The extracellular matrix (ECM) Matrigel™ has frequently and successfully been used to generate new adipose tissue experimentally, but is unsuitable for human application. This study sought to compare the adipogenic potential of a number of alternative, biologically derived or synthetic ECMs with potential for human application, with and without growth factors and a small fat autograft. Eight groups, with six severe combined immunodeficient (SCID) mice per group, were created with bilateral chambers (silicone tubes) implanted around the epigastric vascular pedicle, with one chamber/animal containing a 5 mg fat autograft. Two animal groups were created for each of four ECMs (Matrigel™, Myogel, Cymetra® and PuraMatrix™) which filled the bilateral chambers. One group/ECM had no growth factors added to chambers whilst the other group had growth factors (GFs) (vascular endothelial growth factor-A (VEGF-A) plus fibroblast growth factor-2 (FGF-2) plus platelet-derived growth factor-BB (PDGF-BB)) added to both chambers. At 6 weeks, chamber tissue was morphometrically assessed for percent and absolute adipose tissue volume. Overall, the triple GF regime significantly increased percent1 and absolute# adipose tissue volume (p < 0.00051#) compared to chambers without triple GF treatment. The fat autograft also significantly increased percent (p < 0.0005) and absolute (p < 0.011) adipose tissue volume. Cymetra® (human collagen) constructs yielded the largest total tissue and absolute adipose tissue volume. We found that the pro-angiogenic FGF-2, VEGF-A and PDGF-BB combination in ECMs of synthetic and biological origin produced an overall significantly increased adipose tissue volume at 6 weeks and may have clinical application, particularly with Cymetra. 相似文献

Background and aimsMenopause may reduce fat oxidation. We investigated whether sex hormone profile explains resting fat oxidation (RFO) or peak fat oxidation (PFO) during incremental cycling in middle-aged women. Secondarily, we studied associations of RFO and PFO with glucose regulation.Method and resultsWe measured RFO and PFO of 42 women (age 52–58 years) with indirect calorimetry. Seven participants were pre- or perimenopausal, 26 were postmenopausal, and nine were postmenopausal hormone therapy users. Serum estradiol (E2), follicle-stimulating hormone, progesterone, and testosterone levels were quantified with immunoassays. Insulin sensitivity (Matsuda index) and glucose tolerance (area under the curve) were determined by glucose tolerance testing. Body composition was assessed with dual-energy X-ray absorptiometry; physical activity with self-report and accelerometry; and diet, with food diaries. Menopausal status or sex hormone levels were not associated with the fat oxidation outcomes. RFO determinants were fat mass (β = 0.44, P = 0.006) and preceding energy intake (β = ?0.40, P = 0.019). Cardiorespiratory fitness (β = 0.59, P = 0.002), lean mass (β = 0.49, P = 0.002) and physical activity (self-reported β = 0.37, P = 0.020; accelerometer-measured β = 0.35, P = 0.024) explained PFO. RFO and PFO were not related to insulin sensitivity. Higher RFO was associated with poorer glucose tolerance (β = 0.52, P = 0.002).ConclusionAmong studied middle-aged women, sex hormone profile did not explain RFO or PFO, and higher fat oxidation capacity did not indicate better glucose control. 相似文献

目的:尝试建立基层医院肠外营养(PN)临床合理应用评价方法。方法:收集脂肪乳氨基酸(17)葡萄糖(11%)注射液的使用病例进行调查,统计分析病例资料的完整性,评价配伍用药情况,计算基础代谢值(BMR)并作相关评估。结果:营养风险评估基本参数不全的病例占66.7%,各病例疗程差异显著但用量无差别,脂肪乳氨基酸(17)葡萄糖(11%)注射液非蛋白热卡占个体BMR比例,平均值为72.1%(S=8.9)。脂肪乳氨基酸(17)葡萄糖(11%)注射液配伍用药中加入氯化钾注射液的病例占63.0%,其中70.6%超推荐剂量;加入丙氨酰谷氨酰胺的病例为62.9%,用量全部超推荐剂量。结论:本院PN应用中营养风险评估基本缺失,合理的个体化方案制订水平与指南要求有较大差距,建议医院临床药师在PN应用方面加大相关监测评价等工作力度。 相似文献

Cosmetic facial filler-related ophthalmic artery occlusion is rare but is a devastating complication, while the exact pathophysiology is still elusive. Cerebral angiography provides more detailed information on blood flow of ophthalmic artery as well as surrounding orbital area which cannot be covered by fundus fluorescein angiography. This study aimed to evaluate cerebral angiographic features of cosmetic facial filler-related ophthalmic artery occlusion patients. We retrospectively reviewed cerebral angiography of 7 patients (4 hyaluronic acid [HA] and 3 autologous fat-injected cases) showing ophthalmic artery and its branches occlusion after cosmetic facial filler injections, and underwent intra-arterial thrombolysis. On selective ophthalmic artery angiograms, all fat-injected patients showed a large filling defect on the proximal ophthalmic artery, whereas the HA-injected patients showed occlusion of the distal branches of the ophthalmic artery. Three HA-injected patients revealed diminished distal runoff of the internal maxillary and facial arteries, which clinically corresponded with skin necrosis. However, all fat-injected patients and one HA-injected patient who were immediately treated with subcutaneous hyaluronidase injection showed preserved distal runoff of the internal maxillary and facial arteries and mild skin problems. The size difference between injected materials seems to be associated with different angiographic findings. Autologous fat is more prone to obstruct proximal part of ophthalmic artery, whereas HA obstructs distal branches. In addition, hydrophilic and volume-expansion property of HA might exacerbate blood flow on injected area, which is also related to skin necrosis. Intra-arterial thrombolysis has a limited role in reconstituting blood flow or regaining vision in cosmetic facial filler-associated ophthalmic artery occlusions. 相似文献

