HPV6b L1蛋白在昆虫细胞中的表达及其作为特异性诊断抗原的初步研究 总被引:6,自引:3,他引:3
目的:研究人乳头瘤病毒6b结构蛋白(HPV6b L1)在昆虫细胞系统的表达及其作为特异性诊断抗原的可行性,为HPV感染的血清学诊断及基因工程疫苗研究作准备。方法:采用HPV6b L1重组杆状病毒感染sf-9昆虫细胞制备HPV6b L1,经氯化梯度离心纯化,Western blot分析及电镜观察鉴定;用1μg、5μg和20μg三种剂量免疫小鼠,以检测小鼠血清抗体反应;并用HPV6b L1作为诊断抗原检测HPV感染者(尖锐湿疣及宫颈癌)和健康对照者共112例血清特异性抗体。结果:HPV6b L1在昆虫细胞中得到表达,并可自行组装形成VLPs,且予1μg剂量免疫小鼠就能产生阳性血清抗体反应,HPV6b L1 VLPs特异性抗体对HPV6感染的敏感性和特异性分别为78.6%和97.1%。结论:HPV6b L1 VLPs具较强的免疫原性和抗原性,血清中检测到的HPV6b L1抗体对HPV6感染具较高的特异性,可进一步用于HPV感染的血清学诊断及防治尖锐湿疣的基因工程疫苗研究。 相似文献

目的 :建立一种简易快速 ,适于基层或流行病学调查的自测式胶体金免疫层析法 (GICA)用于检测恶性疟原虫中 H RPII抗原 ,判定是否现症感染。方法 :用 GICA测试条检测全血及滤纸血样中的 H RPII抗原。结果 :40例恶性疟全血及滤纸血 H RPII抗原阳性检出率均为 1 0 0 ,与镜检结果相符合 ;2 5例间日疟均为阴性。结论 :GICA检测血中恶性疟原虫 H RPII抗原特异性强、灵敏度高、简便快速 ,无需特殊仪器设备 ,滤纸血的应用使样本的采集、保存和运输更为便利 ,适合大规模流行病学调查和基层使用 ,有广泛应用价值。 相似文献

As mononuclear cell infiltration and growth of pannus critically depend on synovial neovascularization in rheumatoid arthritis (RA), inhibition of the synovial blood vessels would have the potential to reduce rheumatoid inflammation. In this investigation, we studied the effect of gold sodium thiomalate (GST) and auranofin (AUR) on neovascularization in vivo by using a micropocket technique. Both GST and AUR suppressed rabbit corneal neovascularization in a dose-dependent fashion. Significant inhibition was observed by 3 mg/kg GST and 1 mg/kg AUR injected intravenously every other day. These injections maintained serum gold concentrations at the level of 2–5 g/ml and less than 2 g/ml in GST-and AUR-injected rabbits, respectively. These are concentrations attained in the serum or synovium of rheumatoid patients treated by gold compounds. Similar inhibition was observed by both intramuscular administration of GST and oral administration of AUR. In contrast, no inhibition was observed when non-steroidal anti-inflammatory drugs (NSAIDs; 20 mg/kg acetylsalicylic acid, 10 mg/kg ibuprofen and 10 mg/kg indomethacin) were injected intravenously on a daily basis. These results suggested that gold compounds have an antiangiogenic effect in vivo. The inhibition of neovascularization by gold compounds suggested that they may suppress rheumatoid synovitis by reducing the number of small blood vessels required for mononuclear cell infiltration and synovial tissue proliferation. 相似文献

We have used patch-clamp techniques to study the effect of the sulfhydryl group oxidizing agents mercury and thimerosal on calcium-activated nonselective cation channels from brown adipose tissue. 100 nmol/l mercury and 50 mol/l thimerosal induced a complete block. Blockade could be reversed by reduction of the mercaptide by dithiotreitol (DTT). Mercury was found to be the most potent blocker (IC50-value 21×10–9 mol/l), whereas thimerosal (IC50-value 1.5×10–6 mol/l) was as effective as 3,5-dichlorodiphenylamine-2-carboxylic acid (DCDPC). The DCDPC effect, however, could not be reversed by DTT, indicating different blocking mechanisms. Semi-quantitative urinalysis with urine reagent strips (URS) for erythrocyturia (EU), leucocyturia (LU) and proteinuria (PU) was performed in Congolese and Sudanese school children withSchistosoma haematobium and/orS.mansoni infection. Quantitative urinalysis was performed on the same specimen using microscopy and a Neubauer counting chamber for EU and LU and the Coomassie blue dye-binding assay for PU. Microscopically detectable EU of more than 10 cells/l was found in 63% of all samples and LU of more than 20 cells/l was found in 60% of all samples. With the Coomassie blue method, PU of more than 150 mg/l was detected in 51% of all samples. URS gave positive results of grade 1–3 for EU in 69% of all samples, for LU in 63% of all samples and for PU in 66% of all samples. The sensitivity and specificity of URS compared with standard reference methods were as follows: EU 95% and 75%, LU 81% and 81% and PU 90% and 56%. When the results of all three test were combined, URS differentiated abnormal from normal urine specimens with a sensitivity of 94% and a specificity of 70%. Median quantitative results showed a good correlation with semiquantitative URS readings for all parameters, but there was a wide range of URS scores.We concluded that URS sensitively detect urinary abnormalities and thus may be used as a general screening method under field conditions when more specific methods cannot be performed. In the hospital laboratory,urine microscopy with a counting chamber would be preferred to URS as a sole method for EU and LU detection; URS is useful for the detection of PU in the tropical hospital laboratory where an appropriate quantitative method with a better specificity may not be available. 相似文献

The spatial distribution of pulmonary blood flow and its change over time was investigated in term fetal rabbits, using the plasma tracer fluorescein-isothiocyanate-labeled bovine serum albumin (FITC-BSA). A tracer bolus was injected intravenously and allowed to circulate in vivo for increasing periods of time (2-30 minutes) prior to arrest of the circulation and tissue preparation. Initially, fluorescence was present in the vasculature of 43% of lung parenchymal tissue, disposed as discrete regions or "lobules." Interspersed regions of lung tissue received no tracer inflow. With increasing tracer circulation times (10, 20, and 30 minutes), a greater percentage of lung cross-sectional area contained vessels exhibiting tracer fluorescence (64, 96, and 100%, respectively). In the fetal lung, a high pulmonary vascular resistance (PVR) is maintained. Our studies indicate that, at any given moment, fetal pulmonary blood flow is distributed only to a proportion of discrete lung "lobules," while interspersed "lobules" receive no flow at all. The "lobules" alternate between these "high" and "low" vascular resistance states with a periodicity of approximately 35 minutes, comprising 22 minutes of non-perfusion followed by 13 minutes of perfusion. This circulatory pattern permits both the maintenance of high PVR and uniform lung development. Further, by directing flow to only a portion of the vasculature, greater microvascular flow rates are achieved and hence the risk of blood sludging and stasis is reduced.
