Synthetic fragment of human interferon-2α (124th–138th amino acid residue, laboratory code 2438) inhibits the growth of B lymphocytes (Daudi cell line) in a dose-dependent manner. Radiolabeled peptide 2438 binds to specific receptors on the cell surface and competes with interferon-2α for the common binding sites. 目的:通过研究CO和低氧作用后大鼠肺血管细胞增殖和凋亡状况,探讨低氧肺动脉高压的发病机制及防治措施。方法:应用免疫组织化学,原位末端标记及Western杂交等方法检测常压低氧大鼠肺血管壁细胞增殖,凋亡及c-myc基因的表达状况。结果:正常组,低氧和锡原卟啉组,低浓度CO和血晶素组大鼠肺动脉均存在增殖和凋亡的阳性细胞,两类细胞在肺内呈不均匀散在分布,低氧和锡原卟啉组大鼠肺血管增殖细胞数显著升高而凋亡细胞数显著减少,细胞增殖凋亡比值分别为对照组的5倍或4倍,而低浓度CO和血晶素组肺血管增殖细胞和凋亡细胞系数均显著增加,细胞增殖凋亡比值均为1.2,c-myc在低氧和锡原卟啉组大鼠肺内表达显著增加,在低浓度CO和血晶素组大鼠肺内表达减少。结论:增殖和凋亡现象共存在于正常和处理大鼠的肺血管细胞中,也许c-myc等基因的异常表达导致了细胞增殖和凋亡的失衡,进而调节了慢性低氧肺血管结构的改建。  相似文献

Embryonic cholinesterases are assigned important functions during morphogenesis. Here we describe the expression of butyrylcholinesterase and acetylcholinesterase, and the binding of peanut agglutinin, and relate the results to mitotic activity in chick wing and leg buds from embryonic day 4 to embryonic day 9. During early stages, butyrylcholinesterase is elevated in cells under the apical ectodermal ridge and around invading motoraxons, while acetylcholinesterase is found in the chondrogenic core, on motoraxons and along the ectoderm. Peanut agglutinin binds to the apical ectodermal ridge and most prominently to the chondrogenic core. Measurements of thymidine incorporation and enzyme activities were consistent with our histological findings. Butyrylcholinesterase is concentrated near proliferative zones and periods, while acetylcholinesterase is associated with low proliferative activity. At late stages of limb development, acetylcholinesterase is concentrated in muscles and nonexistent within bones, while butyrylcholinesterase shows an inverse pattern. Thus, as in other systems, in limb formation butyrylcholinesterase is a transmitotic marker preceding differentiation, acetylcholinesterase is found on navigating axons, while peanut agglutinin appears in non-invaded regions. These data suggest roles for cholinesterases as positive regulators and peanut-agglutinin-binding proteins as negative regulators of neural differentiation.  相似文献

Summary Cell proliferation of 51 human renal cell carcinomas and 9 larynx and hypopharynx carcinomas has been studied in vitro and using xenotransplants. The proliferative activity ([3H]thymidine labelling index) increases during the first passages in nude mice and then remains almost constant throughout subsequent passages. A comparison of cell kinetic parameters of 8 human renal cell carcinomas, 1 hypopharynx and 2 larynx carcinomas, with data of xenografts and of human tumours in situ published up to now, shows that the cell kinetic parameters of human tumour xenografts presently studied range between those of human tumours in situ and those of autochthonous or transplantable mouse tumours. S-phase durations and potential doubling times are considerably shorter in xenotransplants than in human tumours in situ, whereas the cycle time is about the same. This means that the growth fraction increases considerably after xenotransplantation. Interleukin (IL)-2 and IL-4 are considered as important regulators of growth and differentiation of lymphocytes. We report that in mice made deficient for both IL-2 and IL-4 by gene targeting all major T cell subsets and B cells were normal, indicating that IL-2 and IL-4 are not essential for development of the immune system. Paradoxically, proliferation of T cells was increased in both IL-2- and IL-4-deficient homozygous mice.  相似文献

目的 研究鹿茸生长素对关节软骨细胞增殖的影响。方法 采用家兔关节软骨细胞体外培养和加入同位素3H—TdR标记方法,观察其对3H—TdR掺入细胞DNA合成的影响。结果 加入鹿茸生长素组的3H—TdR的掺入值与对照组有明显差异,细胞增殖作用明显。结论 鹿茸生长素能明显刺激关节软骨细胞的增殖。  相似文献

