重症肌无力中枢神经系统受损模型 总被引:26,自引:2,他引:24
目的近年研究结果表明,重症肌无力(MG)病变部位并不仅仅局限于神经肌接头(NMJ)处突触后膜烟碱型乙酰胆碱受体(nAChR),烟碱型乙酰胆碱受体抗体(AChR-ab)病理作用可能波及到中枢神经系统(CNS)。因此,有必要建立模拟MG患者CNS损害的动物模型,研究MG患者脑脊液中存在的AChR-ab引起CNS损害的机制。方法从MG患者血中提取的AChR-ab经侧脑室穿刺注入到大鼠脑室系统,然后观察其症状和体征,以及用脑干听觉诱发电位仪(BAEP)检测鼠脑干听觉传导中枢功能。用免疫组化法(ABC)研究AChR-ab与CNS神经-nAChR之间免疫结合反应及其分布。结果大鼠除了出现脑干听觉传导中枢功能障碍外,还出现类似于MG动物模型表现的症状。免疫组化研究结果显示,神经-nAChR样阳性免疫反应广泛分布于CNS许多部位。结论脑室内注入的AChR-ab与神经-AChR结合引起CNS功能障碍和出现MG动物模型样症状。我们首次建立的中枢受损的MG模型将有助于阐明AChR-ab引起中枢受损和CNS下位运动神经元引起横纹肌收缩无力的机制。 相似文献

Severe anaemia is a frequent complication in advanced HIV infection. In our study we investigated the interaction between cytokine network, HIV infection and erythropoietin (Epo) response with increasing anaemia levels. No correlations could be established between circulating tumour necrosis factor (TNF)-alpha and any of the examined parameters. However, a negative correlation was found between haemoglobin values and soluble TNF receptor levels (sTNF-R-I: r = −0.54; P < 0.001; sTNF-R II: r = −0.47; P < 0.001) as well as interleukin-6 levels ( r = −0.29; P < 0.001). In contrast, no significant increase in log[Epo], counterbalancing haemoglobin decline and paralleling the rise in sTNF receptors, was found. In patients classified as stage III, according to the Centers for Disease Control (CDC) classification, the erythropoietin response was significantly more impaired than in patients from CDC groups I and II ( P < 0.001). The results of this study suggest that similar to its action in vitro , activation of the TNF/TNF-R system may impair erythropoietin production in HIV-associated anaemia. Due to the brief half-life of TNF-α, this activation is particularly reflected by elevations of soluble TNF receptor levels. 相似文献

. We have estimated sarcoplasmic reticulum calcium content using rapid application of caffeine on voltage clamped, isolated
guinea-pig ventricular myocytes. Caffeine induces the release of calcium from the sarcoplasmic reticulum and this calcium
is extruded from the cells by the sarcolemmal Na/Ca exchange. Integrating the inward Na/Ca exchange current thus allows estimations
of sarcoplasmic reticulum calcium content. Ventricular myocytes were stimulated to reach new steady-states by action potential
voltage clamps of varying duration. Once contractile steady-state had been reached caffeine was rapidly applied in place of
the next action potential and sarcoplasmic reticulum calcium content measured. Prolonging the action potential duration increased
sarcoplasmic reticulum calcium content and vice-versa. This calcium loading may underlie the positive inotropic effect of
increased action potential duration.

Mice homozygous for the lpr mutation have B and T cell defects and develop autoantibodies, suggesting that lpr plays a role in their genesis. The lpr defect has been identified as a mutation in the apoptosis-associated Fas receptor (FasR) gene. To begin to define the role of FasR in B cells, we have surveyed FasR expression on B-lineage cells from early progenitors in the bone marrow through their maturation in the periphery. Contrary to some reports, we found that FasR is expressed on B cells at all stages of their development and is highest on germinal center B cells. FasR is not expressed on lpr/lpr-derived cells. These data are consistent with the idea that lpr/lpr mice have an intrinsic B cell defect that may be manifested in developing as well as peripheral B cells. An unexpected finding is that B-1 (CD5) B cells do not constitutively express FasR: FasR becomes detectable on B-1 B cells only after activation. 相似文献

The temporal structure of spike trains recorded from optic fibers and single units of the lateral geniculate nucleus (LGN) and primary visual cortex of the cat was studied with a novel method of inter-spike interval analysis. ON type relay cells of the LGN exhibited a multimodal interval distribution preferring a distinct interval (fundamental interval) and its multiples during the sustained light response, whereas most OFF cells showed a broad, unimodal distribution. The general pattern of the interval distribution was relatively independent of stimulus size and contrast and the degree of light adaptation. Simultaneously recorded S-potentials originating from the retinal input generally produced only a single peak at the fundamental interval length. Therefore, the multimodal interval distribution of LGN cells seems to be a result of intra-geniculate inhibition. Cortical cells also showed a weak tendency to fire with spike intervals similar to LGN cells. Therefore, the regular firing pattern observed at peripheral stages of the visual pathway can persist at higher levels and might promote the occurrence of oscillatory activity. 相似文献

We have shown earlier that nicotinic agonists induce the release of noradrenaline from chick sympathetic neurons in culture in two ways: (a) by activating the postsynaptic nicotinic receptors on nerve cell bodies, giving rise to spreading electrical activity and opening of voltage operated calcium channels in neuronal processes; (b) by activating the presynaptic nicotinic receptors on neuronal processes. In the present work, we investigated the contribution of various pathways to the observed Ca2+ influx and subsequent noradrenaline release. Sympathetic neurons in culture were stimulated either by the nicotinic agonist dimethylphenylpiperazinium or electrically, in the presence or absence of tetrodotoxin and of specific blockers of calcium or nicotinic channels, and the effects on [Ca2+]i in the area of neuronal processes and on noradrenaline release were measured. Under control conditions, the N-type channel blocker ω-conotoxin (0.1 μmol/1) diminished the release of noradrenaline and the increase of intraterminal Ca2+ by 48% and 55%, respectively, whereas the L-type channel blocker (+)Bay k 8644 (1 μmol/1) diminished the release of noradrenaline by 25% and the increase of [Ca2+]i by 39%. The P-type channel blocker ω-agatoxin (0.3 μmol/1) had no effect. The effects of the L-type channel ligands were complex and could only be explained on the assumption that, at high concentrations, these drugs also act as nicotinic antagonists. Tetrodotoxin blocked the Ca2+ response evoked by electrical stimulation whereas DMPP applied in the presence of tetrodotoxin still evoked an increase of [Ca2+]i and the release of noradrenaline (27% and 30% of control without tetrodotoxin, respectively). These residual responses were not blocked by any of the calcium channel blockers used or by their combination. Apparently, a substantial part of the influx of Ca2+ induced by the activation of presynaptic nicotinic receptors is not carried by the N-, L- or P-type channels and probably occurs directly via the open channels of nicotinic receptors. 相似文献
