Recycled concrete aggregate (RCA) is a promising substitute for natural aggregates and the reuse of this material can benefit construction projects both economically and environmentally. RCA has received great attention in recent years in the form of aggregate as well as a geotechnical material of sand size. Next to RCA, another recycled material, which reduces the waste volume and is a part of the present challenges in civil engineering, is tire waste. Despite the good engineering properties of recycled tire waste (RTW), its use is still limited, even after almost 30 years since they were first introduced. To broaden the applicability of reused concrete and rubber, a further understanding of their properties and engineering behavior is required. For this reason, the main subject of this paper is composite materials that consist of anthropogenic soil recycled concrete aggregate (RCA) and crushed pieces of recycled tire waste (RTW). In this study, a series of isotropic consolidated drained triaxial tests were undertaken to characterize the shear strength of eight mixtures of variable grain-size distribution, rubber inclusion (RC), and fine fraction (FF) content. The results show that the introduction of rubber waste leads to changes in the strength parameters of the tested mixtures. Improvements in RCA shear strength were observed, the largest for the mixture M7 with 10% of recycled tire waste. Similarly, the effect of fine fraction content on the angle of internal friction and cohesion was found. Dilation characteristics were observed in all analyzed composites. Based on the results of all tests performed, including physical, geometric, chemical, and mechanical properties of the created composites, it can be stated that the samples would meet local road authority requirements for sub-base applications.  相似文献

Road construction consumes a lot of resources and produces a lot of waste and other pollutants. With the emergence of a resource and energy crisis, how to make efficient use of rap has become the research focus of scientific researchers. The interface fusion effect of old and new asphalt in plant mixing and cooling recycling mode is analyzed in order to improve the utilization rate of old asphalt in reclaimed asphalt pavement. In this paper, Materials Studio software was used to establish a bitumen model using the method of four components of bitumen, and then the rationality of the model was verified by density, solubility number and atomic radial distribution function, and the diffusion coefficient obtained from the mean square displacement (MSD) was taken as its evaluation index. The results showed that the diffusion model tends to be stable after 20 ps, and the degree of diffusion increases with the increase in temperature. The degree of diffusion of new asphalt to old asphalt and the degree of diffusion of old asphalt to new asphalt are basically very similar; however, there are some differences at different temperatures. Only a small part of the surface contact between old and new asphalt has been fused, which accords with the partial fusion theory. Compared with Panjin 90# asphalt, the diffusion coefficient of Zhonghaiyou asphalt increases faster with the increase in temperature. The diffusion coefficient increases by 64.3% with the increase of the content of rejuvenators after adding different rejuvenators into the new asphalt. Clarifying the interface fusion effect will be helpful to guide the optimization design of cold-mixing recycled asphalt mixture more scientifically and reasonably. Future research should focus on increasing the fusion effect of old and new asphalt, and explore its influence on the conventional road performance of asphalt mixture.  相似文献

A proinsulin-transferrin (ProINS-Tf) recombinant fusion protein was designed and characterized for the sustained release of an active form of insulin (INS) by hepatoma cells. During incubation with H4IIE hepatoma cells, a gradual decline of ProINS-Tf concentration, with a concomitant generation of the immuno-reactive insulin-transferrin (irINS-Tf), was detected in the culture medium by using INS- or proinsulin (ProINS)-specific radioimmunoassay (RIA) system. Further studies indicated that the conversion of ProINS-Tf to irINS-Tf was a transferrin receptor (TfR) mediated process that was pH-sensitive, and temperature- and microtubule-dependent. These results suggest that the conversion occurred during the slow recycling route of transferrin (Tf)-TfR pathway, possibly processed by proteases in the slow recycling compartments juxtaposed to the trans-Golgi network (TGN). ProINS-Tf exhibited little activity in the short-term promotion of glucose uptake in adipocytes, indicating that it was in an inactive form similar to ProINS. Stimulation of Akt phosphorylation by ProINS-Tf was detected only after prolonged incubation with H4IIE cells. On the other hand, ProINS-Tf pre-incubated with H4IIE cells for 24 h acquired an immediate activity of stimulating Akt phosphorylation. Furthermore, ProINS-Tf elicited a strong activity in the inhibition of glucose production following 24 h incubation with H4IIE cells. Based on these findings, we conclude that the Tf-TfR endocytosis and recycling pathway enables the conversion and release of ProINS-Tf in an active form of irINS-Tf. Results from this study suggest that the Tf-TfR pathway can be exploited for the design of prohormone-Tf fusion proteins as protein prodrugs for their sustained and targeted activation.  相似文献

