目的 研究核受体共激活因子5 (NCOA5)对卵巢癌细胞侵袭和转移能力的作用及分子机制。方法 常规培养卵巢癌细胞株SKOV-3、A2780、OC3、HO-8910和正常卵巢上皮细胞系IOSE80,Western blot检测各细胞中NCOA5蛋白的表达水平。选取高表达NCOA5的卵巢癌细胞SKOV-3 和A2780,各分为si-NC组、si-NCOA5组,并分别转染NC siRNA和NCOA5 siRNA。Boyden实验检测各组细胞侵袭能力,Transwell实验检测各组细胞转移能力,Western blot检测各组细胞上皮间质转(EMT)相关蛋白E-cadherin、N-cadherin、vimentin和Slug蛋白的表达水平。结果 Western blot结果显示NCOA5在卵巢癌细胞中的表达显著上调(P<0.05)。在SKOV-3和A2780细胞中干扰NCOA5的表达均显著抑制卵巢癌细胞侵袭和转移能力 (P<0.05),并促进EMT相关蛋白E-cadherin的表达,抑制细胞中EMT相关蛋白N-cadherin、vimentin和Slug蛋白的表达(P<0.05)。结论 NCOA5促进卵巢癌侵袭转移能力,具有作为卵巢癌新型预后生物标志物和治疗分子靶点的潜力。 相似文献

Summary Biotinylated neoglycoproteins are useful to determine the expression of sugar receptors (lectins) histochemically in routinely processed tissue sections. Assessment of the presence of distinct receptor classes with specificity to-galactosides and to- or-N-acetylgalactosamine, selected on the basis of their potential relevance for recognition processes within the metastatic cascade in murine model systems, was performed for a common human tumour type, colorectal cancer. The four different types of neoglycoproteins, derived from covalent attachment of commercially available derivatives of-N-acetylgalactosamine, differed only quantitatively in their capacity to detect specific binding on cultured cells and tissue sections, thus posing no major restriction on the choice of synthetic process for histochemical efficiency of the product. Glycocytological application revealed specific probe binding and a regulation of level of receptor expression for a human colon carcinoma cell line primarily forN-acetylgalactosamine-specific receptors upon retinoic acid-induced differentiation. Monitoring of sections of the 12 cases of primary and secondary colorectal lesions invariably disclosed the presence of the respective receptors, the extent of cell labelling in primary tumours and metastases being similar. This study aims to investigate MMP2 and MT1-MMP protein as well as VEGF-C and VEGF-D mRNA expression in tumor cells and distant organs considered to be targets for metastasis in a tumor spontaneous metastasis model previously described. Cultured tumor cells, able to express pro-MMP2, MMP2, pro-MMP9, and MT1-MMP, develop tumor growth and metastasis, mainly in the liver and spleen, when they are injected in the mammary pad gland of Wistar rats. Immunohistochemical studies of tumor masses showed small groups of tumor cells staining for MT1-MMP but not for MMP2. In the liver, tumor metastatic foci and a stromal positive staining for both MMP2 and MT1-MMP were shown. The spleen and lymph nodes, with only scattered metastatic cells, did not show MMPs immunostaining. Using RT-PCR, a significantly higher VEGF-C and VEGF-D gene expression was shown in the liver of tumor-bearing rats respect to normal rats, whereas spleen and lymph nodes did not show significant differences in mRNA VEGF-C/D levels. Taken together, our results suggest that the stroma microenvironment of target organs for metastasis has the ability to produce MMPs and VEGFs that facilitate the anchorage of tumor cells and promote tumor cell growth and angiogenesis. 相似文献

