网格蛋白介导的突触囊泡内吞机制

突触囊泡在神经末梢进行着精确而快速地胞吐、内吞循环。近年来进行的研究发现网格蛋白介导的内吞是囊泡蛋白回收的主要途径。本文介绍了这些在内吞早期阶段发生的事件和参与分子的研究进展。     

酒精厂酒糟滤液全回流新工艺固形物循环分析

提出以固形物回流比X作为描述酒糟粗滤液全回流工艺的参数,并推导了固形物回流比X与体积回流比Y及酒糟、滤液和滤渣固形物含量的关系。证明酒糟、滤液和滤渣固形物含量在全回流无限循环时存在极限。     

Audit of waste collected over one week from ten dental practices. A pilot study

An audit of the waste practices of ten general dental surgeries identified problems that have occurred due to the lack of specific dental guidelines or codes of practice in this area. Occupational health and safety requirements for types and locations of sharps containers, and lack of consensus on what constitutes a sharp, were identified as areas needing attention. Cross-infection control items, such as gloves, masks, single-use cups, and protective coverings, were found to constitute up to 91 per cent of total waste. When infectious waste was reclassified by the audit team as 'that waste which was visibly blood stained,' a reduction in waste in this category was made, during the audit, at each practice. The practice of disposing of radiographic fixer and developer into the sewerage system occurred in three out of the ten practices, even though the Australian Dental Association Inc. has discouraged this practice.     

妇科再造胶囊结合腹腔镜治疗不孕症的临床疗效观察

目的 评价妇科再造胶囊结合腹腔镜治疗不孕症的临床疗效.方法 收集我院2009年1月至2011年1月期间收治的女性不孕症患者资料完整者62例,将患者完全随机分为对照组和观察组,各31例.对照组患者进行腹腔镜手术治疗,观察组在进行腹腔镜手术后加用妇科再造胶囊治疗.对2组患者进行为期24个月的随访,比较分析2组患者术后6、12、24个月内疼痛的复发率以及妊娠率.结果 2组患者一般症状均好转,术后6、12、24个月内对照组患者疼痛的复发率分别为:19.4％(6例)、12.9％(4例)、9.7％(3例),累计复发率为41.9％(13例),妊娠率分别为9.7％(3例)、3.2％(1例)、3.2％(1例),累计妊娠率为16.1％(10例);观察组患者的疼痛复发率分别为9.7％(3例)、6.5％(2例)、3.2％(1例),累计复发率为19.4％(6例),妊娠率分别为19.4％(6例)、12.9％(4例)、6.5％(2例),累计妊娠率为38.7％(12例),差异均有统计学意义(均P＜0.05).结论 妇科再造胶囊辅助腹腔镜手术治疗不孕症的临床疗效良好.     

Population pharmacokinetic analysis of clopidogrel in healthy Jordanian subjects with emphasis optimal sampling strategy

Aim: Clopidogrel is metabolized primarily into an inactive carboxyl metabolite (clopidogrel‐IM) or to a lesser extent an active thiol metabolite. A population pharmacokinetic (PK) model was developed using NONMEM® to describe the time course of clopidogrel‐IM in plasma and to design a sparse‐sampling strategy to predict clopidogrel‐IM exposures for use in characterizing anti‐platelet activity.Methods: Serial blood samples from 76 healthy Jordanian subjects administered a single 75 mg oral dose of clopidogrel were collected and assayed for clopidogrel‐IM using reverse phase high performance liquid chromatography. A two‐compartment (2‐CMT) PK model with first‐order absorption and elimination plus an absorption lag‐time was evaluated, as well as a variation of this model designed to mimic enterohepatic recycling (EHC). Optimal PK sampling strategies (OSS) were determined using WinPOPT based upon collection of 3–12 post‐dose samples.Results: A two‐compartment model with EHC provided the best fit and reduced bias in C_max (median prediction error (PE%) of 9.58% versus 12.2%) relative to the basic two‐compartment model, AUC_0‐24 was similar for both models (median PE% = 1.39%). The OSS for fitting the two‐compartment model with EHC required the collection of seven samples (0.25, 1, 2, 4, 5, 6 and 12 h). Reasonably unbiased and precise exposures were obtained when re‐fitting this model to a reduced dataset considering only these sampling times.Conclusions: A two‐compartment model considering EHC best characterized the time course of clopidogrel‐IM in plasma. Use of the suggested OSS will allow for the collection of fewer PK samples when assessing clopidogrel‐IM exposures. Copyright © 2013 John Wiley & Sons, Ltd.     

血液回收机在腹腔镜脾切除术中的应用

目的 探讨血液回收机在腹腔镜脾切除术中的应用价值.方法 2008年6月～2012年5月,该科行腹腔镜脾切除术21例,术中均使用血液回收机.结果 18例顺利完成手术,3例中转开腹.自体脾血回输150～600mL,平均(300±70)mL,未输异体血,术后恢复顺利.结论 腹腔镜脾切除术中使用血液回收机可以有效地避免或减少异体血输注,提高手术安全性.     

Endothelial Src kinase regulates membrane recycling from the lateral border recycling compartment during leukocyte transendothelial migration

When leukocytes cross endothelial cells during the inflammatory response, membrane from the recently described lateral border recycling compartment (LBRC) is selectively targeted around diapedesing leukocytes. This "targeted recycling" is critical for leukocyte transendothelial migration. Blocking homophilic PECAM interactions between leukocytes and endothelial cells blocks targeted recycling from the LBRC and blocks diapedesis. However, the cellular signaling pathways that trigger targeted recycling are not known. We show that targeted recycling from the LBRC is dependent on Src kinase. The selective Src kinase inhibitor PP2 blocked targeted recycling and blocked diapedesis by over 70%. However, Src kinase inhibition did not affect the structure or normal constitutive recycling of membrane from the LBRC in the absence of leukocytes. PECAM, a Src kinase substrate, traffics between the LBRC and the endothelial surface at the cell border. However, virtually all of the PECAM in the cell that was phosphorylated on tyrosine residues was found in the LBRC. These findings demonstrate that Src kinase activity is critical for the targeted recycling of membrane from the LBRC to the site of transendothelial migration and that the PECAM in the LBRC is qualitatively different from the PECAM on the surface of endothelial cells.
