This research aims to study the effect of the dosage of anhydrous sodium metasilicate activator on the long-term properties of fly ash-based one-part alkali-activated binders (OPAAB) cured at ambient conditions. Powdered sodium metasilicate activator was utilized in the range of 8–16% by weight of the fly ash in producing the OPAAB. The properties examined are hardened density, compressive strength, flexural strength, water absorption, efflorescence formation, and microstructural analysis. The experimental result revealed that the binders exhibited excellent long-term strength properties. The compressive strength of the OPAAP is well correlated with its hardened density. The pastes were found to exhibit good soundness characteristics over the long-term. The absorption of water decreases with an increase in the activator dosage from 8–12%, and beyond that, the water absorption relatively remains the same. Field emission scanning electron microscope (FESEM) micrograph revealed uniformly formed solid matrices with the micro-crack present were observed in the samples. The larger pore size promotes the crystallization of the resulting hydrate substances (N, C)-A-S-H gel. The initial dissolution of the OPAAP occurred within the first 30 min. At longer age of curing, mixtures with a higher dosage of powdered activator tend to absorb less water. Strength properties beyond 28 days are considered as the long-term strength.  相似文献

Talking about helping others makes a person seem warm and leads to social approval. This work examines the real world consequences of this basic, social-cognitive phenomenon by examining whether record-low levels of public approval of the US Congress may, in part, be a product of declining use of prosocial language during Congressional debates. A text analysis of all 124 million words spoken in the House of Representatives between 1996 and 2014 found that declining levels of prosocial language strongly predicted public disapproval of Congress 6 mo later. Warm, prosocial language still predicted public approval when removing the effects of societal and global factors (e.g., the September 11 attacks) and Congressional efficacy (e.g., passing bills), suggesting that prosocial language has an independent, direct effect on social approval.As recently as 2002, public approval of Congress was reliably over 50% and as high as 84%. In late 2013, though, public approval reached an all-time low, with less than 10% of Americans expressing support (1). What caused this dramatic decline in public approval in just over a decade? One explanation is that the public held Congress responsible for societal and global problems (e.g., a weak economy) (2, 3). A second explanation is that the public disapproves of ineffective governance. For example, public approval of Congress tends to drop when Republicans and Democrats are polarized against one another and when Congress conflicts with the President (4, 5). We test a third explanation that has less to do with action and more to do with talk (6). We suggest that recent public disapproval partly resulted from the disappearance of warm, prosocial language in Congressional discourse.Previous experimental research has shown that presenting a warm and prosocial demeanor increases social approval (7). People reveal a wealth of information about their feelings and intentions through verbal communication (810). The speaker’s underlying motives notwithstanding, talking about helping others makes positive impressions upon an audience (11). We investigated whether this well-documented finding can explain public perceptions of Congress. Specifically, we asked whether the recent rise of public disapproval of Congress is predicted by declining prosocial language of elected representatives.To measure prosocial language, we computer analyzed all 123,927,807 words spoken in session of the US House of Representatives between 1996 and 2014. Our approach was to look for linguistic markers of prosocial language; we used content analysis software (12) to calculate the proportion of words in the target text that matched entries in a validated dictionary of prosocial words (13). We then compared levels of prosocial language within each month of Congress with their approval ratings by the American public (14) and found a striking match. Fig. 1 shows that levels of prosocial language and the public’s approval followed the same trajectory between 1996 and 2014, r(204) = 0.55, P < 0.001. Notably, the language of both Democrats, r(204) = 0.53, P < 0.001, and Republicans, r(204) = 0.54, P < 0.001, predicted the public’s approval of Congress.Open in a separate windowFig. 1.Prosocial language within the US Congress predicts the public’s approval of Congress. Prosocial language followed a nearly identical trajectory. In the years spanning 2002 and 2014, a small (19%) decrease in prosocial language ushered in a large (75%) decrease in public approval. The individual words whose use most strongly predicted public approval were as follows: gentle, involve, educate, contribute, concerned, give, tolerate, trust, and cooperate.The correlation between prosocial language and public approval does not necessarily imply that representatives’ language caused the public to approve of them. The reverse could be true: The public’s approval could cause changes in the topics that representatives raised. If one variable were causing the other, the causal factor would have changed first and the effected factor second. We tested which variable changed first in time by assessing the association between prosocial language in the present and public approval with time lags of up to 50 mo in the past and future (Fig. 2). The distribution of the associations across the time lags nearly perfectly fitted a normal distribution curve, r(98) = 0.97, P < 0.001. We reasoned that, if operative, societal and global factors would also have influenced the US President’s language and/or the economy. Alternatively, dysfunctional governance may cause both politicians’ rhetoric to be less civil and the public to disapprove (SI Text and Table S1). To test whether prosocial language has an independent, direct effect on the public sentiment, we ran a regression analysis in which we controlled for the effects of both societal/global factors (in the form of the President’s prosocial language, US unemployment rate, and US consumer expectations about the economy) and competent governance (in the form of partisan conflict, the number of bills that Congress passed, and Presidential vetoes). Since 1979, the television station C-SPAN has broadcast Congressional debates to the public, and a large number (47 million) of Americans watch C-SPAN at least once a week (15), the equivalent of 15% of the total population and 57% of the voting population in Congressional elections. These politically active viewers may hear what representatives say and form impressions, which they may then spread contagiously within their social networks (16, 17).A second possible mechanism is through the news media. Journalists may watch floor debates of Congress and influence the public through journalistic slant. We tested this hypothesis by sampling and coding the tone of news editorials. Our results suggested that prosocial language in Congress predicted positive media coverage, r(175) = 0.22, P = 0.004. And positive media coverage predicted public approval, r(193) = 0.26, P < 0.001. Media coverage explained the link between Congressional language and public approval, B = 1.82, 95% confidence interval (CI) = [0.31, 5.04] in a mediation analysis (SI Text) (18). In addition to an indirect effect via media coverage, a direct effect of Congressional language on public approval remained, B = 36.30, 95% CI = [29.05, 43.55], suggesting that the direct (C-SPAN) and indirect (media) channels may work in tandem to explain how Congressional language influences public opinion.Laboratory research has established that prosocial language can influence whether an audience thinks highly of a speaker (7). The effects of IL-1 and IL-4 on the Epo-independent erythroid progenitor in polycythaemia vera

