During orthostatic hypotension we evaluated whether presyncopal symptoms relate to a reduced brain oxygenation. Nine subjects performed 50° head-up tilt for 1 h and eight subjects were followed during 2 h of supine rest and during 1 h of 10° head-down tilt. Cerebral perfusion was assessed by transcranial Doppler determined middle cerebral artery blood velocity (MCA vmean), while brain blood oxygenation was assessed by near-infrared spectrophotometry determined concentration changes for oxygenated (ΔHbO2) and deoxygenated haemoglobin and brain cell oxygenation by the oxidized cytochrome c concentration (ΔCytO2). During head-up tilt, six volunteers developed presyncopal symptoms and mean arterial pressure (88 (78–103) to 68 (57–79) mmHg; median and range), heart rate (96 (72–111) to 65 (50–107) beats min?1), MCA vmean (59 (51–82) to 41 (29–56) cm s?1), ΔHbO2 (by ?5.3 (?3.0 to ?14.8) μmol l?1) and ΔCytO2 were reduced (by ?0.2 (?0.1 to ?0.4) μmol l?1; P < 0.05). During tilt down the cardiovascular variables recovered immediately and ΔHbO2 increased to 2.2 (?0.9–12.0) mmol L?1 above the resting value and also ΔCytO2 recovered. In the nonsyncopal head-up tilted subjects as in the controls, blood pressure, heart rate, MCA vmean and brain oxygenation indices remained stable. The results suggest that during orthostasis, presyncopal symptoms relate not only to cerebral hypoperfusion but also to reduced brain oxygenation. 相似文献

the kinetics of spatial distribution of cellsin vitro. In particular, we have previously observed the preferential spatial distribution of bone cells on the EDS regions of EDS/DMS
patterned surfaces (10). In this study, we examined whether the mechanism of spatial distribution of cells on the EDS regions
was adhesion mediated. Homogeneous layers of EDS and DMS were immobilized on quartz substrates and characterized by contact
angle, X-ray photoelectron spectroscopy, and spectroscopic ellipsometry. The strength of bone cell attachment to the modified
substrates was examined using a radial flow apparatus, within either 20 min or 2 hr of cell incubation in the presence of
serum. A Weibull distribution was chosen to characterize the strength of cell-substratum adhesion. Within 20 min of cell exposure,
the strength of adhesion was significantly larger on EDS and clean surfaces, compared with DMS surfaces (p<0.0001). Within 2 hr of cell incubation, there was no statistical difference between the strength of cell adhesion to EDS,
DMS, and clean surfaces. The results of this study suggest that the surface chemistry mediates adhesion-based spatial cell

We tested the hypothesis that pharmacological reduction of the increase in cardiac output during dynamic exercise with a large muscle mass would influence the cerebral blood velocity/perfusion. We studied the relationship between changes in cerebral blood velocity (transcranial Doppler), rectus femoris blood oxygenation (near-infrared spectroscopy) and systemic blood flow (cardiac output from model flow analysis of the arterial pressure wave) as induced by dynamic exercise of large (cycling) vs. small muscle groups (rhythmic handgrip) before and after cardioselective β1 adrenergic blockade (0.15 mg kg?1 metoprolol i.v.). During rhythmic handgrip, the increments in systemic haemodynamic variables as in middle cerebral artery mean blood velocity were not influenced significantly by metoprolol. In contrast, during cycling (e.g. 113 W), metoprolol reduced the increase in cardiac output (222 ± 13 vs. 260 ± 16%), heart rate (114 ± 3 vs. 135 ± 7 beats min?1) and mean arterial pressure (103 ± 3 vs.112 ± 4 mmHg), and the increase in cerebral artery mean blood velocity also became lower (from 59 ± 3 to 66 ± 3 vs. 60 ± 2 to 72 ± 3 cm s?1; P < 0.05). Likewise, during cycling with metoprolol, oxyhaemoglobin in the rectus femoris muscle became reduced (compared to rest; ?4.8 ± 1.8 vs. 1.2 ± 1.7 μmol L?1, P < 0.05). Neither during rhythmic handgrip nor during cycling was the arterial carbon dioxide tension affected significantly by metoprolol. The results suggest that as for the muscle blood flow, the cerebral circulation is also affected by a reduced cardiac output during exercise with a large muscle mass. 相似文献

