目的观察CO中毒致迟发性脑病(DNS)大鼠脑内CD4^+T淋巴细胞浸润以及神经胶质酸性蛋白(GFAP)的表达情况,探讨CO中毒致DNS的病理过程。方法25只SD雄性大鼠随机分为对照组、染毒后3、7、10、20d组,每组5只。采用HE和免疫组织化学染色方法,观察染毒后各时间点大鼠脑内病理形态学变化,及CD4^+T淋巴细胞浸润和GFAP的表达情况。结果HE染色结果显示:各染毒组在大脑皮层及海马均出现神经细胞不同程度的变性、坏死,染毒后7d组最重。免疫组织化学染色结果显示:对照组无CD4^+T淋巴细胞浸润,有少量GFAP表达;各染毒组不同脑区CD4^+T淋巴细胞、GFAP均有不同程度的浸润和表达。CD4^+T淋巴细胞染毒后3d开始浸润,7d达峰值,两者在数量上差异有统计学意义(P〈0.01)。各组染毒后GFAP均有大量表达,随染毒时间延长表达数量呈上升趋势。结论CD4^+T淋巴细胞可能参与了CO中毒致DNS的免疫病理过程,GFAP阳性细胞对CO中毒引发的DNS可能具有保护作用。 相似文献

目的 通过对T淋巴细胞亚群的检测 ,从免疫学方面探讨同种异体细胞来源的组织工程化骨植入猕猴体内修复长段骨缺损的可行性。方法 用骨髓基质干细胞 (MSCs)经诱导分化为成骨样细胞后与生物衍生骨材料复合培养 ,体外构建组织工程化骨 ,植入 15只异体猕猴体内桥接桡骨 2 .5cm节段骨缺损作为实验组 ;用单纯生物衍生骨材料桥接对侧同样大小骨缺损作为对照组 ;分别于术后 1、2、3、6、12周抽取静脉血和作双侧局部组织取材 ,样本应用流式细胞术检测T淋巴细胞亚群CD3 /CD4 /CD45 、CD3 /CD8 /CD45 和CD2 8 阳性率。结果 术后第 1、2、3、6、12周实验组和对照组T淋巴细胞亚群CD3 /CD4 /CD45 、CD3 /CD8 /CD45 和CD2 8 阳性率两者差异均无显著性 (P >0 .0 5 )。结论 生物衍生骨材料和猕猴MSCs复合构建组织工程化骨植入异体猕猴体内其术后 12周内免疫反应水平低 ,可用以修复猕猴节段骨缺损。 相似文献

Investigators often study rats by microCT to investigate the pathogenesis and treatment of skeletal disorders in humans. However, microCT measurements provide information only on bone mineral content and not the solid matrix. CT scans are often carried out on cancellous bone, which contains a significant volume of marrow cells, stroma, water, and fat, and thus the apparent bone mineral density (BMD) does not reflect the mineral density within the matrix, where the mineral crystals are localized. Water- and fat-suppressed solid-state proton projection imaging (WASPI) was utilized in this study to image the solid matrix content (collagen, tightly bound water, and other immobile molecules) of rat femur specimens, and meet the challenges of small sample size and demanding submillimeter resolution. A method is introduced to recover the central region of k-space, which is always lost in the receiver dead time when free induction decays (FIDs) are acquired. With this approach, points near the k-space origin are sampled under a small number of radial projections at reduced gradient strength. The typical scan time for the current WASPI experiments was 2 hr. Proton solid-matrix images of rat femurs with 0.4-mm resolution and 12-mm field of view (FOV) were obtained. The origin of image artifacts in an off-resonance spin-locking experiment is shown to be imperfections in the excitation flip angle. A pulse sequence for off-resonance spin locking is implemented that compensates for imperfections in the excitation flip angle through an off-resonance rotary echo. The off-resonance rotary echo alternates the frequency offset and phase of the RF transmitter during two spin-locking pulses of equal duration. The underlying theory is detailed, and MR images demonstrate the effectiveness of the technique in agarose gel phantoms and in in vivo human brain at 3T. 相似文献

Patients after kidney, heart and lung transplantation differ in their immunosuppressive drug regimens and in susceptibility to infectious complications with cytomegalovirus (CMV). In this study, CMV-specific T-cell responses were characterized in long-term transplant recipients and associated with the frequency of infectious complications. CMV-reactive CD4 T cells from 50 healthy controls, 68 renal, 14 heart and 24 lung transplant recipients were flow cytometrically quantified by the induction of cytokines after specific stimulation. Moreover, the immunosuppressive effect of calcineurin inhibitors on specific T-cell reactivity was quantified in vitro and compared with responses in vivo. Median CMV-specific T-cell frequencies in long-term renal (1.48%; range 0.06-17.26%) and heart transplant recipients (0.90%; 0.13-12.49%) did not differ from controls (1.82%; 0.26-21.00%). In contrast, CMV-specific T-cell levels were significantly lower in lung transplant recipients (0.50%; <0.05-4.98%) and showed a significant correlation with the frequency of infectious episodes (r =-0.57, p = 0.005). The differences within the groups were associated with increasing dosages of immunosuppressive drugs, as exemplified for calcineurin inhibitors that dose dependently reduced specific T-cell reactivity in vitro. In conclusion, monitoring CMV-specific CD4 T cells may serve as a measure for long-term disease susceptibility and may contribute to an improved management of CMV complications after lung transplantation. 相似文献