Summary. Human recombinant interleukin-1 (IL-1) was studied for its effects on the erythroid progenitors from normal subjects and from patients with polycythaemia vera (PV). No supportive effect of IL-1 was noticed on the normal erythropoietin (Epo) dependent, erythroid burst-forming unit (BFU-E) using peripheral blood or bone marrow. In contrast, the Epo-independent BFU-E from peripheral blood of PV patients could be stimulated significantly. This enhancing effect of IL-1 was not only observed with unsorted but also with sorted CD34+ cells. In addition, it was shown that IL-1 indirectly stimulated the Epo-independent BFU-E because anti-GM-CSF could abrogate the supportive effects of IL-1. In contrast to the Epo-independent BFU-E, the Epo-dependent erythroid colony formation from PV patients could not be augmented by IL-1. Finally, we studied the effects of IL-4 on the Epo-independent BFU-E, because IL-4 can affect the erythroid colony formation and modulate the effects of IL-1. IL-4 suppressed the Epo-independent BFU-E. This effect could be counteracted by the addition of IL-1 to the culture medium. However, the suppressive effect of IL-4 was not related to a decline in spontaneous release of IL-1, because an anti-IL-1 antibody did not modify the spontaneous erythroid colony formation. These data indicate that IL-1 and IL-4 exert separate influences on the Epo-independent erythroid colony formation in PV.  相似文献

Summary In order to investigate the respective role of plasma and platelet von Willebrand factor (vWF) in mediating platelet adhesion and thrombus formation, we performed ex vivo perfusion studies with native blood from patients with various subtypes of type I von Willebrand disease (vWD). We studied 34 patients with type I vWD (19 'platelet normal', five 'platelet low', two 'platelet discordant', eight 'Vicenza'). Parallel studies were carried out on nine patients with severe vWD (type III). At high shear rate (2600 s-1) we found that the defect in platelet-vessel wall interactions in patients having a normal platelet vWF content ('platelet normal' and 'Vicenza') involved thrombus formation, whereas platelet adhesion was normal. At this high shear rate, platelet adhesion and thrombus volume were significantly decreased in patients with subtypes 'platelet low' and 'platelet discordant', i.e. when platelet vWF is either low or dysfunctional. These results indicate that platelet vWF may substitute for plasma vWF to promote platelet adhesion. emphasizing the important role of platelet vWF. They also confirm the role of vWF in thrombus formation at high shear rate because an abnormal thrombus volume was observed in all patients. even when platelet adhesion was normal.  相似文献