目的探讨稳定、可靠建立神经干细胞体外增殖的方法,并对增殖培养的细胞进行鉴定。方法获取胚胎大鼠的脑组织,通过加入神经生长因子和采用保留细胞联系技术操作,使脑组织中的神经干细胞在体外克隆增殖并稳定传代。以免疫荧光方法对增殖克隆的神经干细胞球进行鉴定。结果神经干细胞不断增殖形成神经干细胞球且神经干细胞能快速稳定传代增殖。培养的细胞为神经干细胞特异性巢蛋白(nestin)染色阳性细胞。结论在神经生长因子的作用下,利用保留细胞联系技术操作,神经干细胞可以在体外快速、稳定克隆增殖并传代。  相似文献

ABSTRACT: Short-chain fatty acids, such as butyrate and propionate, induce fetal globin gene expression and are under clinical investigation in the β-hemoglobinopathies. Limitations of the short-chain fatty acids as therapeutics include their rapid metabolism and a tendency to induce cell growth arrest if administered for prolonged periods. In studies described here, the cellular effects of other inducers of fetal globin, phenoxyacetic acid and derivatives of short-chain fatty acids and cinnamic acids, were investigated in the human erythroid cell line K562, the IL-3 dependent multi-lineage cell line (32D), and in mice and primates. Several test compounds supported 32D cell proliferation despite a 50-fold depletion of IL-3, which resulted in growth arrest and apoptotic death in control cells. The degree of proliferation induced by certain test compounds was similar to the degree of proliferation induced by Erythropoietin and G-CSF in the cells. Eight of ten compounds induced γ globin mRNA in K562 cells. A 2.5 to 6-fold increase in reticulocytosis was observedin vivoin mice treated with two prototype compounds. Pharmacokinetic studies of three prototype compounds demonstrated millimolar plasma concentrations after single oral doses for many hours in primates. These findings identify orally bioavailable compounds which induce γ globin gene expression and hematopoietic cell proliferation through an activity which partially abrogates requirements for IL-3. Such compounds provide potential for oral therapeutics which stimulate proliferation of hematopoietic cells of multiple lineages, as well as inducing fetal globin.  相似文献

A pilot study was conducted of the biological characteristics of the leukemia cells of newly diagnosed patients with poor prognosis acute myelogenous leukemia (AML). This study included measurements of the pretherapy proliferative rate of the leukemia cells in vivo, assessment of differentiation in vivo during remission induction therapy, and the level of expression of the fms, myc, and IL1β genes in pretherapy leukemia cells. Short cell cycle times were characteristic of the best prognostic category and were associated with a rapid reduction in marrow leukemia cells in cytosine arabinoside (araC)-sensitive patients. Expression of c-fms was associated with rapid reduction in marrow leukemia cells during araC therapy and with a successful treatment outcome. Expression of the IL1β gene was associated with short remissions. These studies suggest that when compared to newly diagnosed standard prognosis AML, the leukemia of poor prognosis patients is more likely to exhibit long cell cycle times, low levels of fms expression, and is less likely to be associated with myeloid differentiation during remission induction therapy. © 1993 Wiley-Liss, Inc.  相似文献

Glycosaminoglycan administration has favourable effects on morphologicaland functional renal abnormalities in different models. Thepossibility that exogenous glycosaminoglycans modulate glomerularmatrix synthesis was explored in both primary and SV40-MES13murine mesangial cell cultures. On both cell types, both low-molecular-weightheparin and different glycosaminoglycans showed dose-dependentinhibition of proliferation and increase of 35SO2–4 uptake.After 36 h the cell compartment contained a spectrum of 35S-moleculesof less than 200 kDa; under heparin treatment, the two main35SO2–4 components (high and medium MW) increased by 16and 37% respectively. Susceptibility to glycosidases revealedthat heparin promotes the expression of heparan sulphate andincreases that of chondroitin sulphate. Moreover, heparin modifiesthe expression of decorin and bigly-can, involved in adhesionand fibrillogenesis, while not affecting perlecan. The extracellularmatrix modulation in renal cells, for which the sulphation typeand ratio of heparin are crucial, may thus explain the beneficialrenal effects of heparin.  相似文献
