The effect of cyclic 3,5-adenosine monophosphate (cAMP) on production of the enzyme chloramphenicol acetyltransferase (CAT) by whole bacterial cells was studied in strainsEscherichia coli CSH-2/R222 and WZ-78/R222 (cya855). CAT synthesis in strainE. coli WZ-78/R222 was shown to have an intensity only half as great as that of strainE. coli CSH-2/R222. 目的:将人表皮生长因子(hEGF)与人碱性成纤维细胞生长因子(hbFGF)的C端连接,构建出既可与肝素结合,又具有促进细胞生长活性的融合蛋白,并在大肠杆菌中高效表达。方法:将PCR扩增hEGF全基因与PCR扩增hbFGF5端的231bp的片段连接,克隆到表达载体pJN,构建出含融合基因的表达质粒pJRH。将pJRH转化BL21(DE3)表达菌株,获得融合蛋白的表达菌株pJRH(BL)。Westernblot检测表达产物免疫学活性。表达产物上肝素亲和柱和分子筛进行纯化。结果:融合蛋白表达产量为30%,融合蛋白不仅能与肝素结合,而且具有促细胞增殖的活性。Westernblot检测结果显示融合蛋白有EGF的免疫活性。融合蛋白的等电点为5.2。结论:本研究构建了一个hEGF和hbFGF的C端的融合蛋白,该蛋白基本保持了两者原有的生物活性。作为一个新的活性因子,本研究为探讨活性因子蛋白结构与功能的关系奠定基础。  相似文献

Administration of human recombinant granulocyte colony-stimulating factor (G-CSF, 100 g/kg/day, s.c) to rats for 4 days significantly increased circulating neutrophil counts (by 1130%), together with an increase in mononuclear leukocyte counts (by 119%). Infiltrated pleural neutrophil counts in G-CSF-treated rats (G-CSF-r) 5h after the intrapleural injection of zymosanactivated serum were significantly higher (by 155%) than those in control rats (Vehicle-r). In carrageenin-induced pleurisy, counts of infiltrated pleural neutrophils in G-CSF-r 5 and 7h after carrageenin were significantly higher (by 119% and 116%) than those in Vehicle-r. G-CSF treatment increased the volume of pleural exudate and the plasma exudation rate by 122% and 226%, compared to values in Vehicle-r 5h after carrageenin. Cobra venom factor (75 g/kg, i.v.) significantly reduced pleural neutrophil migration in G-CSF-r (by 53%) and Vehicle-r (by 49%). Bromelain (10 mg/kg, i.v.) and aspirin (100 mg/kg, p.o.) reduced pleural neutrophil migration and reduced exudate volume and plasma exudation. Intrapleural bradykinin-(1–5) and prostaglandin E2 levels were significantly higher in G-CSF-r than in Vehicle-r. The increased neutrophil migration in G-CSF-r may be atributed to enhanced activation of the complement system facilitated by increased plasma exudation due to bradykinin and prostaglandins.accepted by M. 用Westem Blot方法确认周围神经于内含有S-100,然后运用免疫组化PAP方法对Wistar大鼠坐骨神经的近侧和远侧进行S-100检测,染色结果用电子图像进行系统分析处理。结果显示:S-100分布于周围神经束间膜,雪旺氏细胞浆内及细胞膜上,神经轴内未见有S-100存在,图像处理结果证实,正常大鼠坐骨神经干内的S-100呈均匀分布。  相似文献

Summary A screening investigation for the presence of risk factors for the development of atherosclerosis demonstrates a plasma factor deficiency in 0,8% in the Viennese population. These findings are in agreement with the data of a newborn screening performed earlier. All the persons were clinically healthy. In 4 of them at least 1 family member suffered from the same defect. The pathogenetic relevance of the plasma factor defect for thrombophilia at young age is discussed.

In order to investigate the interdependent action of the insertion/deletion polymorphism of the angiotensin-converting enzyme (ACE) gene and polymorphism in exon 11 (C1136-->T; Ala379Val) of the platelet-activating factor acetylhydrolase (PAF-AH) gene, which encodes a functional antagonist of PAF, on the progression of immunoglobulin A (IgA) nephropathy, we analysed both polymorphisms in patients with primary IgA nephropathy, who were followed-up for longer than 3 years. During the follow-up (87.3 +/- 50.0 months), the disease progressed in 38 of the 191 patients (19.9%). The D allele of the ACE gene in the absence of the T allele of the PAF-AH gene did not affect the prognosis [odds ratio (OR), 3.6; 95% confidence interval (CI), 0.8-16.4] and neither did the T allele in the absence of the D allele (OR, 3.0; 95% CI, 0.4-24.2). However, the presence of both was a significant prognostic factor (OR, 6.6; 95% CI, 1.4-31.3). After adjusting for other risk factors, the presence of both proved to be an independent risk factor (OR, 4.5; 95% CI, 1.6-12.7). These results suggest that the interdependent effects of ACE and PAF-AH polymorphisms on the progression of IgA nephropathy might be more important than the effect of the individual polymorphisms.  相似文献

The effect of a long strenuous endurance exercise on the phagocytic function of neutrophils was examined. 9 athletes [7 males, 2 females, age: 36–68 years, body mass: 64 (SD 10) kg, height: 175 (SD 10) cm] completed a competetive 100 km run in 8:07 (median value; range: 7:29–9:50 hours). In a whole blood assay the phagocytosis of opsonized E. coli, the receptor density of the Fc receptor 3 (CD16) and the complement receptor 3 (CD11b, direct immunofluorescence) of neutrophils were measured on a per cell basis by flow cytometry before and up to 3 hours after the race. The phagocytic rate (percentage of neutrophils incorporating bacteria) was unchanged after exercise, whereas the phagocytic activity (number of incorporated bacteria per cell) was significantly reduced by –34 (SD 8) % (Wilcoxon test, P<0.001). The total phagocytic capacity of the blood increased 2-3fold post exercise. The surface antigen expressions of CD11b and CD16 were unaffected by the ultradistance run. The results indicate either a reduced phagocytic function of neutrophils on a single cell basis or the mobilization of neutrophils of the marginal pool with a lower phagocytic activity. However, after a long endurance exercise the phagocytotic capacity of the blood was enhanced due to increased cell concentrations.  相似文献

探讨八珍汤对由环磷酰胺引起的小鼠骨髓造血功能抑制的调控作用。采用环磷酰胺致小鼠血虚模型 ,测定八珍汤对骨髓抑制小鼠外周血象及其细胞因子产生的影响。结果表明 ,八珍汤对环磷酰胺所致血虚模型小鼠骨髓细胞有促进增殖作用 ;经八珍汤诱导制备的巨噬细胞、脾细胞、肺条件培养液和骨骼肌条件培养液能促进血虚模型小鼠骨髓细胞增殖 ,促进血虚模型小鼠骨髓基质细胞分泌肿瘤坏死因子 (TNF)。八珍汤对环磷酰胺所致化疗损伤的造血调控作用可能与直接或间接刺激造血微环境的基质细胞分泌正性和负性造血生长因子有关。  相似文献