CCR5/CD4/CXCR4 oligomerization prevents HIV-1 gp120IIIB binding to the cell surface

CCR5 and CXCR4, the respective cell surface coreceptors of R5 and X4 HIV-1 strains, both form heterodimers with CD4, the principal HIV-1 receptor. Using several resonance energy transfer techniques, we determined that CD4, CXCR4, and CCR5 formed heterotrimers, and that CCR5 coexpression altered the conformation of both CXCR4/CXCR4 homodimers and CD4/CXCR4 heterodimers. As a result, binding of the HIV-1 envelope protein gp120IIIB to the CD4/CXCR4/CCR5 heterooligomer was negligible, and the gp120-induced cytoskeletal rearrangements necessary for HIV-1 entry were prevented. CCR5 reduced HIV-1 envelope-induced CD4/CXCR4-mediated cell-cell fusion. In nucleofected Jurkat CD4 cells and primary human CD4+ T cells, CCR5 expression led to a reduction in X4 HIV-1 infectivity. These findings can help to understand why X4 HIV-1 strains infection affect T-cell types differently during AIDS development and indicate that receptor oligomerization might be a target for previously unidentified therapeutic approaches for AIDS intervention.For HIV-1 to enter a target cell, the viral envelope glycoprotein gp120 must interact with a set of cell surface molecules that include the primary receptor, CD4 (1), and a chemokine receptor (CCR5 or CXCR4) that acts as a coreceptor (2, 3). These molecules form CD4/chemokine receptor complexes, as deduced from coprecipitation data for CXCR4 or CCR5 with CD4 (48).Most HIV-1 variants isolated from newly infected individuals use CCR5 and CD4 to enter host cells; these M-tropic R5 strains are predominant in acute and asymptomatic phases of HIV infection. CD4+ T helper type 1 (Th1) cells, which express high CCR5 levels (9, 10), are implicated in maintaining asymptomatic status (11, 12). The “viral shift” from R5 to T-tropic X4 HIV-1 strains correlates with AIDS progression (13, 14). X4 strains infect mainly CD4+ Th2 cells, which express little CCR5 and whose CXCR4 levels resemble those of Th1 cells (15, 16), which suggests that cell susceptibility to HIV-1 infection depends on the CD4/coreceptor ratio and on receptor levels during cell activation and/or differentiation (17). CXCR4 and CCR5 are present as homodimers and heterodimers at the plasma membrane (1820). In addition, gp120-mediated CD4/CXCR4 and CD4/CCR5 association and clustering is reported (2123). Nonetheless, little is known of how CCR5 expression influences the CD4/CXCR4 interaction, or of the molecular basis that underlies the differences in X4 strains infection relative to CCR5 levels at the cell surface.Here, we identify CD4/CXCR4/CCR5 oligomers at the cell membrane, even in the absence of ligands. CCR5 expression in these complexes modifies the heterodimeric CD4/CXCR4 conformation and blocks gp120IIIB binding, without altering binding of the CXCR4 ligand CXCL12 and its subsequent signaling. gp120IIIB-triggered LIMK1 activation, cofilin dephosphorylation, and the actin cytoskeleton rearrangement necessary for cell-cell fusion were impeded in CD4/CXCR4/CCR5-expressing cells. The data obtained using recombinant gp120IIIB glycoprotein were confirmed by experiments showing that X4 HIV-1 infection of Jurkat and primary T cells is regulated by CCR5 expression.  相似文献

Functional Films from Silica/Polymer Nanoparticles

High performance functional coatings, based on hybrid organic/inorganic materials, are being developed to combine the polymer flexibility and ease of processing with the mechanical properties and versatility of inorganic materials. By incorporating silica nanoparticles (SiNPs) in the polymeric matrices, it is possible to obtain hybrid polymer films with increased tensile strength and impact resistance, without decreasing the flexural properties of the polymer matrix. The SiNPs can further be used as carriers to impart other functionalities (optical, etc.) to the hybrid films. By using polymer-coated SiNPs, it is possible to reduce particle aggregation in the films and, thus, achieve more homogeneous distributions of the inorganic components and, therefore, better properties. On the other hand, by coating polymer particles with silica, one can create hierarchically structured materials, for example to obtain superhydrophobic coatings. In this review, we will cover the latest developments in films prepared from hybrid polymer/silica functional systems.  相似文献

Objective

Regular physical exercise within structured lifestyle programs may improve weight status and minimize metabolic risk factors in childhood obesity. The aim of this study was to evaluate the effect of the one-year combined physical exercise/lifestyle program KLAKS on anthropometric and metabolic parameters and glycemic control in childhood obesity.

