Genotyping with flanking DNA markers was used to ascertain Treacher Collins Franceschetti syndrome (TCOF1) in a subject affected by tetralogy of Fallot and cryptorchidism. The proband's family consisted of a father and sister who were affected by the disease, and a healthy mother. Since cardiac malformation and cryptorchidism have been associated with the TCOF1 syndrome, the proband was suspected to be a carrier of the mutated gene. Microsatellite markers D5S527, SPARC and D5S519, which previously mapped the TCOF1 gene within a 2.1-cM interval on chromosome 5 (5q32–33.1), were used to follow the transmission of the TCOf 1 mutated locus. Flanking markers D5S519 and D5S527 were informative and enabled us to exclude inheritance of a TCOF1 mutation to the proband, while showing that cardiac malformation and cryptorchidism were unrelated in mis patient.  相似文献

Cell mediated lympholysis (CML) has been proposed as an in vitro model of the rejection process that results from transplantation of allogeneic tissue. To date, the absolute frequencies of cytotoxic T lymphocytes (CTL) and their precursors (CTL.P) have not been directly estimated in man because of technical difficulties. Through optimizing the conditions for radiometric detection of 51Cr release and the attendant improvement in CML sensitivity, direct CTL frequency estimates have been determined in peripheral blood (PBL), spleen (SPL), and lymph nodes (LNC) after in vitro allostimulation using unrelated human cells and limiting dilution assays. The mean frequency of CTL generated from PBL is 1 in 826 cells (0.121% +/- 0.101%) which, from preliminary experiments, is significantly greater than that generated from either LNC or SPL (p less than 0.05). With restimulation of primed cells on day 10, the frequency of CTL generated from PBL was increased 400%. The CTL.P frequency (0.0064% +/- 0.0050%) was approximately 5% of the corresponding CTL frequency. The CTL.P frequencies were found to be minimal estimates as both accessory "filler" cells and T cell growth factors increased the level of detection of CTL.P an average of threefold. The limiting cell dilution assay as detailed in this report should be a powerful tool for defining the cellular requirements and related factors necessary for optimal induction of a CTL response and should provide the means for determination of the immunogenetic requirements and the allospecificity of human cytotoxic lymphocytes.  相似文献

To investigate airway physiology by use of inhaled aerosols, it is frequently necessary to measure the actual amount of material deposited on the airway wall as well as the site of particle deposition. To satisfy these needs, radiolabeled aerosols and gamma camera techniques have been used to measure regional deposition of inhaled particles. To make quantitative measurements of the amount deposited, previous investigators have used a "phantom" technique to indirectly calibrate the gamma camera for the attenuation of gamma rays through the lungs and chest wall. For this calibration, the phantom is a simulated lung containing a known amount of radioactivity. Radioactive counts emitted from the phantom are assumed to be attenuated in the same manner as the intact human lung. The present article describes a technique to determine directly the amount of inhaled aerosol deposited in the lung and simultaneously to calibrate the gamma camera for each individual subject. We used right angle light scattering and a gamma camera to measure individual values of the deposition fraction (DF) of inhaled aerosol deposited in the lung and the coefficient of attenuation (AC) of gamma rays in normal and obstructed lungs of human subjects. Radiolabeled monodisperse aerosols 1 and 2 microns in diameter were used. Knowledge of the activity of the inhaled aerosol (microcurie per liter), the volume inhaled, and the measured DF determined each subject's AC (counts per minute per microcurie). 如何从体质中寻找中医病因--兼述一个中西医结合方法   总被引:1,自引:1,他引:1  
体质,是对个体生命结构及生命活动基本特点的概括。体质异常是遗传和遗传倾向性疾病发生的根本原因,因此确定了体质类型也就找到了这种疾病的病因。具体讲,我们目前所用的方法是:1、以现代医学病名定中医病名。先确定一定样本病人发病前体质的共有特点,即某种疾病的易患体质,找出体质中的不正常成分(按中医理论);2、分析发病后的证候演变规律,审证求因;3、将发病前体质的不正常成分与病后证候演变综合分析,找出其中的因果关系,并与审计所求之因对照,初步确定病因;4、通过四诊微观化/四诊延伸方法,将西医的病理指标、细菌/分子生物学诊断指标和研究成果等纳入中医诊断体系,再与发病后中医诊断及审证求因结果综合分析,将病因诊断细致化,并确定病机;5、在此病机基础上确定治则治法,并在实验室和临床进行反证。以急性白血病病因病机的确定过程为例进行了演示,同时论述了体质学检验方法对疾病和亚健康状态病因诊断的普遍适用价值。  相似文献