Summary The growth and vascularization patterns of B16 melanoma colonies in the liver and lungs were measured and compared by histological techniques and dye diffusion patterns after injection of the fluorochrome Hoechst 33342. In the liver, the fluorescent pattern of dye diffusion revealed that uninodular tumours measuring up to 146 n in diameter were not functionally vascularized. However, when the nodules fused to give rise to multinodular tumours measuring between 256 and 366 n in diameter, a reticular dye diffusion pattern revealed functional tumour vascularization. In the lungs, subpleural, parenchymal and peritubular (i.e. surrounding blood vessels and airways) tumours were observed. The two former classes were vascularized down to thicknesses and diameters of 49 and 24 m respectively. In contrast, dye diffusion was never seen in peritubular tumour cuffs up to 609 m in thickness. The results indicate differences in vascularization patterns in B16 tumours in the liver and lungs, and differences between tumours growing in different sites within the lungs. If these results are applicable to metastases in these two organs, they indicate potential diffusion-mediated resistance to chemotherapy, and potential hypoxia-mediated resistance to radiotherapy of both metastases and micrometastases. 相似文献

目的 观察胃癌组织中cNOSmRNA表达分布情况 ,探讨其与胃癌侵袭转移和预后的关系。 方法 运用原位杂交技术 ,检测 119例原发性胃癌标本中cNOSmRNA的表达 ,并对所获得的 82份病例随访资料进行生存分析。 结果 胃癌组织中胃腺癌细胞、血管 淋巴管内皮细胞及巨噬细胞中均可见有cNOSmRNA表达 ;肿瘤细胞分化程度越低、恶性度越高 ,胃腺癌细胞和血管 淋巴管内皮细胞cNOSmRNA阳性表达率就越高 ;胃腺癌细胞和血管 淋巴管内皮细胞cNOSmRNA阳性表达率与胃癌侵袭深度、淋巴结转移以及TNM分期呈正相关 ;胃癌血管 淋巴管内皮细胞cNOSmRNA阳性表达者术后生存率较低 ;巨噬细胞中cNOSmRNA表达与胃癌组织学分型、侵袭深度、淋巴结转移、TNM分期及患者术后生存期无关。 结论 胃腺癌细胞和血管 淋巴管内皮细胞cNOSmRNA表达情况与肿瘤恶性程度、侵袭和转移潜能有关 ,血管 淋巴管内皮细胞上cNOSmRNA阳性表达提示患者预后不良 相似文献

Systemic spread of primary intracranial neoplasms is rare and may be due to ventriculoperitoneal shunt (VPS). The most common tumors to metastasize via VPS are germinoma of the pineal gland and medulloblastoma. We report a case of 16-yr-old girl with central nervous system malignant melanosis who developed subsequent peritoneal implants via VPS. To the best of our knowledge, this patient represents the third reported case of meningeal melanosis or melanoma which metastasized to the peritoneal cavity via VPS. We report the cytological and clinical findings of 16 fine-needle aspirates (FNAs) performed on recurrent (n = 6) and metastatic (n = 10) mixed mesodermal tumors (MMMTs). The median interval between the primary diagnosis and FNA was 16 mo. Primary sites were the endometrium (n = 11), the ovary (n = 3), the cervix (n = 1), and pelvic soft tissue (n = 1). Primary tumors showed carcinoma with homologous mesenchymal components in 13 cases and focal heterologous elements in three (two chondrosarcomas and one rhabdomyosarcoma). The FNAs showed carcinoma in all 16 cases, with adenocarcinoma differentiation in three, Mesenchymal elements were identified in aspirates of three recurrent and two metastatic lesions. They were all homologous. No heterologous mesenchymal elements were identified in the aspirates. We conclude that mesenchymal components in FNAs of MMMTs are less likely to be seen in metastatic lesions, and that heterologous mesenchymal components are rarely seen in these aspirates even in recurrent disease. These findings confirm that the epithelial component is responsible for the malignant behavior of MMMTs, and suggest that these lesions may need to be classified as sarcomatoid carcinomas rather than true carcinosarcomas. 本实验研究了环磷酰胺和LAK细胞被动输入联合应用对SHR大鼠乳腺癌肺转移的影响。结果表明当投给50mg/kg环磷酰胺和被动输入LAK细胞(4×10~7/只)可以明显地抑制SHR大鼠乳腺癌的肺转移(P<0.01),并且能有效地延长荷瘤动物的生存时间(P<0.01);半治疗量的LAK细胞和环磷酰胺具有协同抗转移作用(P<0.05)。提示此方法是一个有效的免疫化学疗法。 相似文献