Materials and Methods

142 overweight/obese (BMI > 90th percentile) candidates (7–18 years) were enrolled, 115 participants completed the program. Anthropometrics and biochemical parameters were obtained at beginning and completion. An oral glucose tolerance test (OGTT) was performed in a subgroup of participants. Course of glucose and insulin levels within OGTT was correlated with several parameters and is reported here for those who completed the program.

Results

The mean standard deviation scores (SDS) decreased significantly for BMI, waist circumference, waist-to-height ratio (WHtR) and percentage body fat (all p ≤ 0.01). Improved metabolic risk markers included mean glucose levels within an OGTT at follow-up compared to baseline (p < 0.0001) and HbA1c (p = 0.05) as well as indications of improvement for gamma-glutamyl-transferase and free fatty acids.

Conclusions

The one-year combined exercise/lifestyle program KLAKS significantly improves markers of obesity and glycemic control. Impaired cardiometabolic risk markers, even subclinical, are also favorably influenced by program participation.  相似文献

The reaction of 2,6-diformyl-4-methylphenol (DFMF) with 1-amino-2-propanol (AP) and tris(hydroxymethyl)aminomethane (THMAM) was investigated in the presence of Cobalt(II) salts, (X = ClO4, CH3CO2, Cl, NO3), sodium azide (NaN3), and triethylamine (TEA). In one pot, the variation in Cobalt(II) salt results in the self-assembly of dinuclear, tetranuclear, and H-bonding-directed polynuclear coordination complexes of Cobalt(III), Cobalt(II), and mixed-valence CoIICoIII: [Co2III(H2L1)2(AP1)(N3)](ClO4)2 (1), [Co4(H2L1)23-1,1,1-N3)2(µ-1,1-N3)2Cl2(CH3OH)2]·4CH3OH (2), [Co2IICo2III(HL2)2(µ-CH3CO2)23-OH)2](NO3)2·2CH3CH2OH (3), and [Co2IICo2III (H2L12)2(THMAM−1)2](NO3)4 (4). In 1, two cobalt(III) ions are connected via three single atom bridges; two from deprotonated ethanolic oxygen atoms in the side arms of the ligands and one from the1-amino-2-propanol moiety forming a dinuclear unit with a very short (2.5430(11) Å) Co-Co intermetallic separation with a coordination number of 7, a rare feature for cobalt(III). In 2, two cobalt(II) ions in a dinuclear unit are bridged through phenoxide O and μ3-1,1,1-N3 azido bridges, and the two dinuclear units are interconnected by two μ-1,1-N3 and two μ3-1,1,1-N3 azido bridges generating tetranuclear cationic [Co4(H2L1)23-1,1,1-N3)2(µ-1,1-N3)2Cl2(CH3OH)2]2+ units with an incomplete double cubane core, which grow into polynuclear 1D-single chains along the a-axis through H-bonding. In 3, HL2− holds mixed-valent Co(II)/Co(III) ions in a dinuclear unit bridged via phenoxide O, μ-1,3-CH3CO2, and μ3-OH bridges, and the dinuclear units are interconnected through two deprotonated ethanolic O in the side arms of the ligands and two μ3-OH bridges generating cationic tetranuclear [Co2IICo2III(HL2)2(µ-CH3CO2)23-OH)2]2+ units with an incomplete double cubane core. In 4, H2L1−2 holds mixed-valent Co(II)/Co(III) ions in dinuclear units which dimerize through two ethanolic O (μ-RO) in the side arms of the ligands and two ethanolic O (μ3-RO) of THMAM bridges producing centrosymmetric cationic tetranuclear [Co2IICo2III (H2L12)2(THMAM−1)2]4+ units which grow into 2D-sheets along the bc-axis through a network of H-bonding.