In certain cases of endothelial dysfunction l-arginine becomes rate-limiting for NO synthesis in spite of sufficiently high plasma concentrations of the amino acid. To better understand this phenomenon, we investigated routes of substrate supply to endothelial nitric oxide synthase (eNOS). Our previous data with human umbilical vein (HUVEC) and EA.hy.926 endothelial cells demonstrated that eNOS can obtain its substrate from the conversion of l-citrulline to l-arginine and from protein breakdown. In the present study, we determined the quantitative contribution of proteasomal and lysosomal protein degradation and investigated to what extent extracellular peptides and l-citrulline can provide substrate to eNOS. The RFL-6 reporter cell assay was used to measure eNOS activity in human EA.hy926 endothelial cells. Individual proteasome and lysosome inhibition reduced eNOS activity in EA.hy926 cells only slightly. However, the combined inhibition had a pronounced reducing effect. eNOS activity was fully restored by supplementing either l-citrulline or l-arginine-containing dipeptides. Histidine prevented the restoration of eNOS activity by the dipeptide, suggesting that a transporter accepting both, peptides and histidine, mediates the uptake of the extracellular peptide. In fact, the peptide and histidine transporter PHT1 was expressed in EA.hy926 cells and HUVECs (qRT/PCR). Our study thus demonstrates that l-citrulline and l-arginine-containing peptides derived from either intracellular protein breakdown or from the extracellular space seem to be good substrate sources for eNOS.  相似文献

Quantitative electron probe microanalysis and electron spectroscopic diffraction analysis was used to determine the gradient of distribution of calcium and its crystalline pattern at different levels (lower gelatinous membrane, upper gelatinous membrane and otoliths) in the otoconial membrane of adult OF1 mice. Our quantitative electron probe microanalytical data, obtained with scanning-transmission electron microscopy, indicated that there was a gradient in calcium concentration which increased from the vestibular surface towards the otoliths. Differences between the three regions of the otoconial membrane were statistically significant in both the utricle and saccule. Our results with electron spectroscopic diffraction revealed an increasing crystalline development from the lower gelatinous membrane towards the otoliths. Our findings with both techniques suggest that the gelatinous membrane is involved in the maturation and crystallization of the otoliths.  相似文献

To determine whether cones and Müller cells in the rod dominated retina cooperate to regenerate the 11-cis retinal chromophore via the retinoid cycle, two cell lines from the rod dominated retinas of Murine were used for this study: 661W, a mouse cell line derived from cones, and rMC-1, a rat Müller cell line. Retinoid cycle enzymes were analyzed by RT-PCR, and their catalytic activity was detected by incubation with retinoids and analyzed by HPLC. We found that 661W cells are capable of reducing all-trans retinal to all-trans retinol due to the presence of multiple dehydrogenases and to generate minor amounts of retinyl-ester. The rMC-1 cells take up all-trans retinol and oxidize it to all-trans retinal or esterify it to retinyl-ester, but are incapable of isomerizing all-trans retinoids to 11-cis retinoids. This could be a reflection of lack of necessary activities in Müller cells in vivo, which suggests that Müller cells do not contribute to retinoid cycling by regenerating 11-cis retinoids. Alternatively, this could be due to the potential that rMC-1, as a transformed cell line, has stopped expressing the proteins needed for the regeneration of 11-cis retinoids.  相似文献

Today, the scientific community is facing crucial challenges in delivering a healthier world for future generations. Among these, the quest for circular and sustainable approaches for plastic recycling is one of the most demanding for several reasons. Indeed, the massive use of plastic materials over the last century has generated large amounts of long-lasting waste, which, for much time, has not been object of adequate recovery and disposal politics. Most of this waste is generated by packaging materials. Nevertheless, in the last decade, a new trend imposed by environmental concerns brought this topic under the magnifying glass, as testified by the increasing number of related publications. Several methods have been proposed for the recycling of polymeric plastic materials based on chemical or mechanical methods. A panorama of the most promising studies related to the recycling of polyethylene (PE), polypropylene (PP), polyethylene terephthalate (PET), and polystyrene (PS) is given within this review.  相似文献

The research demonstrates microstructural changes and development of specific texture in Ti-6Al-4V specimens produced by electron beam melting (EBM) under different conditions. The effect of two factors, namely, raw material (powder) recycling and hot isostatic pressing (HIP), on the EBM produced samples structure and properties, has been explored. The as-printed and treated samples were investigated using electron backscattered diffraction (EBSD) analysis. Modification of mechanical properties after the EBM and HIP are explained by the EBSD data on microstructural phenomena and phase transformations. The work is devoted to assessing the possibility of reusing the residual titanium alloy powder for the manufacture of titanium components by the combination of EBM and HIP methods.  相似文献