目的:比较国产与进口阿托代他汀治疗高脂血症的疗效。方法:选择原发性高脂血症患者76例,随机分为国产阿托伐他汀(10mg·d~(-1))组和进口阿托伐他汀(10mg·d~(-1))组各38例,均治疗8周。结果:2组治疗4周时总胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白胆固醇(LDL-C)均开始显著下降(P<0.01),治疗8周高密度脂蛋白胆固醇(HDL-C)开始显著上升(P<0.05),但2组间比较差异无显著意义(P>0.05);不良反应发生率国产组10.5%,进口组7.9%,2组差别无显著意义(P>0.05)。结论:国产阿托伐他汀与进口阿托伐他汀均有明显的调脂效果,两者使用均较安全。  相似文献

本文采用实验研究方法,通过建立大鼠脂肪肝模型,研究了有氧运动对脂肪肝的预防作用效果,探讨其可能机理。实验中选取2个月龄雄性SD大鼠30只,随机分为对照组、单纯高脂饮食组、高脂饮食+运动组等三组。5周后对各组大鼠进行SOD、MDA指标的检测,并做病理组织学观察。实验结果表明,高脂饮食组大鼠肝脏MDA升高、SOD活性较对照组活性降低。HE组MDA值低于H组,SOD值明显高于H组。病理学检查肝脏脂肪变性不明显。  相似文献

目的 :观察醒脑开窍法针刺治疗动脉粥样硬化对高脂血症的影响。方法 :选取中风合并高血压患者 34例 ,另取年龄、性别相同其它条件相当的健康人群 34例作对照 ,观察针刺前两组 HDL、TC的差别及治疗组治疗前后的变化。结果 :治疗组治疗前与对照组相比 ,HDL - C明显降低 (P <0 .0 1) ,TC显著升高 (P <0 .0 1) ;治疗后 ,治疗组 HDL - C显著升高 (P <0 .0 1) ,TC明显降低 (P <0 .0 1)。结论 :醒脑开窍法针刺治疗中风合并高血压者可调节血脂的变化 ,从而起到治疗该病的作用。  相似文献

Ethnopharmacological relevance

Traditional Chinese medical herbs have been used in China for a long time to treat different diseases. Based on traditional Chinese medicine (TCM) principle, Chaihuang-Yishen granule (CHYS) was developed and has been employed clinically to treat chronic kidney disease including diabetic nephropathy (DN). The present study was designed to investigate its mechanism of action in treatment of DN.

Materials and methods

Diabetic rats were established by having a right uninephrectomy plus a single intraperitoneal injection of STZ. Rats were divided into four groups of sham, diabetes, diabetes with CHYS and diabetes with fosinopril. CHYS and fosinopril were given to rats by gavage for 20 weeks. Samples from blood, urine and kidney were collected for biochemical, histological, immunohistochemical and molecular analyses.

Results

Rats treated with CHYS showed reduced 24 h urinary protein excretion, decreased serum TC and TG levels, but CHYS treatment did not affect blood glucose level. Glomerular mesangial expansion and tubulointerstitial fibrosis in diabetic rats were significantly alleviated by CHYS treatment. Moreover, CHYS administration markedly reduced mRNA levels of NF-κB p65 and TGF-β1, as well as decreased protein levels of NF-κB p65, MCP-1, TNF-α and TGF-β1 in the kidney of diabetic rats.

