目的;研究3p24EAβMD位点与食管癌的关系。为寻找该位点附近可能存在的抑癌基因奠定基础。方法:采用PCR—RFLP法检测45例食管癌患者3p24位点杂合缺失情况。并对该片段进行克隆。结果:在22例食管癌信息个体中共检出9例杂合缺失。并通过序列分析可知变化为点突变。结论:3p24EAβMD较高的杂合缺失率显示该位点附近可能存在潜在的抑癌基因。  相似文献

采用限制性酶切片段长度多态性分析法,检测在胃癌高发区庄河检出的19例胃癌高系家族成员胃蛋白酶原C基因多态性,发现三种常见片段及一种稀有片段。对其中4例携带稀有片段的受检者进行了胃镜追踪观察,2.5年后发现1例早期胃癌。本文对此作了初步探讨。  相似文献

The previously described cyclic delta opioid receptor-selective tetrapeptide H-Tyr-d -Cys-Phe-d -Pen-OH (JOM-13) was modified at residue 3 by incorporation of both natural and unnatural amino acids with varying steric, electronic, and lipophilic properties. Effects on mu and delta opioid receptor binding affinities were evaluated by testing the compounds for displacement of radiolabeled receptor-selective ligands in a guinea pig brain receptor binding assay. Results obtained with the bulky aromatic 1-Nal3 and 2-Nal3 substitutions suggest that the shape of the receptor subsite with which the side chain of the internal aromatic residue interacts differs for delta and mu receptors. This subsite of either receptor can accommodate the transverse steric bulk of the 1-Nal3 side chain but only the delta receptor can readily accept the more elongated 2-Nal3 side chain. Several analogs with pi-excessive heteroaromatic side chains in residue 3 were examined. In general, these analogs display diminished binding to mu and delta receptors, consistent with previous findings for analogs with residue 3 substitutions of modified electronic character. Several analogs with alkyl side chains in residue 3 were also examined. While delta receptor binding affinity is severely diminished with Val3, Ile3, and Leu3 substitutions, Cha3 substitution is very well tolerated, indicating that, contrary to the widely held belief, an aromatic side chain in this portion of the ligand is not required for delta receptor binding. Where possible, comparison of results in this delta-selective tetrapeptide series with those reported for analogous modification in the cyclic delta-selective pentapeptide [d -Pen2, d -Pen5]enkephalin (DPDPE) and linear pentapeptide enkephalins reveals similar trends.  相似文献

目的:建立流行性乙型脑炎病毒(JEV)C、prME,E等结构蛋白编码基因克隆.方法:病毒感染C6/36细胞,RT-PCR法依次获取该病毒结构蛋白基因并进行DNA测序分析,PCR克隆法将PCR产物分别构建于真核表达载体pcDNA 3.1的BamHI与EcoRI酶切位点并依次命名为pJC、pJME与pJE,通过DNA测序、电泳以及限制性内切酶分析加以鉴定.结果:JEV C、prME与E基因片段分别为380、2001以及1 500bp,各基因测序结果与发表的JEV JaGAr-01株相对应序列相一致,从重组质粒释出的插入子分别与各基因大小相符合.结论:pJC、pJME与pJE重组质粒分别含有C、prME与E蛋白编码基因.  相似文献

INTRODUCTION: The first-order kernel response of multifocal electroretinogram (mfERG) decreases in myopia. A recent study indicates that the flash ERG is also reduced with increased axial length. The aim of this study was to investigate the variations in the first-order response (K1) and the first slice of second-order response (K2.1) across the retina for different axial lengths. METHODS: Thirty healthy subjects with axial length from 23.72 to 28.13 mm (spherical equivalent refractive errors from plano to -10.50 D) were recruited for mfERG measurement using VERIS 4.0. All subjects were fully corrected after cycloplegic refraction and pupils were dilated prior to mfERG recording. There is one trough, n1, and one peak, p1, in the K1 response and three troughs, n1, n2, n3, and three peaks, p1, p2, p3, in the K2.1 response. The amplitudes and implicit times of K1 and K2.1 responses were analysed to determine the characteristic of the responses across retina and the correlation to axial length. RESULTS: The amplitudes of p1 (in the first-order kernel-K1) decreased in the central region and the paracentral region (ring 3) as the axial length increased. The central retinal region showed high rates of reduction in both n1 and p1 (in K1). The amplitudes of n1p1 and n2p2 (in the first slice of the second-order kernel-K2.1) were reduced in the paracentral region (from ring 2 to ring 5) as axial length increased. The average n1 and p1 in K1, and n1p1 and n2p2 in K2.1 mfERG responses are decreased in amplitude by 6-10% per millimetre elongation of axial length. CONCLUSION: Eyes with longer axial lengths, usually with high myopia, have a weaker mfERG response and this attenuation is across the measured retina (from central to paracentral regions) but different kernel responses show a different pattern of attenuation at different retinal eccentricities. The weaker mfERG responses may be related to the morphological changes associated with increased axial length.  相似文献

