[目的]观察神经干细胞静脉移植对损伤大鼠脊髓功能的治疗作用。[方法]取孕14—16dSD胎鼠的脑室下区组织,体外培养后鉴定细胞。制作脊髓全切模型,伤后1周将Brdu标记好的神经干细胞通过尾静脉注射移植到大鼠体内,移植后及8周行皮层体感诱发电位(CSEP)检测和BBB功能评分,并留损伤脊髓处作病理切片及免疫组化染色。[结果](1)移植后8周BBB评分损伤组、移植组都有所恢复,但都未达到正常水平,移植组恢复较好;(2)模型制作后,CSEP波均消失,细胞移植后8周移植组的波形有不同程度的恢复,但潜伏期延长;(3)移植组大鼠脊髓损伤处存在大量Brdu染色阳性细胞,表明移植的细胞在体内可到达损伤脊髓处并能存活;脊髓损伤部位NF-200及GFAP染色阳性的细胞表明移植的细胞可以分化为具有神经元和胶质细胞特性的细胞。[结论]静脉移植的神经干细胞能到达损伤区代替受损的神经元及神经胶质细胞,使损伤的脊髓功能得到一定程度的恢复。 相似文献

Lesion evolution during focal cerebral ischemia may depend on flow restrictions or on accumulation of toxic mediators within the infarct and expansion of these factors to the periinfarct region. So far, the precise contribution of flow dependent versus spreading-mediated impairment of viable periinfarct tissue has not been determined. Therefore, we measured lesion expansion, flow restrictions and glutamate distribution on serial brain sections at different time points after experimental focal ischemia.Permanent focal ischemia was induced by occlusion of the right middle cerebral artery in male rats and the flow reduction was subsequently measured at 1, 12 and 24 h using iodo[14C]antipyrine autoradiography. Additionally, the necrotic volume was determined on serial brain sections and the glutamate content was measured in tissue samples from adjacent microdissections.Twelve hours after focal ischemia no noteworthy viable areas with blood flow restrictions of 20-40 ml 100 g− 1 min− 1 existed but at 24 h the necrotic tissue exceeded the hemodynamically compromised region by 40 ± 21 mm3 (24%). Furthermore, at 12 and 24 h the glutamate content was elevated in areas surrounding the infarct.Relevant flow restrictions are detectable only during early stages of infarct maturation, whereas the propagation of secondary factors may be the predominant mechanism for delayed infarct evolution. 相似文献

Objectives – To assess long‐term functioning and disability after traumatic brain injury (TBI). Material and methods – Individuals (n = 88) in Norrbotten, northern Sweden, who had been transferred for neurosurgical care were assessed with internationally established TBI outcome measures 6–15 years post‐injury. Results – There was an improvement in overall outcome from discharge from inpatient rehabilitation to follow‐up. Many individuals had a high degree of motor and cognitive functioning, which enabled them to live independently in their own home without assistance, but there remained a disability related to community reintegration and social participation. This affected their productivity and to some degree their marital stability. The remaining disability and reduced productivity were related to the age at injury and the injury severity. Conclusions – Our data showed that individuals with a TBI can achieve and maintain a high degree of functioning many years after the injury. Increasing age and a greater injury severity contributed to their long‐term disability. 相似文献

Data from the Rochester-Olmsted County Medical Records Linkage Project were utilized to assess fertility in persons with epilepsy. Population age-specific reproduction rates for Rochester residents for the years 1935-1974 were estimated using the number of live births from the Minnesota Department of Health Statistics and Vital Statistics of the U.S. for comparison with rates in affected persons. Overall, fertility rates were significantly reduced to 80% of expected for affected males and 85% for affected females. Individuals with partial seizures (simple and complex) were disadvantaged, whereas those with generalized onset were not. During the last 20 years of the study period, males were more disadvantaged than females. The male-female difference was greatest during the time of low population fertility (after 1965). Male deficits were more marked after diagnosis; female deficits were more marked before diagnosis. Differences in the proportion of ever-married person-years between the sexes only partially explain the observed differences. 相似文献

The parameters of myelin development were ascertained in two specific regions of the corpus callosum in a series of cats aged 12 postnatal days through adult. The posteriormost portion of the splenium and the anterior-most portion of the genu were examined in cross section by using the electron microscope. Measurements were made to determine the age at which myelin first appeared, the number and distribution of myelinated fibers, the number and distribution of myelin lamellae, and cross-sectional area of ensheathed axons during development. The results indicate that myelination begins and ends earlier in the anterior region of the callosum. Measurements of myelin lamellae indicate similarities between anterior and posterior regions of the callosum, although development occurs earlier in the genu than in the splenium. No evidence was found for a sequence in the size of axons acquiring myelin sheaths, except that extremely small fibers are the last to begin myelinating. Myelination of the splenium of the corpus callosum begins at the very end of the behavioral and physiological critical period for the corpus callosum's role in visual functional development (Elberger: Behav. Brain Res. 11:223-231, '84; Elberger and Smith: Exp. Brain Res. 57:213-223, '85). Since myelination of a pathway is used as an index of functional reliability, this indicates that the basis for the callosal role in developing visual functions is probably not based on its physiological input to visual cortex. 相似文献

We sought to simultaneously confirm that substantial recovery at day 1 and day 7 after acute ischaemic stroke onset is associated with subsequent neurological deterioration in patients of the Acute Stroke Therapy by Inhibition of Neutrophils randomized clinical trial. Substantial recovery was assessed by improvement in the National Institutes of Health Stroke Score (NIHSS). Neurological deterioration was defined as any stroke event or NIHSS worsening from recovery assessment to day 90. After adjusting for age, t-PA and day 1 NIHSS, there was a non-significant tendency of substantial (pre-specified as 75%) recovery at day 1 to be associated with later deterioration [odds ratio (OR) 2.47; 95% CI, 0.95–6.50]. The corresponding OR for substantial (pre-defined as 65%) recovery at day 7 was 1.84 (0.85–3.96). Other thresholds for recovery were significantly associated with later deterioration: >50%, 80%, 90% and 100% for day 1 and >50%, 60%, 70%, 90% and 100% for day 7. The effect of recovery at day 1 was more important than that of later recovery. This study confirms the association between recovery and subsequent neurological deterioration and is the first to indicate the greater importance of acute recovery at day 1 in comparison with later recovery. 相似文献