Conclusions

CHYS ameliorates renal injury in diabetic rats through reduction of inflammatory cytokines and their intracellular signaling.  相似文献

目的:观察枝管藻多糖对实验性高脂血大鼠血脂和过氧化水平的影响。方法:以Wistar大鼠为研究对象,用高脂饲料建立高脂血症大鼠模型,分别ig枝管藻多糖和烟酸肌醇酯(枝管藻多糖的剂量为150mg·kg~(-1)·d~(-1),300mg·kg~(-1)·d~(-1),烟酸肌醇酯的剂量为 100mg·kg~(-1)·d~(-1)),连续40d。于实验的第 30天和第40天,各采血一次,分别测定血清 TC,TG,LDL-C,HDL-C,MDA,ROS含量和 SOD活性。结果:枝管藻多糖能显著抑制们喂高脂饲料大鼠的 TG(P<0.05)水平的升高,显著降低高脂血症大鼠的血清 TC(P<0.05)和TG(P<0.05)含量,但对血清LDL-C和HDL-C含量的变化几乎无影响;明显降低高脂血大鼠血清ROS,MDA含量,提高血清 SOD活性(P<0.01或 P<0.05)。结论:枝管藻多糖能够显著改善高脂血大鼠体内过氧化状态,但其抗高脂血效果低于烟酸肌醇酯。  相似文献

Ethnopharmacology relevance

Raw and processed Polygoni Multiflori Radix (PMR and PMRP) are used in the prevention and treatment of non-alcoholic fatty liver disease (NAFLD), hyperlipidemia or related diseases. In our previous research, 2, 3, 5, 4′-tetrahydroxy-stilbene-2-O-β-d-glucoside (TSG) displayed the most important role in the total cholesterol (TC) lowering effect among all the chemical constituents of Polygonum multiflorum. Emodin and physcion displayed more favorable triglyceride (TG) reducing effects than TSG. However, there are few researches focus on the approach and mechanism of how do Polygonum multiflorum exhibit good lipid regulation activity. The targeted sites of active substances of Polygonum multiflorum are still not clearly elucidated. This research pays close attention to how major chemical components of Polygonum multiflorum affect the TC and TG contents in liver cells.

Materials and methods

In this research, a sensitive, accurate and rapid in vitro model, steatosis hepatic L02 cell, was used to explore target sites of active chemical substances of Polygonum multiflorum for 48 h. Steatosis hepatic L02 cell was exposed to emodin, physcion and TSG, respectively. The contents of four key enzymes in the pathway of synthesis and decomposition of TC and TG were investigated after exposure. Meanwhile, the contents of lipid transfer protein were also tested. The diacylgycerol acyltransferase 1 (DGAT1) controlled the biosynthesis of TG from free fatty acids while 3-hydroxy-3-methyl-glutaryl-CoA reductase (HMG-CoA reductase) limited the biosynthesis of TC. Hepatic triglyceride lipase (HTGL) and cholesterol 7α-hydroxylase (CYP7A) played the key role in the lipolysis procedure of TG and TC.

Results

The synthesis of TC and TG in steatosis L02 cells were apparently increased in the model group compared to the control group. Intracellular contents of HMG-CoA reductase and DGAT1 increased 32.33% and 56.52%, while contents of CYP7A and HTGL decreased 21.61% and 47.37%. Emodin, physcion and TSG all showed down-regulation effects on HMG-CoA reductase, while up-regulation effects on CYP7A. The most remarkable effect on HMG-CoA reductase was found on emodin. Emodin could reduce the DGAT1 content from 438.44±4.51 pg/mL in model group to 192.55±9.85 pg/mL (100 μm). The content of HTGL in 300 μm physcion group was 3.15±0.15 U/mL, which was more significantly effective than the control, lovastatin and fenofibrate group.

Conclusions

TSG could raise the content of CYP7A and then promote the lipolysis of cholesterol. Moreover, TSG also showed the best LDL-reducing effect. Emodin could inhibit HMG-CoA reductase and DGAT1, which were key enzymes in the synthesis of TC and TG. Physcion increased the content of HTGL, and then could boost the lipolysis of triglyceride. At the same time, physcion showed the best VLDL-reducing effect. In view of the above conclusions, we contributed the lipid regulation activity to an overall synergy of TSG, emodin and physcion.  相似文献