The Stiles-Crawford effect of the first kind (SCE-I) was measured on both emmetropic and myopic subjects at six different retinal locations. The results revealed a number of significant discrepancies in receptor alignment between the groups of different refractive errors. In myopic subjects, the receptors in the nasal retina (i.e. between the fovea and the optic nerve head) were found to be aligned nasally towards the optic nerve head, whereas the receptors in the temporal retina were aligned towards the centre of the exit pupil. In emmetropic subjects, the receptors across the retina were finely tuned towards the centre of the exit pupil. The magnitude of the receptor displacement in myopic subjects was found to be directly associated with the length of the eyeball.  相似文献

目的:探讨心肌营养素1(cardiotrophin 1,CT1)/破伤风毒素重链C端片段(tentanus toxin C fragment,TTC)(CT1/TTC)融合蛋白的构建及其对大鼠嗜铬细胞瘤(pheochromocytoma,PC12)细胞的靶向性。方法:采用聚合酶链式反应(PCR)、T-A克隆等分子生物学方法构建CT1/TTC融合蛋白。体外培养PC12细胞,并将CT1/TTC融合蛋白与PC12细胞共培养,红色荧光免疫组化染色后在激光共聚焦显微镜下观察融合蛋白能否在TTC的靶向作用下进入PC12细胞。结果:成功构建了CT1/TTC融合蛋白,测序显示融合基因序列正确,免疫组化染色结果显示融合蛋白能够进入PC12细胞,并发出红色荧光。结论:采用PCR和T-A克隆等分子生物学方法能成功构建CT1/TTC融合蛋白.并且TTC能够将CT1靶向进入PC12细胞。  相似文献

目的:探讨载脂蛋白A1、B基因多态性对非刨伤性股骨头坏死(avascular necrosis of the femoral head,ANFH)发生的影响.方法:应用聚合酶链反应对中国北方汉族143例ANFH患者和92例正常人分别扩增含Apo AI基因启动子-75 bp和第一内舍子 83 bp及Apo B基因Eco RI、XbaI和3-VNTR的DNA片段,限制性内切酶酶切扩增产物,琼脂糖凝胶电泳分离基因多态性.结果:Apo A1基因启动子-75 bp处,ANFH患者中A/A基因型频率明显高于正常组(P<0.01),而G/A基因型频率明显低于正常组(P<0.01).Apo AI内舍子 83 bp位点,Apo B基因Eco RI、Xba I位点和3-VNTR区域ANFH患者组和正常组基因型及等位基因频率分布无统计学差异.结论:Apo A1基因启动子区域-75bp位点A/A型可能是非创伤性股骨头坏死易感基因之一,但未能发现Apo A1第一内舍子 83 bp位点及Apo B基因Eco RI、XbaI和3-VNTR位点多态性与非创伤性股骨头坏死发生有明显的关系.  相似文献

目的:研究新疆汉族青年手长、掌长与身高的关系,为人类学、法医学提供参考,同时为中国人体质调查积累资料。方法:应用Hrdlicas标准和我国普遍采用的方法测量了575名(男270名,女305名)18~24岁汉族青年学生的手长、掌长和身高,并将原始数据进行医学统计学处理。结果:按年龄组和性别组计算出各组手长、掌长和身高的均值,身高与手长、掌长的比值。并提出由手长、掌长推算身高的简单公式(身高=手长×比值,身高=掌长×比值)和回归方程(身高=回归系数×手长+截距,身高=回归系数×掌长+截距)。结论:手长、掌长和身高各组性差有显著性(P<0.01);由手长、掌长推算身高的简单公式和回归方程成立(P<0.05)。  相似文献