The term lipidome is mentioned to the total amount of the lipids inside the biological cells. The lipid enters the human gastrointestinal tract through external source and internal source. The absorption pathway of lipids in the gastrointestinal tract has many ways; the 1st way, the lipid molecules are digested in the lumen before go through the enterocytes, digested products are re-esterified into complex lipid molecules. The 2nd way, the intracellular lipids are accumulated into lipoproteins (chylomicrons) which transport lipids throughout the whole body. The lipids are re-synthesis again inside the human body where the gastrointestinal lipids are: (1) Transferred into the endoplasmic reticulum; (2) Collected as lipoproteins such as chylomicrons; or (3) Stored as lipid droplets in the cytosol. The lipids play an important role in many stages of the viral replication cycle. The specific lipid change occurs during viral infection in advanced viral replication cycle. There are 47 lipids within 11 lipid classes were significantly disturbed after viral infection. The virus connects with blood-borne lipoproteins and apolipoprotein E to change viral infectivity. The viral interest is cholesterol- and lipid raft-dependent molecules. In conclusion, lipidome is important in gastrointestinal fat absorption and coronavirus disease 2019 (COVID-19) infection so lipidome is basic in gut metabolism and in COVID-19 infection success. 相似文献

To evaluate the significance of micrometastases in relation to survival rate, specimens from 48 colorectal carcinoma patients
were analysed after fat clearance. The number and size of the lymph nodes harbouring metastases and the significance of micrometastases
for patients' survival were assessed. We found that although the majority of metastatic lymph nodes (71.8%) were 5 mm or less
in diameter, their size had no effect on survival. Immunohistochemical staining of lymph nodes revealed that 15 of 25 patients
with Dukes' stage B diagnosed by routine staining had micrometastases, 86% of these lymph nodes being less than 5 mm in diameter.
The survival rate of this subgroup was found to be considerably poorer than that of Dukes' stage B patients with no micrometastases.
None of the three patients with Dukes' stage A carcinoma had micrometastases. Since most of the metastases and micrometastases
occur in lymph nodes of 5 mm and less and can be easily missed by routine examination, we suggest that fat clearance and routine
immunohistochemical analysis of Dukes' stage B improve the prediction of outcome of colorectal cancer patients.

Background: The assessment of Fat Mass and Fat-Free Mass indices provides valuable information about changes in body composition.Aim: To identify cut-off points for Fat Mass Index (FMI) to predict an upper limit of percentage body fat of men (25%) and women (30%) for defining obesity and its association with hypertension.Subjects and methods: A total of 436 men and 596 women were included in the study. Fat mass was calculated using skin-fold measurements. FMI cut-off points to predict an upper limit of percentage body fat of 25% (men) and 30% (women) for defining obesity were assessed using Receiver Operating Characteristic (ROC) curve analysis.Results: ROC curve analysis indicated that the level of FMI was 6.59 kg/m2 in men and 6.64 kg/m2 in women at 25% and 30% body fat, respectively. Risk estimation for hypertension with FMI indicated high risk of hypertension in men (OR: 3.4, CI: 2.1–5.5) as well as in women (OR: 5.3, CI: 2.3–12.4).Conclusion: The level of FMI was 6.6 kg/m2 in men and women predicted at upper limits of 25% and 30% body fat, respectively, for defining obesity. 相似文献

The aim of the study was to evaluate the surgical outcome of patients with advanced medication-related osteonecrosis of the jaw (MRONJ) treated with different flaps for soft tissue closure. Only MRONJ stage III patients with a minimum follow-up of 6 months were included in our prospective study. The soft tissue closure techniques were: Mylohyoid Muscle Flap, Buccal Fat Flap and mucoperiosteal flap alone. Potential risk factors and surgical side effects were analyzed. Relapses occurred in 12 of 44 included cases and 38 reached mucosal integrity within the follow-up. Cases treated with the muscle or fat flap showed better results regarding the recurrence rate (p < 0.001) and soft tissue healing (p = 0.002): only 3 of 33 developed a relapse, and 31 of 33 reached mucosal integrity. The outcome was worse if MRONJ occurred at the front areas of the jaw (p = 0.025). Postoperative, the pain level was reduced significantly (p < 0.001). Partial hypoesthesia of the lip arose in 18 cases. An impairment of the long-term prosthetic rehabilitation has not been seen.Patients with MRONJ stage III undergoing surgery benefit from extensive soft tissue closure. 相似文献
