD组、假手术组和NBP组,NBP组以丁苯酞灌胃1个月,VD组和假手术组给予等量的植物油,以Morris水迷宫测试各组大鼠的学习记忆能力,HE染色观察大鼠海马区脑组织形态.结果 NBP组和假手术组和VD组的隐蔽平台逃避潜伏期[分别为(38.34±2.46)s,(14.83±3.77s),(75.74±6.33)s];原平台象限时间[分别为(26.45±4.66)s,(35.21±3.78)s,(18.67±5.39)s].与VD组相比,NBP组明显缩短隐蔽平台逃避潜伏期,增加原平台象限时间,并在HE染色中减少神经元坏死.结论 NBP能显著改善VD大鼠的学习记忆能力.
Abstract:
Objective To investigate the effect of 3-n-butylphthalide (NBP) on learning and memory abilities in vascular demensia (VD) rats. Methods VD model rats was established by two-vessel method. 60 3-month-old Wistar male rats were randomly divided into VD group, sham-operate group and NBP group. Rats in NBP group were given NBP 120 mg · kg-1 · d-1 ,VD and sham-operate group were given equal quality vegetable oil.Morris water maze was used to assess the learning and memory abilities in each group and HE staining was used to observe the hippocampus morphology of the rats. Results The escape latency in hidden plat test was( (38.34 ±2.46 ) s, ( 14.83 ± 3.77s ), ( 75.74 ± 6.33 ) s ) and the original platform quadrant time was ( ( 26.45 ± 4.66 ) s,(35.21 ±3.78)s, ( 18.67 ±5.39)s) in NBP group,sham-operate group and VD group respectively. Compared to VD group, NBP group had obviously decreased escape latency in hidden plat test, increased original platform quadrant time and distinctly decreased the necrosis of the neurons in HE dyeing. Conclusion NBP can improve the learning and memory deficits in VD rats.  相似文献

目的研究影像实践教学中开展案例式PBL教学法对构建新型师生关系的影响。方法在我院影像诊断专业班级进行案例式PBL实验教学后,在评价学生分析能力、自主能力、创新能力的同时,重点研究此新型教学模式在构建新型师生关系方面的意义。结果学生不仅在影像诊断分析能力方面有显著提高,师生关系也由传统“教师权威制”、“师道尊严制”转变为“平等”、“尊重”为主的师生交互关系。结论案例式PBL教学法在构建新型师生关系方面取得非常好的效果。  相似文献

ObjectiveThe aim of this study was to compare the qualitative and quantitative targets in the detection of proteinuria in children with HSP.MethodsThree hundred and forty children were taken as the research subjects. Qualitative and a series of quantitative indicators of urine protein were tested.ResultsThe qualitative and quantitative of protein in 340 children were analyzed retrospectively. Correlation analysis found that 24hUTP had a strong correlation with age, RUMA, RUPCR, and qualitative urine occult blood (< 0.01, for all). After regression analysis, RUMA and RUPCR were still significantly correlated with 24hUTP (< 0.01). At the same time, RUMA and RUPCR had good diagnostic accuracy for proteinuria. When the RUPCR ratio was set as 0.415, the diagnostic sensitivity was 83%, and the specificity was 98.7%. When RUMA was set as 68.215, the sensitivity was 94.3% and the specificity was 96.2%.ConclusionCompared with 24hUTP, RUMA and RUPCR had high sensitivity and specificity for monitoring proteinuria in children with HSP.  相似文献

目的探讨直接测定红细胞平均血红蛋白含量(celluarHb,CHb)在贫血诊断中的意义。方法通过全自动血液分析仪检测健康人群及不同类型贫血患者的CHb,并进行ROC曲线分析。结果CHb的正常参考范围是27.74~31.21pg;除了正常细胞均一性贫血患者的CHb与健康人群无显著性差异(P>0.05)外,下述四类贫血患者的CHb与健康人群均有极显著性差异(P<0.01):在大细胞均一性贫血诊断时,若以CHb>31.2pg为分界值,其敏感度0.94、特异性0.99、诊断效率0.97;在大细胞不均一性贫血诊断时,以CHb>31.2pg为分界值,其敏感度1.00、特异性0.99、诊断效率0.99;在诊断正常细胞不均一性贫血时,以CHb<29.4pg为分界值:敏感度0.62、特异性0.54、诊断效率0.57;在诊断小细胞性贫血时,以CHb<27.2pg为分界值,诊断敏感度1.00、特异性1.00、诊断效率1.00。结论直接测定能真实反映机体红细胞血红蛋白含量,结合合适的分界值,在各种贫血的诊断与鉴别诊断中,尤其是小细胞性贫血,CHb是一项理想的诊断指标,值得进一步推广应用。  相似文献