Vascular smooth muscle cells (VSM) are normally exposed to transmural fluid flow shear stresses, and after vascular injury, blood flow shear stresses are imposed upon them. Since Ca2+ is a ubiquitous intracellular signaling molecule, we examined the effects of fluid flow on intracellular Ca2+ concentration in rat aortic smooth muscle cells to assess VSM responsiveness to shear stress. Cells loaded with fura 2 were exposed to steady flow shear stress levels of 0.5–10.0 dyn/cm2 in a parallel-plate flow chamber. The percentage of cells displaying a rise in cytosolic Ca2+ ion concentration ([Ca2+]i) increased in response to increasing flow, but there was no effect of flow on the ([Ca2+]i) amplitude of responding cells. Addition of Gd3+ (10 M) or thapsigargin (50 nM) significantly reduced the percentage of cells responding and the response amplitude, suggesting that influx of Ca2+ through ion channels and release from intracellular stores contribute to the rise in ([Ca2+]i) in response to flow. The addition of nifedipine (1 or 10 M) or ryanodine (10 M) also significantly reduced the response amplitude, further defining the role of ion channels and intracellular stores in the Ca2+ response. © 2002 Biomedical Engineering Society.

目的:探讨超短波对慢性肺心病CCP患者的临床应用价值。方法:急性发作期CCP患者87例分为超短波治疗组45例和对照组42例,治疗组在常规治疗的基础上给予超短波治疗,对照组行常规治疗。治疗前后分别测定两组患者血浆中血管内皮生长因子VEGF、内皮素(ET-1)含量,动脉血氧分压(PaO2)和平均肺动脉压(mPAP)水平。结果:与对照组比较治疗组治疗后PaO2显著升高(P<0.01),VEGF、ET-1、mPAP明显降低(P<0.01)。VEGF、ET-1与PaO2呈负相关(P<0.01)、与mPAP呈正相关(P<0.01)。结论:超短波可以通过降低肺动脉高压对CCP患者起治疗作用。 相似文献

for the measurement of peripheral venous oxyhaemaglobin saturation (SvO2) in the adult forearm by a non-invasive technique, near infrared spectroscopy (NIRS) with venous occlusion. A series of studies
is performed on healthy adults to compare measurements of forearm SvO2 made by NIRS with measurements of superficial venous SvO2 made by co-oximetry, and to study the effect of different optode spacings. There is a significant correlation between forearm
SvO2 measured by NIRS and SvO2 of superficial venous blood measured by cooximetry (n=19, r=0.7, p<0.0001). Higher values for SvO2 were obtained using a 2.5 cm spacing than with a 4 cm spacing (mean difference=4.1% (95% Cl 1.4%–6.8%) n=16). This difference
is likely to have been due to a more superficial volume of tissue being studied with the closer optode spacing. Peripheral
SvO2 can be measured non-invasively using NIRS with venous occlusion. It may prove to be a useful method to study circulatory

Dynamic behavior of leukocytes in the microcirculation of solid tumor tissue was visualized using a fluorescent labeling technique combined with the use of a real-time confocal laser-scanning microscope (CLSM) system. Colon tumor cells (RCN-9) were inoculated into the peritoneal cavity of male Fischer 344 rats. Tumor-free rats were similarly injected with physiological saline (intraperitoneally). Ten days after tumor inoculation, the mesentery was exteriorized and subjected to vital microscopic observation under the CLSM system. Leukocytes were labeled with rhodamine 6G (100 g kg–1, intravenously), and their behavior within the microvessels (10–30 m in diameter) was analyzed both in the solid tumor tissues and the normal mesentery. Wall shear rate was calculated from the measured values of vessel diameter and erythrocyte flow velocity. In tumor microvasculature of tumor-bearing rats, the centerline erythrocyte velocity (0.73 ± 0.58 mm s–1, mean±standard deviation) and wall shear rate (210 ± 151 s–1 were significantly lower than those of the tumor-free rats (1.27 ± 0.83 mm s–, 344 ± 236 s–1, respectively). Despite such reduced flow conditions, flux of the rolling leukocytes as well as density of the adhered leukocytes both decreased significantly in tumor microvasculature as compared with normal controls. The methods developed in this work show promise in improving our understanding of tumor biology and pathophysiology. © 1998 Biomedical Engineering Society.