本文总结报告了3例肠道T细胞淋巴瘤(intestinal T-cell lymphoma,ITCL)的诊治资料,并结合文献复习,发现IT—CL多见于中年男性,以腹痛、血便、发热、体质量下降为主要症状,治疗效果差,预后不良。病理改变以肠道溃疡形成为特点,溃疡形态呈多形性、多灶性、不规则,镜下瘤细胞明显异型、弥漫性浸润,中至大细胞多见。肿瘤细胞呈T细胞表型。ITCL临床少见,缺乏特异性临床表现,极易误诊。故临床医师应重视对ITCL临床病理特征、免疫表型和基因型的研究,注意识别,促其早期诊治。 相似文献

Autoimmune mechanisms are postulated to play a role in the development and progression of dysimmune neuropathies (DN). We investigated the relation between lymphocyte number and marker expression, and disease activity in 20 patients with DN under intravenous immunoglobulins (IVIg) treatment. B- and T-lymphocyte markers were studied by flow cytometry of the expression of CD5, CD25, CD23 and CD38 markers on B cells and of CD3, CD4 and CD8 markers, respectively. These parameters were compared with those obtained from matched healthy volunteers. The proportions of CD38+ B cells were higher in patients compared with those of controls. Proportions of activated CD4+ and CD8+ T cells were comparable in peripheral blood mononuclear cells of patients and controls, but a significant reduction of the absolute numbers of CD3+, CD4+ and CD8+ cells were observed in DN patients. The percentages of CD25+ memory T cells were instead significantly increased in DN patients. Lastly, T-cell reduction and the CD19/CD38 ratio over total B (CD19+) cells directly correlated with a poor response to IVIg therapy. In DN, whereas T-cell number is reduced, activated T and B cells are increased, thus suggesting an intrinsic defect of the immune response. 相似文献

Despite the fact that both H-2K and D molecules are up-regulated in the central nervous system (CNS) following Theiler's murine encephalomyelitis virus (TMEV) infection, resistance in this virus model of multiple sclerosis maps exclusively to D. To address this paradox, we examined the ability of the K and D molecules to present viral antigens to cytotoxic T lymphocytes (CTL). Whereas no virus-specific CTL were detected in the CNS of susceptible B10.Q and B10.S mice 7 days post-infection, D-restricted CTL were identified readily in the CNS of resistant B10 animals. There was no evidence of K-restricted CTL in the CNS of B10 mice at day 7 post-infection. The presence of both K- and D-restricted virus-specific CTL in the spleen of immunized B10 mice demonstrates that the exclusive use of D molecules by CTL in the CNS of mice 7 days post-infection is not due to the inability of the K molecules to present viral peptides to lymphocytes. We conclude that the prominent role of the D locus in determining resistance or susceptibility to TMEV-induced demyelination is determined by factors governing the regulation of the immune response, and not by the presence or absence of CTL precursors capable of recognizing viral peptides presented by the K and D antigen-presenting molecules, or by differences in the ability of the K and D molecules to present viral peptides. 相似文献

Abstract. A single dose of rabbit antithymocyte globulin (ATG) was given as the sole immunosuppressive therapy in a model of strong MHC barrier rat heart allotransplantation. PVG/c hearts transplanted to Wistar/Kyoto (WKy) rats resulted in long-term surviving (LTS) grafts and cell-mediated lympholysis (CML) unresponsiveness in 50% of the animals. The effects of ATG treatment on the peripheral blood lymphocyte subsets were studied by flow cytometry. The absolute T-lymphocyte levels decreased to less than 5% and were normalized after 2 weeks. CD8-positive cells were normalized within 1 week, whereas CD4-and CD5-positive cells remained low. Rats with LTS grafts had low levels of all T-lymphocyte markers, especially the CD4-and CD5-positive cells. Rats rejecting their grafts showed an eightfold increase in levels of CD8-and CD5-positive lymphocytes and a twofold increase in levels of CD4-expressing lymphocytes. It is concluded that ATG treatment causes the immediate elimination of large lymphoid populations as well as long-lasting immunomodulation detectable in peripheral blood. 相似文献
